Abstract:With the development of aquaculture industry in our country,diseases are becoming increasingly serious.How to detect and diagnose such diseases correctly and rapidly has become a hot topic.Compared with the classic biological and chemical methods,enzyme linked immunosorbent assay(ELISA) is a quick,accurate,simple and convenient method of high sensitivity.It has become an effective means of pathogen detection in lab.In the paper,the theory of ELSIA,its application in the diagnosis of bacterial and viralpathogens in aquiculture and in the detection of marine biological toxin are described.(1) Theory of ELSIAThe basis of ELSIA is the idiosyncratic reaction of antigen-antibody.Sample solution,enzyme conjugate and antibody react in the microtiter wells and wash away the unbound enzyme conjugate,and then add the substrate or chromogen to the wells for incubation.The bounded enzyme conjugate will change the chromogen color.And finally add terminator and use a suitable wavelength to measure the absorption.(2)Application of ELSIA in the detection of bacterial pathogens in aquicultureThe common bacterial pathogens in aquiculture are Vibrio and Aeromonas hydrophila.The detection rate of Vibrio parahaemolyticus in the liver of Preudosciaena crocea could reach 70% using ELSIA technology.The technical analysis of monoclonal antibody of Aeromonas hydrophila has also developed quickly.ELSIA analysis of monoclonal antibody cell strains of the anti-Aeromonas hydrophila provides a rapid and accurate way to diagnose Aeromonas hydrophila in fish.(3)Application of ELSIA in the detection of viral pathogens in aquicultureIn late 1990s,ELISA was already used to determine the toxicity of grass carp haemorrhagic virus(GCHV),it only took 30 min to find the antigen in the unpurified venom successfully.Several years later,ability tests between dot-ELISA and SPA-CoA showed that sensitivity of dot-ELISA was 10 times higher than that of SPA-CoA.ELISA detection of viral pathogens of shrimps mainly involves baculovirus and white spot syndrome virus(WSSV).Sensitivity of baculovirus detection in Penaeus chinensis was up to 60 nanogram and the positive rate of WSSV in the same pool was up to 61.8%.In order to verify the specificity of ELSIA,cross-disturbing test was made between health Pelodiscus sinesis and the control group in tissue solution,cell culture fluid and extract fluid,and viral hemorrhagic septicemia virus(VHSV) and carp pox virus(CPV) in the reaction system.The results show that VHSV,CPV and tissue solution of health Pelodiscus sinesis all presented a negative reaction to the viral antibody,while tissue solution,cell culture fluid and extract fluid of control group presented positive to virus antibody,which confirmed good specificity of ELSIA.(4)Application of ELSIA in the detection of marine biological toxinRapid,accurate and easy to operate is the basic requirements for ELISA detection of marine biological toxin.In early 1990,Japan developed a competitive ELISA method to analysis Okadaic acid,the main components of Diarrhetic shellfish toxin(DSP).In the following years,ELSIA method was established in USA and German using anti-saxtoxin and anti-neo-saxtoxin polyclonal antibody technology. STX and neo-STX could be detected simultaneously and the sensitivity was 40 μg per hundred grams.Though developments have been made in the marine biological toxin detection,some problems such as antibody cross-reaction,shortage of toxin standards still remain to be solved.
