Comparison between two different bioequivalence designs with within-subject variation around 30%

Abstract:Objective In the bioequivalence (BE) studies, within-subject variations (CVw) around 30% are often encountered. Current paper is to compare the success rates for BEs with CVw around 30% between average bioequivalence (ABE) and referenced-scaled bioequivalence (RSABE) . Methods Mathematical simulation was used to construct the bioequivalence data, and the rates of passing bioequivalence tests were calculated. Results When CVw was around 30%, different BE designs could have different rates of passing BE tests. Under same number of subject times period, ABE method with 2 period cross-over design had higher success rate for BEs with CVw < 30%, while RSABE was more suitable for BEs with CVw> 30%. When RSABE was applied, partial replicate method was a little more efficient than full replicate method for BEs with CVw> 30%; full replicate method was a little more efficient than partial replicate method for BEs with CVw < 30%, however the difference was not noticeable. Conclusion When we perform BE test, the design pattern should be considered.
