Abstract:Magnetic resonance imaging (MRI) is one of the most useful diagnostic techniques in clinical medicine and more than 30% of all MRI examinations are accompanied by administration of a contrast agent which has become a class of pharmaceuticals. The principles, characterization and classification and recent developments of MRI contrast agents are presented in this review. The primary focus of such research is to study the MRI contrast agents with organ-selectivity, high relaxivity and safety.

Abstract:Different tissues and diseases have specific biological signals. The development of magnetic resonance imaging (MRI) contrast agents responding to these signals can not only improve the sensitivity of disease detection by MRI, but also increase the accuracy of disease diagnosis through molecular imaging of these disease signals. This paper gives a review of MRI contrast agents responding to albumin, pH, metal ion, enzyme, redox and other biological signals with the focus on the latest developments and perspectives.

Abstract:Magnetic resonance imaging (MRI) has revolutionized medical imaging diagnostics with the advantages of non-invasive nature, absence of ionizing radiation, unrestricted penetration depth, high-resolution imaging of soft tissues, organs and blood vessels, and multi-parameter and multi-sequence imaging. Contrast agents (CAs) are crucial for enhancing image quality, detecting molecular-level changes, and providing comprehensive diagnostic information in contrast enhanced MRI. However, the performance of clinical Gd-based CAs represents a limitation to the improvement of MRI sensitivity, specificity, and versatility, thereby impeding the achievement of satisfactory imaging outcomes. In recent years, the development of magnetic nanoparticle-based CAs has emerged as a promising avenue to enhance the capabilities of MRI. Here, we review the advances in magnetic nanoparticle-based MRI CAs, including blood pool CAs, biochemically-targeted CAs, stimulus-responsive CAs, and ultra-high field MRI CAs, as well as the use of CAs for cell labeling and tracking. Additionally, we offer insights into the future prospects and challenges associated with the integration of these nanoparticles into clinical practice.

Abstract:Magnetic resonance imaging(MRI) is a powerful and non-invasive imaging technique that provides very high resolution.It shows both physiological and anatomical details to a certain extent and has no ionizing radiation.However,its resolution cannot meet the clinical needs in some cases,so contrast agents should be used to achieve higher resolution MR images.Data show that about 30% of MRI exams need the use of contrast agents.Gadolinium chelates is one of the most common clinical MRI contrast agents.Traditional gadolinium contrast agents lack specific targeting property,have short half-life and low relaxivity,and cause side effects when large doses of them are used.Recently,the field of nanomedicine-based contrast agents holds great promise for overcoming many traditional hurdles of contrast agents including specific targeting property,half-life,relaxivity,and toxicities.These nanomedicine-based contrast agents mainly include liposomes,dendrimers,micelles,nanoparticles,nanotubes and fullerenes.

Abstract:Magnetic resonance imaging(MRI) is one of the common detection methods in clinical medical imaging. The most commonly used longitudinal relaxation (T1) contrast agent in clinical diagnosis is Gadopentetate Dimeglumine(Gd-DTPA). Since their commercialization, gadolinium-based contrast agents have been studied extensively. This paper introduces the working principle of gadolinium-based inorganic nanoparticle(Gd IONPs) contrast agents, expounds the influence of the morphology, size, surface modification, oxygen vacancy of the inorganic nanoparticles, and looks forward to the design of gadolinium-based inorganic nanoparticle contrast agents.

Abstract:In recent times, magnetic resonance imaging (MRI) has emerged as a highly promising modality for diagnosing severe diseases. Its exceptional spatiotemporal resolution and ease of use have established it as an indispensable clinical diagnostic tool. Nevertheless, there are instances where MRI encounters challenges related to low contrast, necessitating the use of contrast agents (CAs). Significant efforts have been made by scientists to enhance the precision of observing diseased body parts by leveraging the synergistic potential of MRI in conjunction with other imaging techniques and thereby modifying the CAs. In this work, our focus is on elucidating the rational designing approach of CAs and optimizing their compatibility for multimodal imaging and other intelligent applications. Additionally, we emphasize the importance of incorporating various artificial intelligence tools, such as machine learning and deep learning, to explore the future prospects of disease diagnosis using MRI. We also address the limitations associated with these techniques and propose reasonable remedies, with the aim of advancing MRI as a cutting-edge diagnostic tool for the future.

Abstract:Magnetic resonance imaging(MRI) has been extensively used in clinical diagnosis and currently over 30% MRI runs are performed in the presence of contrast agents. However, commercially available contrast agents originated from small molecules typically exhibit relatively low relaxivities and insufficient circulation time. Therefore, there is a long pursuit to develop new contrast agents with high relaxivities to discriminate pathological tissues from normal ones. Compared with small molecule MRI contrast agents, the incorporation of small molecule contrast agents into macromolecular scaffolds allows for constructing macromolecular MRI contrast agents, remarkably elevating the relaxivities due in part to increased rotational correlation time(τR). Moreover, if the macromolecular scaffolds are responsive to external stimuli, the MRI signals could be selectively switched on at the desired sites(e.g., pathological tissues), further intensifying the imaging contrast. In this feature article, we outline the recent achievements in the fabrication of stimuli-responsive macromolecular MRI contrast agents. Specifically, macromolecular contrast agents being responsive to acidic p H, redox potentials, and other stimuli including photoirradiation, pathogens, and salt concentration are discussed. These smart contrast agents could affect either longitudinal(T1) or transverse(T2) relaxation times of water protons or other nuclei(e.g.,19 F), exhibiting enhanced signals in pathological tissues yet suppressed signals in normal ones and displaying promising potentials in in vitro and in vivo MRI applications.

Abstract:Science Progress, Volume 107, Issue 1, January-March 2024. X-ray computed tomography (CT) and magnetic resonance (MR) imaging are essential tools in modern medical diagnosis and treatment. However, traditional contrast agents are inadequate in the diagnosis of various health conditions. Consequently, the development of targeted nano-contrast agents has become a crucial area of focus in the development of medical image-enhancing contrast agents. To fully understand the current development of nano-contrast agents, this review provides an overview of the preparation methods and research advancements in CT nano-contrast agents, MR nano-contrast agents, and CT/MR multimodal nano-contrast agents described in previous publications. Due to the physicochemical properties of nanomaterials, such as self-assembly and surface modifiability, these specific nano-contrast agents can greatly improve the targeting of lesions through various preparation methods and clearly highlight the distinction between lesions and normal tissues in both CT and MR. As a result, they have the potential to be used in the early stages of disease to improve diagnostic capacity and level in medical imaging.

Abstract:Magnetic resonance imaging is a powerful diagnostic technology with high spatial resolution and non-invasion. The contrast agents have significant effect on the resolution of the MR imaging. However, the commercial contrast agents (CAs) usually consist of individual Gd3+ chelated with a low molecular weight acyclic or cyclic ligand, and these small-molecule CAs are usually subjected to nonspecificity, thus leading to rapid renal clearance and modest contrast enhancement for tumor imaging. In recent years, the nanostructured materials conjugated with aptamers were widely used and opened a new door in biomedical imaging due to excellent specificity, non-immunogenicity, easily synthesis and chemical modification of aptamers. This review summarizes all kinds of aptamertargeted MRI CAs and their applications.

Abstract:AbstractMagnetic resonance imaging (MRI) is a clinical imaging modality effective for anatomical and functional imaging of diseased soft tissues, including solid tumors. MRI contrast agents (CA) have been routinely used for detecting tumor at an early stage. Gadolinium‐based CA are the most commonly used CA in clinical MRI. There have been significant efforts to design and develop novel Gd(III) CA with high relaxivity, low toxicity, and specific tumor binding. The relaxivity of the Gd(III) CA can be increased by proper chemical modification. The toxicity of Gd(III) CA can be reduced by increasing the agents' thermodynamic and kinetic stability, as well as optimizing their pharmacokinetic properties. The increasing knowledge in the field of cancer genomics and biology provides an opportunity for designing tumor‐specific CA. Various new Gd(III) chelates have been designed and evaluated in animal models for more effective cancer MRI. This review outlines the design and development, physicochemical properties, and in vivo properties of several classes of Gd(III)‐based MR CA tumor imaging. Abstract:Magnetic resonance imaging (MRI) as one of the most important imaging methods is widely used in clinical diagnosis and basic life science research, with advantages including noninvasiveness, high/multiple soft tissue contrast, parametric imaging and the richness of information. During recent years, there have been many advances in the instrument technology and biomedical application of MRI. Advanced techniques, such as ultra-high field, ultra-fast and ultra-sensitive imaging, are now available for routine clinical usage and research settings. Also, the MRI technology is rapidly developing via incorporating with multiple cutting-edge disciplines including big data technology, artificial intelligence and theranostics. Currently, MRI is playing an indispensable role in tumor molecular imaging, functional brain imaging and brain connectome studies. This paper introduces the history and basic principles of MRI, and summarizes the important problems in the research of MRI instrument, methodology and application, providing a perspective on the directions of future MRI research in China.

Abstract:Computed tomography (CT) is one of the most widely used noninvasive clinical imaging modalities because of its wide availability, low cost, high efficiency and high resolution. Although the resolution of CT is much higher than that of conventional radiography and many other imaging techniques, it is still difficult to distinguish subtle changes of soft tissues because of physical limitations. To increase the contrast of soft tissues and improve the accuracy of disease diagnosis, a variety of CT contrast agents have been developed. Small iodinated compounds have been widely used as clinical CT contrast agents, but their toxicity and short circulation time restrict their further application. To resolve these problems, nanosized CT contrast agents have been developed. In addition to iodine, nanoparticles based on heavy atoms such as gold, tantalum, lanthanides, and bismuth have been investigated as more efficient CT contrast agents than small iodinated compounds. These novel nanosized CT contrast agents have high biocompatibility and long circulation times, which are advantageous for visualization of blood vessels and tumors. Moreover, the combination of CT with other imaging techniques, such as magnetic resonance imaging and ultrasound imaging, could realize higher sensitivity and more personalized diagnostic information. In particular, theranostic CT contrast agents that integrate diagnosis with therapy in a single nanoplatform may serve as a nanomedicine capable of both noninvasive imaging and remote-controlled therapy. Herein, we introduce the principles of CT imaging and commonly used nanoscale CT contrast elements, including iodine, gold, bismuth, tantalum, and lanthanides. The recent advances, future development and existing problems of multifunctional nanosized CT contrast agents are reviewed.

Abstract:Magnetic resonance imaging (MRI) is a non‐invasive molecular imaging tool being extensively employed in clinical and biomedical research for the detection of a broad spectrum of diseases. This technique offers remarkable spatial resolution, good tissue penetration and a high soft tissue contrast. Contrast agents (CAs) have been regularly used in MRI tests to enhance the resolution of MR images and to visualize the diseased sites in the body. In the past years, considerable efforts have been devoted towards developing new theranostic MRI agents that can be tailored to integrate the targeting and therapeutic functions in a single agent. In this review, we have underlined the role of the MRI CAs in the developing field of 'theranostics' and their recent applications in the combined imaging and therapy of different types of tumors. In addition, this review also outlines the different categories of MRI CAs and their comprehensive classification based on different criteria such as chemical composition, relaxation mechanism and biodistribution with clinically relevant examples.
