Interventional effects of atorvastatin on the apoptosis of vascular smooth muscle cells

Abstract:AIM: To investigate the effects of atorvastatin on the apoptosis of vascular smooth muscle cells(VSMCs), and probe into the significance of expression of apoptosis inhibitor Survivin. METHODS: The experiment was performed in the National Key Laboratory for Medical Genetics, central laboratory of Xiangya Second Hospital of Central South University from April 2004 to March 2005. 1 The VSMCs were derived from the middle layers of aortas in 5 one-month-old healthy male rats which had been cultured for 5 or 6 passages. Sterile cover glass was placed in the 6-well culture plate before intervention, and the VSMCs were treated in 4 groups when they grew up to 90% of the whole cover glass: normal control group not treated with atorvastatin, atorvastatin 0.1 μmol/L group treated with 0.1 μmol/L atorvastatin, atorvastatin 1 μmol/L group treated with 1 μmol/L atorvastatin, and atorvastatin 10 μmol/L group treated with 10 μmol/L atorvastatin. 2 The apoptosis of VSMCs was ob-served at 6, 12, 24, 48 and 72 hours after atorvastatin treatment by Hoechst33258 fluorescence staining. The expression of Survivin protein in VSMCs was detected by means of immunohistochemistry. 3 Inter-group difference was performed by t test.RESULTS: 1 From the results of fluorescence staining for the detection of VSMC apoptosis, the apoptotic VSMCs were increased in a dose- and time-dependent manner after they were treated with atorvastatin. At 72 hours af-ter treatment with atorvastatin, the apoptosis rate of VSMCs was signifi-cantly higher in the atorvastatin 0.1 μmol/L, 1 μmol/L, 10 μmol/L groups than in the normal control group(11.1%, 27.2%, 50.5% vs 3.8%, P < 0.01), and it was also significantly different in the inter-group comparison(P < 0.01). 2 The expression of Survivin protein in VSMCs was decreased at 24 hours after treated with atorvastatin, and was not detected in VSMCs at 48 hours to 72 hours after treated with atorvastatin. CONCLUSION: 1 Atorvastatin can induce the apoptosis of VSMCs in dose- and time-dependent manners. 2 The mechanisms of atorvastatin in- ducing apoptosis of VSMCs may be associated with down-regulating expres-sion of Survivin protein.
