Abstract:The Alzheimer disease (AD) caused dementia is increasing year by year.Currently, the therapeutic treatment for AD includes cholinesterase inhibitors and memantine, which just release the symptoms.Due to the rapidly rising cost of health care for AD patients, the more efficient therapies, which can reverse the progress of AD are urgently needed to be developed. In AD,misfolded tau is probably a key pathological target. As more and more researches aim to regulate the phosphorylation level or to modulate the accumulation of tau,the recently discoverable prion-like spreading manner of tau provides a new direction for the development of AD treatment. This article describes the strategies directed at reducing misfolded tau and compensating for normal tau function loss.
