Expression of Cripto-1 in hepatocellular carcinoma

Abstract:Objective To evaluate expression of Cripto-1 protein in hepatocellular carcinoma (HCC),nontumor-ous liver tissues,and HCC cell lines with various metastatic potentials,and determine the correlation between the expression level and HCC metastatic potential.Methods Western Blot was used to evaluate Cripto-1 protein ex-pression in HCC tumor tissues,nontumorous liver tissues and HCC cell lines with various metastatic potentials.Im-munohistochemistry was used to evaluate Cripto-1 expression in HCC tumor tissues,and determine the correlation between the expression level and clinicopathologiacal parameters.Results Expression level of Cripto-1 protein was higher in HCC tissues than their paired adjacent nontumorous liver tissues as well as normal liver tissues.Cripto-1 protein expression was positively correlated with metastatic potential of HCC cell lines.High Cripto-1 expression was detected in 64.6% (53 /82)of HCC tumor tissues,and 0 (0 /10)in the normal liver tissues.Clinicopatholog-ical analysis on 82 HCC patients demonstrated that Cripto-1 expression was significantly associated with tumor dif-ferentiation,vascular invasion and status of metastasis (P <0.05).Conclusion The expression level of Cripto-1 protein is elevated in HCC,and positively correlated with the invasion and metastasis of HCC.
