Abstract:Object To investigate the biotransformation of the antimalarial compound artemisinin( Ⅰ ) by Catharanthus roseus and Ginkgo biloba cell suspension cultures. Methods Plant tissue culture technology was employed. The product was isolated on silica gel column chromatography and its structure was elucidated by spectroscopic evidence. Results One product was obtained and its structure was characterized as 3α- hydroxydeoxyartemisinin ( Ⅱ ). Conclusion Both of C. roseus and G. biloba cell suspension cultures can bioconvert artemisinin.

Abstract:Object To study the microbial transformation of antimalarial drug artemisinin ( I ).Methods The enzymes secreted by fermentation of Rhizopus chinensis Saito CICC 3043 and Cunninghamel-la elegans Lendn. AS 3. 1207 in the potato medium were employed to transform artemisinin. ResultsThree products were obtained, among which deoxyartemisinin ( Ⅱ ) was also found in the controls withoutmicroorganisms. The other two were identified as 3α-hydroxydeoxyartemisinin (Ⅲ) and 9β-hydrox-yartemisinin (Ⅳ). Compound (Ⅳ) was identified as a new one. Conclusion Artemisinin could be trans-formed by the two title strains of microorganisms. It is easy to release one oxygen atom by breaking of perexide bridge in potato medium and to form deoxyartemisinin, thus making it lose its antimalarial activi-ty. The iron (Fe) in the potato medium may have the above function.

Abstract:Artemisinin is a compound of current interest in the treatment of drug-resistant malaria. An accurate, sensitive, and reproducible reversed-phase HPLC with UV detection method is described for the determination of artemisinin in the herb of Artemisa annua L. Artemisinin in the extracts was converted into a UV-absorbing compound, Q260 by being treated with 0.2% (by weight) NaOH solution and then 0.08M acetic acid. A reversed-phase C-18 silica column (Discovery 250 x 4.6 mm, 5 mum) was selected as a fixed phase, and kept at 30degreesC. The mobile phase was 45/10/45 (by volume) methanol/acetonitrile/0.9' mM Na2HPO4-3.6mM NaH2PO4 buffer (pH 7.76). The flow rate was 0.5 mL/min. The detection wavelength was 260 nm. The standard calibration curve was linear with correlation coefficient 0.9996. The artemisinin content in the herb of Artemisa annua L. was 0.652%. The relative standard deviation and recovery were 1.1% and 98.9%, respectively.

Abstract:Microbial transformations of artemisinin 1 by Cunninghamella echinulata (AS 3.3400) and Aspergillus niger (AS 3.795) were carried out. Two products, 10β-hydroxyartemisinin 2 and 3α-hydroxydeoxyartemisinin 3, were obtained. Their structures were identified on the basis of chemical and spectroscopic data. 10β-Hydroxyartemisinin is a new compound.

Abstract:OBJECTIVE To study the biotransformation of podophyllotoxin by the cell suspension culture and root culture systems of Rheum palmatum. METHOD Using plant tissue culture technology and HPLC techniques to isolate products. The structures were elucidated by spectroscopic means. RESULT Cell suspension culture of R. palmatum could convert podophyllotoxin to produce picropodophyllotoxin with the yield of 73.8%, while root culture of R. palmatum could convert podophyllotoxin to produce epipodophyllotoxin and apopodophyllotoxin. CONCLUSION Podophyllotoxin did not affect the pH value of the media used in tissue cultures. Both cell suspension culture and root culture of R. palmatum can convert podophyllotoxin.

Abstract:Introduction: Artemisinin ( 1 ) is a milestone compound in malaria treatment, and it exhibits a broad scope of bioactivities. Herein, sequential chemo-reduction and biotransformation of artemisinin were undertaken to obtain a series of artemisinin derivatives. Methods: First, 10-deoxyartemisinin ( 2 ) and 9-ene-10-deoxyartemisinin ( 3 ) were synthesized after simple handling with boron trifluoride/diethyl ether and sodium borohydride. Then, biotransformation of 10-deoxyartemisinin was conducted with Cunninghamella echinulata CGMCC 3.4879 and Cunninghamella elegans CGMCC 3.4832, and the transformed products were separated and identified. The antimalarial activity of these products was tested in vitro against Plasmodium falciparum 3D7. Results: Fifteen metabolites (4–18) , including seven novel compounds, were isolated and identified after cultivation. Compounds 2 , 3 , 13 , 15 , 16 , and 18 displayed moderate-to-good antimalarial activity, with a half-maximal inhibitory concentration ranging from 6 to 223 nM. Discussion: This work explored the combination of chemical and biological transformation to develop a co-environmental, efficient, and cost-efficiency synthetic methodology and applied it to synthesize novel derivatives of artemisinin. The association of the two strategies will hopefully provide an abundant source for the development of novel drugs with bioactivities.

[Anthraquinone Production and Analysis in the Hairy Root Cultures of Rheum Palmatum L].

Abstract:Hairy root culture of the medicinal plant Rheum palmatum L. was established by genetic transformation with Agrobacterium rhizogenes. The effects of various media with different pH on growth of the hairy roots and biosynthesis of free anthraquinones were investigated. The experimental results showed that MS agar medium with pH 5.5-5.8 is suitable for growth of the hairy roots. Dark condition is favourable and 62.5-fold increase in fresh weight was reached within a culture period of 25 days. Auxin (0.1 mg.L-1 IAA) activated the hairy root growth but inhibited the biosynthesis of free anthraquinones. About 28% of the total free anthraquinones was released into the liquid medium from the rhubarb hairy roots.

Abstract:Microbial transformation of artemisinin (1) by Cunninghamella elegans was investigated. Four isolated products were identified as 6β-hydroxyartemisinin (2), 7α-hydroxyartemisinin (3), 7β-hydroxyartemisinin (4), and 6β,7α-dihydroxyartemisinin (5). The structures were elucidated by spectroscopic and X-ray crystallographic analysis. Product 5 is a novel compound and being reported here for the first time. It features two hydroxyl groups in its structure, and this is the first report on dihydroxylation of the artemisinin skeleton. Quantitative structure-activity relationship and molecular modeling studies indicate the modification of artemisinin skeleton will increase antimalarial activity and water solubility. The chemical syntheses of artemisinin derivatives at C6 or C7 position are impossible due to the lack of functional groups. 6β,7α-Dihydroxyartemisinin is hydroxylated at both 6β- and 7α-positions of artemisinin skeleton at the same time. Therefore, this new compound would be a good scaffold for further structural modification in the search for more potent antimalarial drugs.

Abstract:The discovery of the artemisinin involved a number of people in an unusual social background. Most of the published articles of the early history of artemisinin discovery, including that in Cell, remain controversial because they are often based on reports relying on one side of a complicated story. The controversies have not been lessened by the award of the Lasker prize to Youyou Tu. Here we present a balanced account of the history about: 1) the effective crude extract of Qinghao (the sweet wormwood); 2) the extraction process of the effective antimalarial monomer; 3) clinical validation of the effective monomer; 4) the determination of artemisinin structure. Our article is based on original documents, conference records, diaries and interview records. These materials indicate that cooperation among different scientists and institutes played significant roles.

Abstract:When (22S, 23S)-homobrassinolide (SSHB) was added at 1 μg l −1 to hairy root cultures of Artemisia annua , the production of artemisinin reached to 14 mg l −1 , an increment of 57% over the control. SSHB treatment led concomitantly to an increased biomass production of 15 g l −1 . A stimulatory activity of SSHB on nucleic acids and soluble protein content in hairy roots was also observed at the growth stage.

Abstract:INTRODUCTIONArtemisia annua is a rich source of biologically active substances such as terpenoids, coumarins and polyacetylenes. These chemicals have been reported to show beneficial pharmacological properties such as antitumor and antibacterial activities. In genetically transformed root cultures of A. annua, three bioactive metabolites, namely, ponticaepoxide (an insecticidal polyacetylene, 1), drimartol A (an anticancer sesquiterpene coumarin, 2) and (Z)-7-acetoxy-methyl-11-methyl-3-methylene-dodeca-1,6,10-triene (a new anticancer sesquiterpene, 3) were isolated and identified in our recent work. However, no quantitative analysis methods for any of them are yet available, nor for their simultaneous analysis.OBJECTIVETo develop an HPLC-PAD method for simultaneous determination of 1, 2 and 3 in hairy root cultures of A. annua.METHODOLOGYHPLC operating conditions were optimised and the chromatographic separation was performed on a C(18) column with a gradient acetonitrile : water as mobile phase.RESULTSLinear relationships within the range of investigated concentrations were observed for the three metabolites with their correlation coefficients greater than 0.997. The method was validated for repeatability (RSD <3.59%) and intra- and inter-day precision (RSD <3.1%) with recovery between 94.8 and 107.6% and the RSD less than 3.40%. The method was successfully applied to the time-course of accumulation of the bioactive compounds in genetically transformed root cultures of A. annua.CONCLUSIONThe HPLC-PAD method developed for the simultaneous determination of three bioactive metabolites 1, 2 and 3 was simple, reproducible and sensitive.

Abstract:Artemisinin,an endoperoxide sesquiterpene lactone isolated from Artemisia annua L.,is an effective drug against multidrug resistant strains of malarial parasite,Plasmodium.In this paper,the biosynthesis pathway for artemisinin was proposed on the basis of the transformation of some precursor such as arteannuin B and artemisinic acid. The function and constituents of the key enzymes involved in the biosynthesis were discussed to reveal the synthetic path. The genetic manipulation on artemisinin biosynthesis was also introduced for the possibility on artemisinin production by biotechniques.
[A new sesquiterpene lactone from biological transformation of Hericium erinaceus and Artemisiae Annuae Herba]

Abstract:This study aimed to investigate the chemical constituents from the biological transformation of Hericium erinaceus and Artemisiae Annuae Herba(HQ biological transformation). The chemical constituents of ethyl acetate fraction of 75% ethanol extract in HQ biological transformation were separated and purified by silica gel and Sephadex LH-20 gel column chromatographies together with semi-preparative high performance liquid chromatography(HPLC). Their structures were identified by physicochemical properties, spectroscopic analysis, as well as comparisons with the data reported in literature. Nine compounds were isolated and identified as 2α-hydroxydeoxyartemisinin(1), 6β-hydroxy-stigmast-4,22-dien-3-one(2), 3β,5α-dihydroxy-ergosta-7,22-dien-6-one(3), friedelin(4), dankasterone(5), ergosterol endoperoxide(6), 3β-hydroxy-5,9-epoxy-(22E,24R)-ergosta-7,22-dien-6-one(7), 3α,5α,9α-trihydroxy-(22E,24R)-ergosta-7,22-dien-6-one(8), and stigmast-3-one(9). Compound 1 was a new sesquiterpene lactone named 2α-hydroxy-deoxyartemisinin. The activity against Helicobacter pylori(Hp) of compounds 1-9 in vitro was determined by Kirby-Bauer disk diffusion method. The screening results showed compounds 1, 2 and 5 had certain anti-Hp activity.

Abstract:Artemisinin is an endoperoxide sesquiterpene lactone isolated from the aerial parts of Artemisia annua L., and is presently the most potent anti-malarial drug. Owing to the low yield of artemisinin from A. annua as well as the widespread application of artemisinin-based combination therapy recommended by the World Health Organization, the global demand for artemisinin is substantially increasing and is therefore rendering artemisinin in short supply. An economical way to increase artemisinin production is to increase the content of artemisinin in A. annua . In this study, three key genes in the artemisinin biosynthesis pathway, encoding farnesyl diphosphate synthase, amorpha-4, 11-diene C-12 oxidase and its redox partner cytochrome P450 reductase, were over-expressed in A. annua through Agrobacterium -mediated transformation. The transgenic lines were confirmed by Southern blotting and the over-expressions of the genes were demonstrated by real-time PCR assays. The HPLC analysis showed that the artemisinin contents in transgenic lines were increased significantly, with the highest one found to be 3.6-fold higher (2.9 mg/g FW) than that of the control. These results demonstrate that multigene engineering is an effective way to enhance artemisinin content in A. annua .

Abstract:OBJECTIVE To study the relative expression of the genes involved in artemisinin biosynthesis in different tissues including roots, stems, leaves and flowers of Artemisia annua, and establish the relationship between gene expression and artemisinin accumulation, eventually leading to discover the mainly effective genes involved in artemisinin biosynthesis. METHOD The 7 functional genes involved in artemisinin biosynthesis were detected at the level of expression by using qRT-PCR, and simultaneously the content of artemisinin in the 4 investigated tissues was detected in parallel. RESULT The 3 genes including HMGR, DXR and FPS which were involved in the upstream pathway of artemisinin biosynthesis showed the highest expression levels in flowers, and the 4 functional genes including ADS, CYP71AV1, CPR and AAR which were involved in the artemisinin-specific biosynthetic pathway were found to be expressed in all the 4 detected tissues. The highest expression level of ADS was found in leaves, then followed by flowers, and the lowest expression level of ADS was found in roots and stems. CYP71AV1 had highest expression level in flowers and lowest in leaves. CPR showed highest expression level in flowers, and AAR had lower expression levels in the other 3 artemisinin-specific pathway genes in all the tissues. The highest content of artemisinin was found in leaves (0.343 mg x g(-1)), then followed by flowers (0.152 mg x g(-1)), roots (0.062 mg x g(-1)) and stems (0.060 mg x g(-1)). CONCLUSION In the biosynthesis of artemisinin, the upstream genes including HMGR from the MVA pathway, DXR from the MEP pathway and the checkpoint gene FPS were much more active in flowers, and this suggested that flowers might be the tissues of artemisinin precursor biosynthesis, and further DXR contributed more to artemisinin biosynthesis. The positive correlation of ADS expression and artemisinin content in tissues demonstrated that ADS played a very important role in artemisinin biosynthesis, which was the ideal target for engineering the artemisinin biosynthetic pathway. In summary, the functional genes involved in artemisinin biosynthesis do not express at the same level but synergistically.
A New Sesquiterpene from Hairy Root Culture of Artemisia Annua

Abstract:A new sesquiterpene (Z)-7-acetoxy-methyl-11-methyl-3-methylenedodeca-1,6,10-triene (AMDT) was isolated and identified from the methanol extract of the hairy root culture of Artemisia annua. The structure of AMDT was determined based on the analysis of spectroscopic data, notably of the 2D NMR spectra. This new compound showed cytotoxicity against human tumor cell lines 95-D and HeLa with IC50 values of 27.08 and 20.12μmol/L, respectively.

Abstract:Artemisinin, isolated from an annual herbaceous plant Artemisia annua L., is an effective antimalarial compound. However, artemisinin is accumulated in small amounts (0.01–0.1% leaf dry weight) in A. annua , resulting in constant high artemisinin price. Although metabolic engineering of partial artemisinin metabolic pathway in yeast achieved great success, artemisinin from A. annua is still the important business resource. Here, we report on the generation of transgenic plants with simultaneously overexpressing four artemisinin biosynthetic pathway genes, amorpha-4,11-diene synthase gene ( ADS ), amorpha-4,11-diene 12-monooxygenase gene ( CYP71AV1 ), cytochrome P450 reductase gene ( CPR ), and aldehyde dehydrogenase 1 gene ( ALDH1 ) via Agrobacterium -mediated transformation. The qRT-PCR analysis demonstrated that the introduced four genes of the transgenic lines were all highly expressed. Through high-performance liquid chromatography analysis, the artemisinin contents were increased markedly in transformants, with the highest being 3.4-fold higher compared with non-converter. These results indicate that overexpression of multiple artemisinin biosynthetic pathway genes is a promising approach to improve artemisinin yield in A. annua .
Isolation of Dihydroartemisinic Acid from Artemisia annua L. By-Product by Combining Ultrasound-Assisted Extraction with Response Surface Methodology

Abstract:Malaria is the most devastating parasitic disease worldwide. Artemisinin is the only drug that can cure malaria that is resistant to quinine-derived drugs. After the commercial extraction of artemisinin from Artemisia annua, the recovery of dihydroartemisinic acid (DHAA) from artemisinin extraction by-product has the potential to increase artemisinin commercial yield. Here we describe the development and optimization of an ultrasound-assisted alkaline procedure for the extraction of DHAA from artemisinin production waste using response surface methodology. Our results using this methodology established that NaOH at 0.36%, extraction time of 67.96 min, liquid-solid ratio of 5.89, and ultrasonic power of 83.9 W were the optimal conditions to extract DHAA from artemisinin production waste. Under these optimal conditions, we achieved a DHAA yield of 2.7%. Finally, we conducted a validation experiment, and the results confirmed the prediction generated by the regression model developed in this study. This work provides a novel way to increase the production of artemisinin per cultivated area and to reduce artemisinin production costs by recycling its commercial waste to obtain DHAA, an immediate precursor of artemisinin. The use of this technology may reduce the costs of artemisinin-based antimalarial medicines.

Abstract:Artemisinin content in hairy roots of Artemisia annua was increased from 0.8 mg g −1 dry wt to 1 mg g −1 dry wt by using elicitor treatment of mycelial extracts from the endophytic fungus Colletotrichum sp. The increase of artemisinin was dependent on the growth stage of hairy roots as well as on the dose of the elicitor applied. When hairy roots of 23-day-old cultures (later growth phase) were exposed to the elicitor at 0.4 mg total sugar ml −1 for 4 days, the maximum production of artemisinin reached to 13 mg l −1 , a 44% increase over the control. This is the first report on the stimulation of artemisinin production in hairy roots by the elicitor from an endophytic fungus of A. annua .
