Effects of transplanted bone marrow cells on cardiomyocyte apoptosis and expressions of bcl- 2 after myocardial infarction rats

Abstract:Objectives To study the effects of transplanted bone marrow cells on cardiomyocyte apoptosis and cardiac function in a rat ischemic heart model.Methods Myocardial infarction model was developed in SD rats by ligation of left anterior descending artery. 5×10 6 bone marrow mononuclear cells were injected into peri-infarction zone (BMT group). On 4 or 8 weeks post-infarction, apoptosis of cardiomyocytes and expression of bcl-2 protein were determined by TUNEL or immunohistochemistry. Cardiac function was evaluated by echocardiography (UCG).Results TUNEL results indicated that apoptosis indices of peri-infarction zone in the BMT group were significantly lower than those in the control group (4 weeks,0.0946±0.017vs 0.173±0.018,P0.05;8 weeks,0.0916±0.014 vs 0.182±0.015,P 0.05 ). Immunohistochemistry revealed that bcl-2 expressions in the BMT group were higher than those in the control group (4 weeks,12.66±3.37 vs 5.68±2.53, P0.05; 8 weeks,13.08±3.087 vs 5.02±1.91,P0.05). UCG showed significantly improved EF and FS values in the BMT group (P0.05).Conclusion BMT inhibits cardiomyocytes apoptosis by regulating the expressions of bcl-2 and bax,thus improving cardiac function.
