Abstract:An extracellular lipase was purified from the fermentation broth of Bacillus coagulans ZJU318 by CM-Sepharose chromatography, followed by Sephacryl S-200 chromatography. The lipase was purified 14.7-fold with 18% recovery and a specific activity of 141.1 U/mg. The molecular weight of the homogeneous enzyme was (32 kDa), determined by sodium dodecyl sulfate (SDS) polyacrylamide gel electrophoresis. The enzyme activity was maximum at pH 9.0 and was stable over a pH range of 7.0–10.0, and the optimum temperature for the enzyme reaction was 45°C. Little activity loss (6.2%) was observed after 1 h of incubation at 40°C. However, the stability of the lipase decreased sharply at 50 and 60°C. The enzyme activity was strongly inhibited by Ag+ and Cu2+, whereas EDTA caused no inhibition. SDS, Brij 30, and Tween-80 inhibited lipase, whereas Triton X-100 did not significantly inhibit lipase activity.

Abstract:A producing-fibrinolytic enzyme strain was isolated with high yield. The strain was identified as Bacillus amyloliquefaciens. B. amyloliquefaciens GUTU06 fibrinolytic enzyme was purified by acetone precipitation and reverse micelle. Acetone precipitation condition and reverse micelle condition were examined. Results showed that the total reverse micelle extraction efficiency was 64.49 % +/- 1.6 %. The purification fold of the entire process reached 13.38. The optimum pH of purified enzyme is 5, and the optimum temperature is 45 degrees C. Fe3+ and K+ can enhance the fibrinolytic activity of the enzyme. Compared to commercial fibrinolytic enzymes such as urokinase and lumbrukinase, GUTU06 fibrinolytic enzymes have a lower pH optimal range and higher temperature stability. The molecular weight of the enzyme was approximately 28 kDa. Reverse micelle extraction with cetyl trimethylammonium bromide as a surfactant combined with acetone precipitation is suitable for separating and purifying fibrinolytic enzymes and a promising technique for obtaining active proteins.

Abstract:A fibrinolytic enzyme was purified crude the crude extract of Ba - 2, a strain of Bacillus sp., through ammonium sulphate fractional pre cipitation, Sephadex G- 75 gel filtration, Q- Sepharose anion exchange preparative, preparative PAGE and electric elution.The purified component was shown to consist of three subunits with a molecular weight of approximately 23,22 and 19kD,respectively,and the molecular weight of the enzyme was about 64kD.The enzyme showed similar fibrinolytic activity on the fibrin plate and the plasminogen-free fibrin plate,suggesting that it may comtain both strong thrombolytic enzyme activity and plasminogen activator activity.

Abstract:Objective Purification of a fibrinolytic enzyme produced from Bacillus subtilis ZY-1 and investigation of its activity and characterization.MethodsA high fibrinolytic enzyme was purified with(NH4)2SO4 fractionation and sephadex G-100 gelfiltration chromatography,and characterization was investigated by check the enzymatic activities in different conditions.ResultsThe specific activity of enzyme was 26 400 U/mg,and its optimal temperature and pH was 45 ℃ and 8.0,respectively.The enzyme had fine thermal heat stability.ConclusionThe enzyme had potential commercial value.

Abstract:In order to obtain pure fibrinolytic enzyme and reveal the enzymatic properties, a fibrinolytic enzyme-producing strain with high fibrinolytic activity was screened form Douchi, a traditional Chinese soybean food and was identified as Bacillus subtilis. The mutant DC-12N8 of the strain was then processed in the following procedure; centrifugal sterilization, fractional sedimentation with ammonium sulfate, DEAE-Sepharose Fast Flow and CM-Seph-arose Fast Flow ion-exchange chromatography, Sephadex G-75 gel-filtration. Thus, a kind of fibrinolytic enzyme was purified from Douchi. As compared with the crude enzyme extraction, the specific activity of the purified enzyme increases by 52.6 folds with a recovery of 11.7%, and 4.5 mg of active enzyme protein with an activity of 1.18 mol/s can be obtained from 1 L of fermentation liquid. SDS-PAGE results show that the purified enzyme is a kind of single-train protein with a molecular mass of 28 ku.

Abstract:An antithrombus enzyme (ATE) was precipitated by (NH4)2SO4 or ethanol from supernatant of Bacillus subtilis culture broth then purified using ion exchange chromatography on CM-sepharose fast flow. The effects of ionic strength and pH value on protein adsorption, the gradient elution at different flow rates and step elution were examined respectively. The recovery yield of the optimised process was 74.5% with a purification factor 8.1. The ATE molecular weight was estimated as 30ku by SDS-PAGE. The experimental results showed that the enzyme was stable in the range of pH 7 to pH11, and temperature 25℃ to 37℃.

Abstract:Endophytes are a very important resource of microorganisms with potential application values.A novel fibrinolytic enzyme(PPFE-I) was purified from the fermentation supernatant of endophytic bacterium Paenibacillus polymyxa EJS-3 by the following procedure including ammonium sulfate precipitation,Hiprep phenyl FF hydrophobic chromatography,RESOURCEM Q ion exchange chromatography and Sephacryl S-300HR gel filtration.The apparent molecular mass of PPFE-I was estimated to be 63 kDa by SDS-PAGE analysis.The purity of PPFE-I was 94.1% determined by HPLC.Using the above procedure,16 mg of active proteins were obtained from 1 L of the fermentation supernatant of Paenibacillus polymyxa EJS-3 and a 14.5-fold increase in the specific activity of PPFE-I was achieved and the activity recovery was 3.3%.Meanwhile,the final purified product had good thrombolytic activity in vitro.

Abstract:The method of purifying a novel fibrinolytic enzyme subtilisin FS33 by gel chromography was investigated in this paper.The crude enzyme produced by Bacillus subtilis DC33 was precipitated by 30%～65% saturation of(NH4)2SO4,and the protein collected by centrifugation(10000×g,15min) was applied to chromatographic procedures including hydrophobic chromatography,anion exchange and gel filtration chromatography.The finally eluted proteins by Sephadex G-50 gel filtration were subjected to SDS-PAGE.Under these conditions,the purified rate and the recovery of final enzyme were 36.6 fold and 13.0%,respectively.The specific activity of the fibrinolytic enzyme was 15495U/mg,and only one band was observed in the purified sample under reducing or non-reducing conditions.The molecular mass was estimated to be approximately 30kDa.The higher fibrinolytic activities of the purified enzyme band were observed on the plasminogen-free fibrin plate.

Abstract:Bacillus subtilis WY34 secreted a dihydrolipoamide dehydrogenase.The crude enzyme solution was purified by weak ion-exchange chromatography and blue sepharose fast flow chromatography.The purified enzyme was of 59.7 times higher homogeneity.The recovering yield was 46.9%.The molecular masses,detected by SDS-PAGE and Superdex 75 gel filtration chromatography,were(64.6?ku) and(67.5?ku) respectively,which indicated that the enzyme was a monomeric protein.A simple and efficient two-step purification protocol resulted in an apparently homogenous enzyme preparation with a high yield.

Abstract:A fibrinolytic enzyme isolated from marine Bacillus subtilis HQS-3 was purified to electrophoretic homogeneity using ammonium sulphate precipitation, alkaline solution treatment, membrane concentration, dialysis, ion exchange, and gel filtration chromatography. SDS-PAGE and gel filtration chromatography showed that it was a monomeric protein with an apparent molecular weight of 26kDa. The purified enzyme was active at pH 6.0–10.0 with an optimum pH of 8.0. It was stable at temperatures ranging from 25 to 37°C, exhibiting maximum activity between 45°C and 50°C. The isoelectric point of the enzyme was 9.0–9.2, which was higher than those of other known fibrinolytic enzymes from Bacillus species. PMSF, EDTA, Cu2+, Zn2+, and Co2+ inhibited the enzyme activity significantly. This enzyme did not cause hemolysis in vitro and preferred direct degradation of fibrin in the following order: α, β, and γ–γ chains. Thus, these results suggest that the marine-derived enzyme is a plasmin-like serine metalloprotease, which is distinct from other fibrinolytic enzymes from genus Bacillus.

Abstract:Fermented bean foods are a crucial source of fibrinolytic enzymes. The presented study aimed to purify, characterize, and chemically modify Bacillus velezensis SN-14 fibrinolytic enzyme. The fibrinolytic enzyme was purified using CTAB/isooctane/hexyl alcohol/n-butyl alcohol reverse micellar system, and the purified enzymewas chemically modified to improve its enzymatic activity and stability. Enzyme activity recovery and the purification fold for this enzymewere 44.5 +/- 1.9% and 4.93 +/- 0.05 fold, respectively. SDS-PAGE results showed that themolecular weight of the purified fibrinolytic enzyme was around 28 kDa. Besides, the optimum temperature and pH of the purified fibrinolytic enzymewere 37 degrees C and 8-9, respectively. Fe2+, mPEG5000, and pepsinwere used for chemical modification and for improving the activity and stability of the purified enzyme. Thermal and acid-base stability of chemically modified enzymes increased significantly, whereas enzymatic activity increased by 7.3 times. After 30 d of frozen storage, the modified enzyme's activity was remarkably lower (33.2%) than the unmodified enzyme (60.6%). The current study on B. velezensis SN-14 fibrinolytic enzyme and chemicalmodificationmethod using Fe2+, mPEG5000, and pepsin provide a reference for developing fibrinolytic drugs and foods. Abstract:Bacillus subtilis DC33 producing a novel fibrinolytic enzyme was isolated from Ba-bao Douchi , a traditional soybean-fermented food in China. The strong fibrin-specific enzyme subtilisin FS33 was purified to electrophoretic homogeneity using the combination of various chromatographic steps. The optimum temperature, pH value, and p I of subtilisin FS33 were 55°C, 8.0, and 8.7, respectively. The molecular weight was 30 kDa measured by SDS–PAGE under both reducing and non-reducing conditions. The enzyme showed a level of fibrinolytic activity that was about six times higher than that of subtilisin Carlsberg. The first 15 amino acid residues of N-terminal sequence of the enzyme were A-Q-S-V-P-Y-G-I-P-Q-I-K-A-P-A, which are different from that of other known fibrinolytic enzymes. The amidolytic activities of subtilisin FS33 were inhibited completely by 5 mM phenylmethanesulfonyl fluoride (PMSF) and 1 mM soybean trypsin inhibitor (SBTI), but 1,4-dithiothreitol (DTT), β-mercaptoethanol, and p-hydroxymercuribenzoate (PHMB) did not affect the enzyme activity; serine and tryptophan are thus essential in the active site of the enzyme. The highest affinity of subtilisin FS33 was towards N-Succ-Ala-Ala-Pro-Phe-pNA. Therefore, the enzyme was considered to be a subtilisin-like serine protease. The fibrinolytic enzyme had a high degrading activity for the Bβ-chains and Aα-chain of fibrin(ogen), and also acted on thrombotic and fibrinolytic factors of blood, such as plasminogen, urokinase, thrombin, and kallikrein. So subtilisin FS33 was able to degrade fibrin clots in two ways, i.e., (a) by forming active plasmin from plasminogen and (b) by direct fibrinolysis.
Production and Characterization of Fibrinolytic Enzyme from Bacillus Amyloliquefaciens in 19 L Fermenter and Investigation of Its Properties

Abstract:As an extension of our previous fermentation experiments in shake flasks, the objective of this study was to optimize the fermentation conditions for the production of fi brinolytic enzyme by Bacillus amyloliquefaciens GZJSI-12-7 in a 19 L fermenter. The results showed that plasmin activity began to appear after incubation for 6 h and remained constant after 78 h at a level of 4 178.39 IU/mL. The peak of enzyme production occurred about 12 h earlier than in shake flasks. Fibrinolytic enzyme was purified from the fermented liquid medium by bactofugation, ammonium sulfate precipitation, dialysis and Sephadex G-75 gel filtration, with a purification fold of 9.84 and an activity recovery of 42.52%. The specific activity of the purified fibrinolytic enzyme was 48 073.193 IU/mg. Enzymatic characterization showed that the molecular weight of the fibrinolytic enzyme was about 30.2 kD, and its optimum temperature and pH were 40 ℃ and 7.5, respectively. The purified fibrinolytic enzyme was stable blow 40 ℃ but sharply decreased above 50 ℃. The enzymatic activity was markedly catalyzed by Mg2+, Ca2+ and Mn2+, but strongly inhibited by Cu2+ and Fe3+.
A thermostable lipolytic enzyme from a thermophilic Bacillus sp.: Purification and characterization

Abstract:A thermophilic Bacillus sp. was isolated that secreted an extracellular, thermostable lipolytic enzyme. The enzyme was purified to 58 folds with a specific activity of 9730 units/mg of protein and yield of 10% activity by ammonium sulphate precipitation, Phenyl Sepharose chromatography, gel-permeation followed by Q Sepharose chromatography. The relative molecular mass of the protein was determined to be 61 kDa by SDS-PAGE and approximately 60 kDa by gel permeation chromatography. The enzyme showed optimal activity at 60–65 ?C and retained 100% activity after incubation at 60 ?C and pH 8.0 for 1 h. The optimum pH was determined to be 8.5. It exhibited 50% of its original activity after 65 min incubation at 70 ?C and 23 min incubation at 80 ?C. Catalytic function of lipase was activated by Mg++ (10 mM), while mercury (10 mM) inactivated the enzyme completely. No effect on enzyme activity was observed with trypsin and chymotrypsin treatment, while 50% inhibition was observed with thermolysin. It was demonstrated that PMSF, SDS, DTT, EDTA, DEPC, ?ME (100 mM each) and eserine (10 mM) inhibited the activity of the lipolytic enzyme. With p-nitrophenyl laurate as a substrate, the enzyme exhibited a Km and Vmax of 0.5 mM and 0.139 ?M/min/ml. The enzyme showed preference for short chain triacylglycerol and hydrolyzes triolein at all positions. In contrast to other thermostable Bacillus lipases, this enzyme has very low content of hydrophobic amino acids (22.58 %). Immunological studies showed that the active site and antigen-binding site of enzyme do not overlap.

Abstract:A novel fibrinolytic enzyme subtilisin FS33 was purified from Bacillus subtilis DC33, isolated from a traditional flavour-rich food in China. The purified subtilisin FS33 was a single chain protein with a molecular mass of 30 kDa measured by SDS-PAGE. After activated SDS-PAGE, the enzyme band exhibited strong fibrinolytic activity on the fibrin plate. Subtilisin FS33 was temperature-stable below 60°C over the pH range 5–12, with a maximum activity at pH 8.0, but the activity completely disappeared after 10 min above 65°C. The NH 2 -terminal amino acid sequence of the enzyme was different from that of other known fibrinolytic enzymes, such as NK, CK, SMCE, KA38, subtilisin E, subtilisin DFE and Katsuwokinase. The amidolytic activities of subtilisin FS33 were inhibited completely by phenylmethanesulfonyl fluoride (PMSF) and soybean trypsin inhibitor (SBTI). EDTA did not affect the enzyme activity, and none of the ions tested activated the activity. Therefore, the enzyme was thought to be a subtilisin-like serine protease. The enzyme degraded the Bβ-chains of fibrin(ogen) very rapidly and then degraded the Aα-chain and at least five fragments from fibrin(ogen) were obtained after hydrolysis. Subtilisin FS33 was also able to cleave blood clots in the absence of endogenous fibrinolytic factors.

Abstract:A fibrinolytic enzyme was purified from the dry body of Whitmania pigra Whitman. The fibrinolytic enzyme was purified to homogeneity with a yield of 0.003% and a purification of 630.7 fold. The molecular weight of the enzyme was estimated to be 26.7 kDa by reduced sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). The enzyme was tested by matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF-MS) and it showed that the enzyme was a novel fibrinolytic enzyme. The optimal pH and temperature of the enzyme were 8.5 and 55 degrees C, respectively. Enzyme activity was enhanced by Na+, Mg-2+,Mg- and K+. On the contrary, the proteolytic activity was significantly inhibited by Mn2+, Fe2+, Fe3+, ethylenediaminetetraacetic acid (EDTA), and ethylenebis(oxyethylenenitrilo)tetraacetic acid (EGTA). Fibrinolytic and fibrinogenolytic assays showed that the enzyme preferentially hydrolyzed fibrinogen A-chains, followed by B- and -chains. The -, -, and --chains of fibrin were also degraded by the enzyme.
The Purification and N-Terminal Sequencing of the Lipase from Penicillium Expansum WMC20718

Abstract:The alkaline lipase produced from penicillium expansum WMC20718 was purified 18.5 fould with the recovery of 48.8% by centrifugation, ammonium sulfate precipitation, phenyl Sepharose CL 4B, DEAE Sepharose and Sephadex G 75 column chromatographies. The purified lipase had a specific activity of 4 200 U/mg protein, which were showed to be homogeneous by SDS Page and RP HPLC. The molecular weight of the native lipase was estimated to be about 28.2 kD by gel filtration using Sephades G 150, and that of the denatured lipase about 27.5 kD by SDS PAGE, indication that the lipase was a monomer. The N terminal amino acid sequence was determined as follows: A T A D A A A F P D L H R A A K L S S A. The optimum temperature of the lipase was 30 ℃ and was stable below 35 ℃, but only 30% of its activity remained after 30 min incubation at 45 ℃. The lipase was stable at pH from 6.5 to 10.5, and its optimal pH was 10.0. Anionic surfactant LAS affected the enzymatic stability greatly, but non ionic surfactant AEO 9 had little effect and could lessen the effect of LAS. The alkaline lipase showed resistance to alkaline protease in proper concentration.

The purified enzyme showed a single band on PAGE and a level of fibrinolysis activity about 5.16 times that of the crude enzyme. The molecular weight was 12.6 kD determined by SDSげ PAGE. The isoげ electric point(pI) was proved to be 7.45 by fast polyacrylamide isoげ electric focus electrophoresis. The fibrinolysis activity was determined by aragoseげ fibrous protein plate method. The optimum temperature and pH were 50 → and 8.0 respectively. It is stable in the basic solution. According to the control of activity of this enzyme between positive and negative plate, this enzyme had high affinity to fibrin, and the ability of activating plasminogen was low. In vitro, this enzyme had strong ability to hydrolyze artificial thrombus.

Abstract:The purification of elastase was achieved by combination methods of ammonium sulfate precipitation,ion-exchange chromatography and gel filtration chromatography.The elastase was purified 13.79-fold to apparent homogeneity with 7.88% overall recovery.It was a single-chain protein with a molecular mass of 31ku,and estimated by SDS-PAGE and gel filtration.Studies on enzymatic properties showed as follows:the elastase belonged to metalloproteinase containing Ca2+.The elastase activity had a temperature optimum of 50℃ along with good thermal stability.Meanwhile it was optimal at pH 7.4 in borax-boric acid buffer or pH 8.6 in Tris-HCl,and tended stability between 8.0 and 11.0.10mmol/L Ca2+ had stabilizing and activation on the elastase activity,whereas SDS and Tween-80 only had activation.Moreover,the elastase activity was inhibited varying degrees by many irons(Li+,Na+,Zn+,K+,Ba2+,Mg2+,Mn2+)and EDTA.The catalyzes dynamics equation was:y=0.186x+ 32.493,Vmax =0.0308 U/mL,Km=0.005 7g/mL.
