Protective effect of SIRT 1 agonist on myocardium in diabetic mice

Abstract:Objective To explore the effects of SIRT 1 agonist on myocardial oxidative stress,myocardial hypertrophy and myocardial diastolic function of diabetic mice.Methods Diabetic animal model was established by streptozotocin.The experimental mice were randomly divided into 4 groups,such as blank control group (no modeling,and intragastric administration of normal saline),model group (intragastric administration of normal saline after modeling),positive control group (no modeling,and intragastric administration of resveratrol,2.5 mg/kg daily),and observation group (intragastric administration of resveratrol after modeling,2.5 mg/kg daily).The indexes of oxidative stress and myocardial hypertrophy were detected,and the echocardiography and hemodynamics were measured.Results Compared with the blank control group,the mice myocardium malondialdehyde (MDA) level in model group was significantly increased,the superoxide dismutase (SOD) level was significantly decreased,the E/A ratio and-dp/dtmax were decreased significantly,and the mRNA expressions of atrial natriuretic peptide (ANP) and β myosin heavy chain (β-MHC) were significantly increased.These differences were statistically significant (P<0.05 or P<0.01).For the administrated resveratrol intervention,compared with the model group,the MDA level in observation group was significantly decreased and the SOD level was increased significantly (P<0.05);the E/A ratio was significantly improved (P<0.05).However,the-dp/dtmax also rose again,but the difference was not statistically significant (P>0.05).The mRNA expressions of β-MHC and ANP in observation group were inhibited significantly (P<0.05 or P<0.01).Conclusion Exogenous SIRT 1 agonist can improve myocardial hypertrophy and myocardial diastolic function in diabetic mice.Its mechanism may be that exogenous SIRT 1 agonist inhibits mitochondrial oxidative stress level and ventricular remodeling.
