Abstract:OBJECTIVE To prepare the cream with excellent properties,good transdermal effect and good curative effect for periocular chloasma by screening and optimization of the formulation of Tranexamic acid cream.METHODS In this study,the abdominal skin of SD rats was used for transdermal test in vitro to study the transdermal properties of tranexamic acid.The factors of matrix type,transdermal absorption enhancer,emulsifier and drug loading were investigated by single factor experiment.Three levels were selected for each factor.The factors which influence the transdermal absorption were evaluated with in vitro accumulative permeability as index,and the concentration of the sample was determined by HPLC.RESULTS The optimal prescription of Tranexamic acid cream was selected by screening:the O/W type was selected,the transdermal absorption enhancer was 5％ nitrogen ketone,the emulsifier was 1％ tween 80,and the best drug content was 6％.CONCLUSION The preparation conforms to the quality requirements of ointment,and can be further developed for clinical study.
