Inhibition Activity of BF-30 against Murine Pancreatic Carcinoma Cell Panc02 in vitro Abstract

Abstract:Pancreatic cancer is one of the common types of digestive system cancers and is known to be highly aggressive and malignant.The chemotherapeutic drugs used for pancreatic cancer in clinic,for example gemcitabine,cause significant side effects.It is of great importance to develop novel anticancer drug candidates with lower toxicity.In the present study,the anticancer effect of Cathelicidin-like peptide BF-30 against murine pancreatic carcinoma cell line Panc02 in vitro was examined.CCK-8 assay indicated that BF30 inhibited the proliferation of Panc02 cells in vitro in a dose-and time-dependent manner,with an IC50 of (21.6 ±0.8) μmol/L.The data of transmission electron microscope (TEM) assay showed that,BF-30 induced the rupture of the cytoplasmic membrane,which was consistent with data obtained from lactate dehydrogenase (LDH) release assays and PI staining.When the plasma membrane integrity was injured,increased release of LDH into the medium supernatant occurred,as well as the increased positive rate of PI staining.Meanwhile,BF-30 could bind to genomic DNA of the Panc02 ceils and inhibited the cell migration,as well as suppressed VEGF mRNA level.The amount of genomic DNA binding in Panc02 cells increased from 46.3% to 89.1% as the concentration of BF-30 increased from 10 to 40 μmol/L,while in the untreated control the amount of genomic DNA binding was only 1.7%.The average widths of the wound in BF-30-treated groups were 81.0% and 90.4% at 20 and 40 μmol/L BF-30,respectively,after 24 incubation.In addition,BF-30 significantly suppressed mRNA expression of the VEGF gene in Panc02 cells in a dose-dependent manner and it was only 0.3 fold at 40 μmol/L BF-30 compared with control.Based on all above,the inhibition of BF-30 against Panc02 cells proliferation was dependent on disrupting the cytoplasmic membrane,binding to genomic DNA,inhibiting cell migration,suggesting that it might be a candidate for the treatment of pancreatic cancer.
