Expression and correlation of glucocorticoid receptors and nuclear factor-κB in oral lichen planus

Abstract:Objective To examine the expression and correlation of glucocorticoid receptors (GR)and nuclear factor-κB(NF-κB) in oral lichen planus(OLP).Methods Thirty-two OLP cases and 12 normal oral mucosa specimens were included.The oral mucosae were examined for GR and NF-κB by Westernblotting and real time reverse transcription-PCR(RT-PCR).The correlation between GR and NF-κB in OLP was analyzed by correlation analysis.Results The proteins of GRα,GRβ,p65 and p50 were expressed in both OLP and control group.The expression of GRα mRNA in the OLP was lower than that in control (0.85 ±0.04 in OLP and 1.03 ± 0.03 in control,P =0.000).But there was no difference of GRβ mRNA between OLP and normal samples (1.02 ± 0.06 in OLP and 0.98 ± 0.03 in control,P =0.069).The expression of p65 and p50 mRNA in the OLP both were higher than that in control(1.30 ±0.14 in OLP and 0.99 ±0.02 in control of p65,1.21 ± 0.08 in OLP and 1.03 ± 0.03 in control of p50,both P =0.000).A negative correlation was noted between GRα and p65 in OLP(r =-0.428,P =0.015),and the same correlation found between GRα and p50 (r =-0.536,P =0.002).Conclusions Low expression of GRα may contribute to the protracted course of inflammation and the efficacy of glucocorticoid in OLP.There is an antagonism between GRα and NF-κB in the inflammation of OLP.
