Synthesis of kidney-targeted prodrug triptolide-glucosamine conjugate

Abstract:OBJECTIVE To synthesize triptolide-glucosamine conjugate (TPG) as a kidney-targeted prodrug.METHODS Triptolide was reacted with succinic anhydride to generate triptolide succinic acid monoester.Then the free carboxyl was connected to the 1-NH2 from the 2-glucosamine derivative 2-amino-1-(2-aminoethoxy)-glucose through amide bond.The compound was confirmed by NMR and HRMS.RESULTS Through this route,TPG was successfully synthesized.Because of the 2-glucosamine,TPG could target renal and its solubility was increased obviously.CONCLUSION The synthetic route is feasible.The prodrug TPG has renal targeting ability and better solubility.
