Cartilage Regeneration from Epiphyseal Chondrocytes Cultured in Biological Gel in Vitro

Abstract:Objective To observe the biological characteristics of the cartilage regenerated from epiphyseal chondrocytes implanted into a self-made biological gel during in vitro cultivation, and evaluate the biological property of the gel as the matrix material of cartilage tissue engineering. Methods The first passage chondrocytes were harvested and implanted into the gel for culture in vitro. The changes of chondrocyte-gel complex in morphology, the syntheses of collagen type Ⅱ and Ⅰ, and aggrecan were investigated grossly and with inverted microscope and immunochemical methods. Results The chondrocyte-gel comlplex contracted to 45% of its original volume and lost its primary shape in the first week of cultivation, but maintained the even and uniform distribution of cells. The complex gradually changed from periphery to center and finally became mature cartilage after 2 weeks of cultivation. At this time, the complex was covered on surface with a membrane-like structure consisting of 1-3 layers of spindle cells, and inside, round cells were separated by extracellular matrix with the formation of chondrocytic lacunas. The complex showed strong positive staining of collagen type Ⅱ and aggrecan in the matrices and light positive staining of collagen type Ⅰ mainly in peripheral membrane. Conclusion The biological gel possesses satisfying cellular compatibility, and is fit for chondrocytes growing in it to regenerate engineered cartilage, but its primary shape is not well-maintained.
