Expression of PTEN Protein and Phosphorylated Protein Kinase B in Endometrial Adenocarcinoma Tissue

Abstract:Aim:To examine the expression of PTEN and phosphorylated protein kinase B(P-AKT)in endometrial adenocarcinoma tissue.Methods:The expressions of PTEN and P-AKT in 12 cases of proliferative endometrium,42 cases of endometrial hyperplasia including 15 cases of simple hyperplasia(SH),19 cases of complex hyperplasia(CH) and 8 cases of atypical hyperplasia(AH),and 46 cases of endometrial adenocarcinoma were detected by immunohistochemistry(SP).Results:The positive expression rate of PTEN declined from proliferative endometrium(91.7%),endometrial hyperplasia(42.9%) to endometrial adenocarcinoma(37.0%)(χ2=11.64,P=0.003).The positive expression rate of PTEN in SH tissue was 73.3%,significantly higher than that in CH tissue(26.3%) and AH tissue(25.0%)(P=0.014).The positive expression rate of P-AKT showed an increased trend in proliferative endometrium(25.0%),endometrial hyperplasia(57.1%),and endometrial adenocarcinoma(87.0%)(χ2=19.37,P0.001).From SH,CH,to AH,the positive expression rates of P-AKT were 26.7%,68.4% and 87.5%,respectively.The positive rate of P-AKT in SH and CH tissue was lower than that in AH tissue(χ2=5.28,P=0.027).The expression of PTEN was related with histological grades(P0.001),but not with FIGO stage(χ2=0.06,P=0.89).P-AKT was not related with histological grades or FIGO stage in endometrial carcinoma(χ2=0.02,P=0.82).The expression of PTEN was related with P-AKT in endometrial adenocarcinoma(rP=-0.368,χ2=6.69,P=0.012).Conclusion:PTEN gene and P-AKT are involved in the carcinogenesis and development of endometrial adenocarcinomas.
