Abstract:The aim of this study was to evaluate the capability of using micro-magnetic resonance imaging (MRI) to visualize and characterize atherosclerotic plaques of mouse models. Twenty five apolipoprotein E-knockout mice were fed atherogenic diet, which enabled creation of aortic atherosclerotic plaques. Aortic plaques were examined in vivo by 4.7 T MRI and then characterized ex vivo by 11.7 T three-dimensional MRI. MR images were correlated with subsequent histological confirmation. In vivo 4.7-T MRI demonstrated unevenly thickened aortic walls due to formation of atherosclerotic plaques. Ex vivo 11.7-T MRI enabled not only to acquire full volume-rendered images of the entire vessels but also to characterize plaque components (such as lipid cores and fibrous caps) at any level and any projection, which were confirmed by histological correlation. Micro-MRI provides an excellent imaging tool for basic science to investigate atherosclerosis in small animal models, which may become a supplement to histopathology of atherosclerotic cardiovascular disease.

Abstract:Objective To investigate the possibility of superparamagnetic iron-oxide(SPIO) particles as a marker of atherosclerotic plaques detected with 1.5T magnetic resonance imaging system in New Zealand white rabbit model.Methods Model of atherosclerosis were established by injuring the right carotid artery endothelium with balloon and feeding with high lipid diet.Five rabbits(group A) were intravenously injected SPIO;and the other five rabbits(group B) received the same volume of saline injection.The MR imaging was performed at 24,48 and 72 hours after injection,and then,histological study was done.Results Twenty-four hours after the intravenous injection of SPIO,3 rabbits in group A showed signal intensity loss in the right carotid artery wall on T2WI,corresponding to Perl's Prussian blue staining-positive particles in the plaques revealed by histopathological study.T2WI change and Perl's Prussian blue staining-positive particles in the plaques were not observed in any one model in group B.Conclusion It is possible to use superparamagnetic iron-oxide (SPIO) particles as a marker of atherosclerotic plaques detected with 1.5T magnetic resonance imaging system in New Zealand white rabbit model.

Abstract:Magnetic resonance imaging (MRI) is a useful tool for non-invasive identification and characterization of atherosclerotic plaques in both basic science and clinical practice. To date, the reported studies on in vivo vascular MRI of small animals, such as mice and rats, are mainly performed on high-field micro-MR scanners, which are not always available in many academic institutions and basic research units. This study aimed to explore the possibility of generating high-resolution MR images of the atherosclerotic aortic walls/plaques of mice using a clinical 3.0 T MR scanner with a dedicated solenoid mouse coil. An atherosclerotic mouse model was first generated by feeding 8 ApoE − / − mice an atherogenic diet. MR images of the ascending aortas of these mice were then achieved using a three-dimensional black-blood turbo spin-echo sequence (repetition time TR = 4 heart echo time TE = 10 ms). The MRI displayed a clear view of the aortic lumens and the atherosclerotic lesions, which correlated significantly well with subsequent histological confirmations (linear regression analysis, r = 0.73, P = 0.04). This study demonstrated that a clinical 3.0 T MR scanner can be used for high-resolution imaging of mouse atherosclerotic lesions to some extent.

Abstract:Background: It has been confirmed that the α7-nicotinic acetylcholine receptor (α7nAChR) is an important target for identifying vulnerable atherosclerotic plaques. Previously, we successfully designed and synthesized a series of 18 F-labeled PET molecular probes targeting α7nAChR, which are mainly used in the diagnosis of Alzheimer's disease. Based on the characteristics of α7nAChR in blood vessels, we have firstly screened for a suitable novel 18 F-labeled PET molecular probe ([ 18 F]YLF-DW), with high selectivity for α7nAChR over α4β2nAChR and a good effect for the imaging of atherosclerotic animal models, to effectively identify vulnerable atherosclerotic plaques at an early stage. Meanwhile, we compared it with the “gold standard” pathological examination of atherosclerosis, to verify the reliability of [ 18 F]YLF-DW in early diagnosis of atherosclerosis. Methods: The vulnerable atherosclerotic plaques model of ApoE-/-mice were successfully established. Then based on the methods of 3D-QSAR and molecular docking, we designed oxazolo[4,5-b] pyridines and fluorenone compounds, which are targeted at α7nAChR. Through further screening, a novel alpha7 nicotinic acetylcholine receptor radioligand ([ 18 F]YLF-DW) was synthesized and automatically 18 F-labeled using a Stynthra RNplus module. Subsequently, we employed [ 18 F]YLF-DW for the targeting of α7nAChR in atherosclerotic plaques and control group, using a micro-PET/CT respectively. After imaging, the mice were sacrificed by air embolism and the carotid arteries taken out for making circular sections. The paraffin embedded specimens were sectioned with 5 μm thickness and stained with oil red. After staining, immunohistochemistry experiment was carried out to verify the effect of micro-PET/CT imaging. Results: The micro-PET/CT imaging successfully identified the vulnerable atherosclerotic plaques in the carotid arteries of ApoE-/-mice; whereas, no signal was observed in normal control mice. In addition, compared with the traditional imaging agent [ 18 F]FDG, [ 18 F]YLF-DW had a significant effect on the early plaques imaging of carotid atherosclerosis. The results of oil red staining and immunohistochemistry also showed early formations of carotid plaques in ApoE-/-mice and provided pathological bases for the evaluation of imaging effect. Conclusion: We innovated to apply the novel molecular probe ([ 18 F]YLF-DW) to the identification of vulnerable atherosclerotic plaques in carotid arteries, to detect atherosclerosis early inflammatory response and provide powerful input for the early diagnosis of atherosclerotic lesions, which may play an early warning role in cardiovascular acute events.

Inhibited Atherosclerotic Plaque Formation by Local Administration of Magnetically Labeled Endothelial Progenitor Cells (epcs) in a Rabbit Model.

Abstract:Purpose: To investigate whether atherosclerosis can be prevented by magnetically labeled endothelial progenitor cells (EPCs) in rabbits.Materials anti methods: EPCs derived from rabbit periphery blood were labeled with a superparamagnetic iron oxide (SPIO) agent Fe2O3-poly-L-lysine (Fe2O3-PLL). Rabbit atherosclerosis was induced by high-cholesterol-diet following balloon injury via catheterization of right common carotid artery (RCCA). Fe2O3-PLL labeled EPCs (2 x 10(6)) and media were allowed to interact with the RCCA for 25 min in EPC-treated rabbits (n = 14) and control rabbits (n = 7) animals respectively. MRI was performed with a 1.5 T-magnet to measure RCCA signal intensity (SI) and caliber at week 1, 2, 3, 6,12, and 15 with animals euthanized in groups for histopathology.Results: In EPC-treated rabbits, T-2-weighted MRI showed SI loss in RCCA at week I and 2 followed by normalization after week 3. MRI outcomes corresponded well to findings of Prussian blue staining. MRI at week 6,12 and 15 showed little stenosis of RCCA in EPC-treated rabbits, but moderate to severe stenoses in control rabbits. Histology at week 15 revealed significantly thinner RCCA wall (277.62 mu m vs. 382.95 mu m, P= 0.026), greater internal diameter (913.33 mu m vs. 789.64 mu m, P= 0.037) and smaller plaque (398.60 mm(2) vs. 597.70 mm(2), P = 0.047) in EPC-treated rabbits relative to control rabbits.Conclusion: Atherosclerosis at RCCA was inhibited by SPIO-labeled EPCs, which was depicted with a clinical MRI scanner over 2 weeks after cell administration, suggesting that EPCs may play a role in restoration of endothelial injury and prevention of atherosclerosis. Abstract:Macrophage recruitment has emerged as the crucial force driving the initiation and progression of atherosclerotic lesions. Therefore, the identification of macrophages in plaques is of vital importance for identifying vulnerable plaques, and noninvasive imaging methods are particularly desirable. Some studies have reported that MRI can detect plaque macrophages through targeted nanoparticles, but it is still hard for an US to detect macrophages in atherosclerotic plaque. In this study, anti-CD68 receptor-targeted Fe-doped hollow silica nanoparticles (CD68-Fe-HSNs) were fabricated as a dual-modal US/MRI contrast agent for identifying macrophages of aorta ventralis atherosclerotic plaques in ApoE-/- mice, confirmed by immunofluorescence and bio-TEM. This system possesses biodegradable characteristics even though it is an inorganic mesoporous nanosystem, indicating its potential high biocompatibility for further in vivo research. We expect that these dual-modal US/MRI nanoparticles will play a role in assessing vulnerable plaque in future research studies.

Abstract:Aim: The diagnosis of vulnerable atherosclerotic plaques remains challenging. This study labeled ultrasmall superparamagnetic iron oxide with rhodamine (USPIO-R) and evaluated USPIO-R for imaging atherosclerotic plaques. Methods: Apolipoprotein E-deficient mice were fed a high-fat diet and underwent MRI before and after an intravenous injection of USPIO-R. Subsequently, an aortic specimen from the mice was removed and sliced for fluorescence imaging and Prussian blue and immunofluorescent staining. Results: T-2 signal loss appeared and persisted in the aortic plaque postinjection, and spontaneous fluorescence from the plaque was observed. The accumulated mechanism of USPIO-R by plaque was the macrophage internalization by Prussian blue and immunofluorescence. Conclusion: USPIO-R is a promising dual-modality probe for diagnosing and monitoring vulnerable atherosclerotic plaques.
In vivo MR imaging of bone marrow cells trafficking to atherosclerotic plaques.

Abstract:Purpose: To develop a magnetic resonance imaging (MRI)-based method to monitor in vivo trafficking of bone marrow (BM) cells to atherosclerotic lesions. Materials and Methods: BM cells from LacZ-transgenic mice were labeled with a supermagnetic iron oxide (Feridex) and then transplanted into ApoE(-/-) recipient mice that were fed an atherogenic diet. Twenty-four ApoE(-/-) mice were divided into three study groups: 1) group I with Feridex-labeled BM transplantation (BMT) cells (N = 9), 2) group II with unlabeled BMT cells (N = 10), and 3) group III with no BMT cells (N = 5). Migrated Feridex/LacZ-BM cells to atherosclertic aortic walls were monitered in vivo using a 4.7T MR scanner and correlated with histopathological findings. Results: In group I with Feridex-BMT cells, histology examination displayed plaques in five of nine animals. In four of these five animals, in vivo MRI showed large MR signal voids of the aorta walls (due to the "blooming" effect of migrated Feridex-BM cells in plaques), which were correlated with Feridex- and/or LacZ-positive cells detected in the atherosclerotic lesions. No signal voids could be visualized in the two control animal groups (groups II and III). Conclusion: This study demonstrates the potential use of in vivo MRI to monitor the trafficking of magnetically labeled BM cells to atherosclerotic lesions.
OxLDL-targeted Iron Oxide Nanoparticles for in Vivo MRI Detection of Perivascular Carotid Collar Induced Atherosclerotic Lesions in ApoE-deficient Mice

Abstract:Atherosclerotic disease is a leading cause of morbidity and mortality in developed countries, and oxidized LDL (OxLDL) plays a key role in the formation, rupture, and subsequent thrombus formation in atherosclerotic plaques. In the current study, anti-mouse OxLDL polyclonal antibody and nonspecific IgG antibody were conjugated to polyethylene glycol-coated ultrasmall superparamagnetic iron oxide (USPIO) nanoparticles, and a carotid perivascular collar model in apolipoprotein E-deficient mice was imaged at 7.0 Tesla MRI before contrast administration and at 8 h and 24 h after injection of 30 mg Fe/kg. The results showed MRI signal loss in the carotid atherosclerotic lesions after administration of targeted anti-OxLDL-USPIO at 8 h and 24 h, which is consistent with the presence of the nanoparticles in the lesions. Immunohistochemistry confirmed the colocalization of the OxLDL/macrophages and iron oxide nanoparticles. The nonspecific IgG-USPIO, unconjugated USPIO nanoparticles, and competitive inhibition groups had limited signal changes (p < 0.05). This report shows that anti-OxLDL-USPIO nanoparticles can be used to directly detect OxLDL and image atherosclerotic lesions within 24 h of nanoparticle administration and suggests a strategy for the therapeutic evaluation of atherosclerotic plaques in vivo.

Abstract:Pro-inflammatory M1 macrophage is identified as a prominent component initializing the progress of vulnerable atherosclerotic plaque. Here, we constructed anti-MARCO NaGdF4:Yb,Er@NaGdF4 upconversion nanoparticles (UCNPs) by conjugating polyclonal MARCO antibody to the surface of NaGdF4:Yb,Er@NaGdF4via condensation reaction. UCNPs displayed highly mono-dispersion with average sizes of 26.7 ± 0.8 nm and favorable biocompatibility. In vivo upconversion optical imaging revealed that distinctive fluorescence signal could be observed in the regions of carotid artery 10 min post-injection, reached peak value at 1 h and decreased back to baseline at 24 h post-injection. The carotid artery wall demonstrated high signal intensity on T1-weighted MR images after anti-MARCO UCNPs injection, as determined by 7.0T MRI. Immunofluorescence staining of tissue section of carotid artery revealed that MARCO was highly abundant in shoulder regions of plaque. Anti-MARCO UCNPs is a promising optical/MRI dual-modality imaging probe which can non-invasively reflect M1 phenotype macrophages behavior in vivo.

Abstract:Abstract Background Cardiovascular disease is the leading cause of death in the world. The majority of cardiovascular events result from the rupture of vulnerable atherosclerotic plaques, which are characterized by high and active macrophage content. The integrin αVβ3 is expressed by activated macrophages and endothelial cells in atherosclerotic lesions and thus is a marker of high-risk plaques. Therefore, 89Zr-RGD-melanin nanoparticle (MNP) positron emission tomography (PET)/photoacoustic imaging (PAI) imaging of αVβ3 expression in plaques might provide a novel noninvasive biomarker of plaque vulnerability. Purpose In this study, the intrinsic photoacoustic signals and the native strong chelating properties with metal ions of MNP, positron-emitting metal ions 89Zr and αVβ3 integrins targeting ability of cyclic c (RGDfC) peptide was employed to construct an efficient nanoplatform. And we evaluated the feasibility of 89Zr-RGD-MNP PET/PAI of αVβ3 expression in vivo and in vitro. Methods We conjugated αVβ3 integrins, cyclic c (RGDfC) peptide, to MNP and chelated the long-lived positron-emitting nuclide 89Zr. The bio-stability and targeting action was detected in macrophages. And the PET/PAI imaging was performed in apoE−/− mice with partial carotid ligation leading to atherosclerosis. In PET imaging, tracer uptake was measured in the stenotic areas of the carotid arteries, as well as on the contralateral side at different time points in vivo. In PAI, photoacoustic signal was measured in the atherosclerosis of the carotid arteries in vivo. Melanin staining and immunohistochemistry of αVβ3 expression were detected in atherosclerosis of the carotid arteries. Results 89Zr-RGD-MNP showed excellent bio-stability and targeting action. PET imaging showed specific tracer accumulation at plaques in the left carotid artery, confirmed by competitive receptor blocking studies and the contrast in the right carotid artery. In the biodistribution studies, the left carotid (5.29%±0.78%) showed higher uptake than the right carotid (2.11%±1.55%). PAI showed the PA signal in the surgery group (452±85 a.u.) were stronger than the control (156±45 a.u.) and blocking group (254±66 a.u). The result was consistent with PET imaging and the presence of nanoparticles, as indicated by pathological examinations. These results presented good in vivo multimodality imaging (PET/PAI) properties. Conclusions We have developed 89Zr-labeled atherosclerotic plaques imaging agents based on the natural melanin nanoparticle. 89Zr-RGD-MNP demonstrates specific tracer accumulation in mice atherosclerotic carotid plaques. In this model, its uptake was associated with αVβ3 expression. 89Zr-RGD-MNP is a potential tracer for noninvasive imaging in atherosclerosis. Imaging Carotid Plaque Burden in living mice via Hybrid Semiconducting Polymer Nanoparticles Based Near-Infrared-II Fluorescence and Magnetic Resonance Imaging

Abstract:The majority of atherothrombotic events (e.g., cerebral or myocardial infarction) often occur as a result of plaque rupture or erosion in the carotid, and thereby it is urgent to assess plaque vulnerability and predict adverse cerebrovascular events. However, the monitoring evolution from stable plaque into life-threatening high-risk plaque in the slender carotid artery is a great challenge, due to not enough spatial resolution for imaging the carotid artery based on most of reported fluorescent probes. Herein, copolymerizing with the small molecules of acceptor-donor-acceptor-donor-acceptor (A-D-A′-D-A) and the electron-donating units (D′), the screened second near-infrared (NIR-II) nanoprobe presents high quantum yield and good stability, so that it enables to image slender carotid vessel with enough spatial resolution. Encouragingly, NIR-II nanoprobe can effectively target to intraplaque macrophage, meanwhile distinguishing vulnerable plaque in carotid atherosclerosis in living mice. Moreover, the NIR-II nanoprobe can dynamically monitor the fresh bleeding spots in carotid plaque, indicating the increased risk of plaque instability. Besides, magnetic resonance imaging is integrated with NIR-II fluorescence imaging to provide contrast for subtle structure (e.g., narrow lumen and lipid pool), via incorporating ultrasmall superparamagnetic iron oxide into the NIR-II nanoprobe. Thus, such hybrid NIR-II/magnetic resonance imaging multimodal nanoprobe provides an effective tool for assessing carotid plaque burden, selecting high-risk plaque, and imaging intraplaque hemorrhage, which is promising for reducing cerebral/ myocardial infarction-associated morbidity and mortality.

Abstract:Plaque rupture is a critical concern due to its potential for severe outcomes such as cerebral infarction and myocardial infarction, underscoring the urgency of noninvasive early diagnosis. Magnetic resonance imaging (MRI) has gained prominence in plaque imaging, leveraging its noninvasiveness, high spatial resolution, and lack of ionizing radiation. Ultrasmall iron oxides, when modified with polyethylene glycol, exhibit prolonged blood circulation and passive targeting toward plaque sites, rendering them conducive for MRI. In this study, we synthesized ultrasmall iron oxide nanoparticles of approximately 3 nm via high-temperature thermal decomposition. Subsequent surface modification facilitated the creation of a dual-modality magnetic resonance/fluorescence probe. Upon intravenous administration of the probes, MRI assessment of atherosclerotic plaques and diagnostic evaluation were conducted. The application of Flash-3D sequence imaging revealed vascular constriction at lesion sites, accompanied by a gradual signal amplification postprobe injection. T1-weighted imaging of the carotid artery unveiled a progressive signal ratio increase between plaques and controls within 72 h post-administration. Fluorescence imaging of isolated carotid arteries exhibited incremental lesion-to-control signal ratios. Additionally, T1 imaging of the aorta demonstrated an evolving signal enhancement over 48 h. Therefore, the ultrasmall iron oxide nanoparticles hold immense promise for early and noninvasive diagnosis of plaques, providing an avenue for dynamic evaluation over an extended time frame.

Abstract:Background: Atherosclerosis is a chronic vascular inflammatory procedure alongside with lipid efflux disorder and foam cell formation. alpha 7-Nicotinic acetylcholine receptor (alpha 7nAChR) is a gated-calcium transmembrane channel widely expressed in neuron and non-neuron cells, such as monocytes and macrophages, activated T cells, dendritic cells, and mast cells. F-18-ASEM is an inhibitor targeted to alpha 7nAChR that had been successfully applied in nervous system diseases. Previous studies had highlighted that alpha 7nAChR was related to the emergency of vulnerable atherosclerotic plaques with excess inflammation cells. Thus, F-18-ASEM could be a complementary diagnostic approach to atherosclerotic plaques. Materials and Methods: The synthesis of ASEM precursor and F-18-labeling had been performed successfully. We had established the ApoE(-/-) mice atherosclerotic plaques model (fed with western diet) and New Zealand rabbits atherosclerotic models (balloon-sprained experiment and western diet). After damage of endothelial cells and primary plaque formation, F-18-ASEM imaging of atherosclerotic plaques linked to alpha 7nAChR had been conducted. In vivo micro-PET/CT imaging of ApoE(-/-) mice and the control group was performed 1 h after injection of F-18-ASEM (100-150 mu Ci); PET/CT imaging for rabbits with atherosclerotic plaques and control ones was also performed. Meanwhile, we also conducted CT scan on the abdominal aorta of these rabbits. After that, the animals were sacrificed, and the carotid and abdominal aorta were separately taken out for circular sections. The paraffin-embedded specimens were sectioned with 5 mu m thickness and stained with hematoxylin-eosin (H&E) and oil red. Results: In vivo vessel binding of F-18-ASEM and alpha 7nAChR expression in the model group with atherosclerosis plaques was significantly higher than that in the control group. PET/CT imaging successfully identified the atherosclerotic plaques in ApoE(-/-) mice and model rabbits, whereas no obvious signals were detected in normal mice or rabbits. Compared with F-18-FDG, F-18-ASEM had more significant effect on the early monitoring of inflammation in carotid atherosclerotic plaques of ApoE(-/-) mice and model rabbits. F-18-ASEM had relatively more palpable effect on the imaging of abdominal aorta with atherosclerosis in rabbits. H&E and oil red staining identified the formation of atherosclerotic plaques in model animals, which provided pathological basis for the evaluation of imaging effects. Conclusion: We first confirmed F-18-ASEM as radiotracer with good imaging properties for precise identification of atherosclerotic diseases.

Detection of Vulnerable Atherosclerosis Plaques with a Dual-Modal Single-Photon-Emission Computed Tomography/Magnetic Resonance Imaging Probe Targeting Apoptotic Macrophages

Abstract:Atherosclerosis (AS), especially the vulnerable AS plaque rupture-induced acute obstructive vascular disease, is a leading cause of death. Accordingly, there is a need for an effective method to draw accurate predictions about AS progression and plaque vulnerability. Herein we report on an approach to constructing a hybrid nanoparticle system using a single-photon-emission computed tomography (SPECT)/magnetic resonance imaging (MRI) multimodal probe, aiming for a comprehensive evaluation of AS progression by achieving high sensitivity along with high resolution. Ultrasmall superparamagnetic iron oxide (USPIO) was covered by aminated poly(ethylene glycol) (PEG) and carboxylated PEG simultaneously and then functionalized with diethylenetriaminepentacetate acid for 99mTc coordination and subsequently Annexin V for targeting apoptotic macrophages abundant in vulnerable plaques. The in vivo accumulations of imaging probe reflected by SPECT and MRI were consistent and accurate in highlighting lesions. Intense radioactive signals detected by SPECT facilitated focus recognization and quantification, while USPIO-based T2-weighted MRI improved the focal localization and volumetry of AS plaques. For subsequent ex vivo planar images, targeting effects were further confirmed by immunohistochemistry, including CD-68 and TUNEL staining; meanwhile, the degree of concentration was proven to be statistically correlated with the Oil Red O staining results. In conclusion, these results indicated that the Annexin V-modified hybrid nanoparticle system specifically targeted the vulnerable AS plaques containing apoptotic macrophages and could be of great value in the invasively accurate detection of vulnerable plaques.

Abstract:Stroke is one of the top three fatal diseases in human history. Inflammatory injury of the artery endo-membrane system is widely accepted as the major starting mechanism for the formation and enlargement of atherosclerosis plaque, which can be diagnosed by B-ultrasound or carotid angiography. However, clinical data reveal that there are asymptomatic patients that are not diagnosed until the stenosis degree is over 70%. Superparamagnetic iron oxide (SPIO), a promising candidate for a next generation contrast agent in medical imaging, has been used in the imaging of carotid artery plaques, but it still faces the challenge of targeted enrichment. Herein, we introduce a mature magnetic nanoparticle contrast agent and a promising method for diagnosing carotid artery inflammatory plaques with a Magnetic Resonance Imaging (MRI) technique. The superparamagnetic nanoparticles (SNPs) were synthesized by directly coating hydrophilic and high-magnetism Fe3O4 nanoparticles with an organosilica layer, which was followed by modification with polyethylene glycol. It was verified that the resultant nanocomposite possesses a higher structural stability, excellent dispersivity, separability, as well as biosafety. More importantly, the strong magnetism was preserved, making it possible to attract the SNPs with an external magnetic field and achieve a target concentration within the lesion. Moreover, an in vitro magnetic collar, designed to produce a stable magnetic field around the superficial common carotid arteries was introduced. The SNPs particles in the blood flow were slowed down by the collar, their motion direction was changed, and they were captured by the inflammatory cells in the plaque. The effectiveness and feasibility of the particles were evaluated via testing the MRI performance on histological levels with a rat carotid plaque model. The SNPs that were concentrated and accumulated in the plaque were verified to present an evident, negative enhancement in the Proton Density-T2 (PD-T2) sequence images. Therefore, it was demonstrated that the superparamagnetic nanoparticles have great potential as an MRI contrast agent to detect early stage carotid artery inflammatory plaques with an external magnetic collar.
Comparing 18F-Fdg Pet and 99Mtc-Labeled Ultrasmall Superparamagnetic Iron Oxide-Conjugated Annexin V Spect/Ct/Mr Imaging Apoptosis in Atherosclerotic Plaques

Abstract:1705 Objectives Considering macrophage apoptosis is tightly related to the vulnerability of atherosclerosis plaque and annexin V could be targeted to apoptosis cells, thus it would be interested to design a corresponded probe. However, limited resolution of nuclear medicine instrumentations hindered accurate localization of plaque, in this connection, we are trying to design a hybrid ultrasmall superparamagnetic iron oxide (USPIO)-Annexin V probe allowing for both MRI imaging with high resolution and SPECT imaging with high sensitivity and capability of quantification. Methods USPIO nanoparticles with the average diameters of less than 20 nm were synthesized, thereafter, they were modified with DTPA,PEG and conjugated with Annexin V. The desired DTPA-USPIO-Annexin V molecule was labeled with 99mTc at pH 8.4. This probe was injected into ApoE-/- mice feeding with high cholesterol diet for 3 months, and micro-SPECT/CT were performed 5 h post injection. The same mice were performed MRI imaging using T2-weighted sequences before injection of this probe and again after micro-SPECT/CT imaging. Two days later, 18F-FDG PET was done with the same mice. To better comparing, the above experiments have been carried out on C57 control mice with the same protocol. Results Both 18F-FDG PET and 99mTc-DTPA-USPIO-Annexin V SPECT/CT showed focused uptake on plaque sites, especially on aorta arch of ApoE-/- mice. However, 18F-FDG PET signal was largely disturbed by higher background, 99mTc-DTPA-USPIO-Annexin V SPECT showed more intense uptake on plaque and clearer background. Moreover, higher resolution MRI images confirmed our nuclear medicine images and showed apparent signal changing on aorta arch site after receiving this probe in ApoE-/- mice, but not in C57 mice. Conclusions This preliminary data show 99mTc-DTPA-USPIO-Annexin V could be targeted to atherosclerosis plaque as a hybrid SPECT/MRI probe. 18F-PBR06 PET/CT Imaging for Evaluating Atherosclerotic Plaques Linked to Macrophage Infiltration

Abstract:Background The present study explored the 18 kDa translocator protein radioligand [F-18]N-fluoroacetyl-N-(2,5-dimethoxybenzyl)-2-phenoxyaniline (F-18-PBR06) targeting macrophages for PET imaging of atherosclerotic plaques and evaluating the vulnerability of atherosclerotic plaques toward rupture. Materials and methods F-18-PBR06 was synthesized using a Synthra RNplus module automatically. RAW264.7 cells were used for cell binding study with F-18-PBR06. In-vivo micro-PET/CT imaging for ApoE(-/-) mice and C57 mice was performed 1 h after injection of F-18-PBR06. CD68(+) and F480 immunofluorescence stainings were performed in the aorta tissues. Results In-vitro cell studies showed uptake of F-18-PBR06 to RAW264.7 cells. Micro-PET/CT imaging identified the atherosclerotic lesions in the aortic arch of ApoE(-/-) mice successfully, whereas no signal was observed in C57 mice. The ratio of plaque-to-muscle in ApoE(-/-) mice of 32 weeks was significantly higher than that in ApoE(-/-) mice of 22 weeks, which was confirmed by CD68(+) immunofluorescence staining and F480 immunofluorescence staining. Conclusion TSPO radioligand F-18-PBR06 allows noninvasive PET/CT imaging of macrophage-abundant atherosclerotic plaques as well as positive correlation between PET imagings and ex-vivo immunofluorescence staining of plaques in mice with different ages, thereby representing a potential attractive tool for evaluating the vulnerability of atherosclerotic plaques towards rupture.

Abstract:Active foamy macrophage enrichment drives atherosclerotic plaque initiation and evolution, and is the prominent target for precisely identifying vulnerable plaque. Precise imaging of high-risk plaque allows promotion of treatment and prevention of vascular pathema. However, current iron oxide (IO) nanoparticles-based magnetic resonance (MR) imaging of plaque is often limited by insufficient perfusion and nonspecific accumulation of peri-aortic lymph nodes. Besides that, intrinsic defects of MR also impede its use for accurately identifying plaque details. Herein, by conjugating with PP1 peptide, a novel magnetic mesoporous silica nanoparticle (PIMI) loaded with near-infrared fluorescence (NIRF) dye (IR820) was fabricated to specifically target and quantify macrophage enrichment of atherosclerotic plaque in ApoE−/− mice using dual MR/NIRF imaging. Biocompatibility experiments ulteriorly confirmed the high safety of PIMI nanoparticles in vivo, which lays the foundation of next-generation contrast agent for recognizing macrophage-rich plaque in the near future.
