Abstract:Objective To isolate and purify the non-alkaloid constituents from Pachysandra terminalis Sieb.et Zucc.using Semi-pre-parative HPLC.Methods The non-alkaloid crude extracts were obtained after the raw materials of Pachysandra terminalis were extracted by ethanol and underwent hydrolysis with 20 mL·L -1hydrochloric acid.Then,the crude extracts were treated with pe-troleum ether,ethyl acetate and n-butyl alcohol in success to finally yield different polarity parts.The ethyl acetate residues were separated on silica gel column and eluted with the mobile phase composed with petroleum ether-ethyl acetate at different ratios.Af-ter that,different polarity fractions were obtained and successively purified by Semi-preparative HPLC.The chemical structures of the isolated compounds from Pachysandra terminalis were identified by 1H-NMR and 13C-NMR.Results 5 compounds were even-tually obtained from the non-alkaloid ethyl acetate part of Pachysandra terminalis and they were identified as scopoletin,isofraxi-din,pinoresinol,protocatechuic acid and quercetin.Conclusion The Semi-preparative HPLC revealed these characteristics of effec-tive,rapid,reproducible and the compounds with high purity,which was applicable to quickly obtain the non-alkaloid constituents from Pachysandra terminalis.
