Agrin reduces heterogenous expression of nicotinic acetylcholine receptors in skeletal muscles of septic rats

Abstract:Objective To explore the effect of agrin on the heterogenous expression of nicotinic acetylcholine receptors (nAChRs) in the skeletal muscles of rats with sepsis. Methods Forty-eight 2-month-old healthy SD rats were randomized equally into sham-operated group, sham+agrin group (ShamA group), cecal ligation and puncture (CLP) group, and CLP+agrin group (CLPA group) and received either CLP to establish models of sepsis or a sham operation of laparotomy without CLP as indicated. In ShamA group and CLPA group, the rats were given an intramuscular injection of 0.1 mL agrin (100 μg/mL) into the anterior tibial muscle after the operation. At 48 h after the modeling, blood samples were collected from the rats for measurement of plasma interleukin-6 (IL-6) level, and the compound muscle action potentials (CMAP) were detected to assess the neuromuscular conduction. The protein expression levels of type γ-nAChR and α7-nAChR in the anterior tibial muscle were detected using Western blotting and immunofluorescence staining. Results The plasma IL-6 levels were significantly lower in the sham-operated and ShamA group than in CLP and CLPA groups (P < 0.05) and were comparable between the latter 2 groups (P>0.05). Compared with those in the 2 sham-operated groups, the duration and latency of CMAP were increased significantly while the amplitude and nerve conduction velocity were decreased obviously in CLP group (P < 0.05). Compared with those in CLP group, the duration and latency of CMAP were decreased and the amplitude and nerve conduction velocity were increased significantly in CLPA group (P < 0.05). In the anterior tibial muscle, the protein expression levels of γ-nAChR and α7-nAChR were significantly higher in CLP and CLPA groups than in the 2 sham-operated groups (P < 0.05), and their levels were the highest in CLP group (P < 0.05). Conclusion Agrin can improve neuromuscular function in rats with sepsis, which is likely attributed to down-regulated heterogeneous expression of γ-nAChR and α7-nAChR in the skeletal muscles.
