Establishment and CT Evaluation of Rabbit VX2 Hepatic Tumor Model

Abstract:Objective To monitor rabbit VX 2 hepatic tumor and to analyze its imaging features with spiral computed tomography (SCT). Methods Hepatic tumors were created by injecting fragments of VX 2 tumor into the left lobe of the liver. Plain CT scan and contrast-enhanced CT scan were performed at 7, 14, 21, 28, and 35 d after implantation, respectively. The biological and histopathological features and the spiral CT features of tumors were observed. Results The success rate of the transplanted tumor was 100%. Three lesions were detected by CT at 7 d after implantation. VX 2 tumor grew slowly at 14 and 21 d after implantation, but grew quickly at 28 and 35 d after implantation. The tumor was fitted well to an exponential curve and metastases were found at 28 d after implantation. Histopathological examination showed that tumor grew in the liver tissue and infiltrated into the margins of the liver and had the similar biological features with squamous cell carcinoma. The tumor appeared as a low-attenuation solitary lesion in unenhanced CT except 2 lesions (10%) to be homogeneous density during the period from 14 d to 21 d. In arterial phase, significant peripheral enhancement was observed. In 17 lesions (85%), the dilated nutritional arteries stretching straight to the margins of 15 tumors (75%) were detected while the main bodies of the tumors showed hypodensity. In portal and equilibrium phase, concentrical enhancement of tumors was detected. The density of the tumors became heterogeneous on plain CT scan at 21 d after implantation. Conclusion SCT is a quick, non-invasive, and repeatable method for the evaluation of the properties and the therapeutic effect of metastatic rabbit VX 2 liver tumor model.
