Genistein Suppresses in Vitro Invasive Growth of Human Hepatocellular Carcinoma Cells

Abstract:Objective To study in vitro inhibitory effects of genistein on invasive growth of Bel 7402 hepatocellular carcinoma cells, and to explore the underlying mechanism. Methods Bel 7402 hepatocellular carcinoma cells were exposed to genistein with 5 μg/mL and 10 μg/mL, respectively. Cell number and viability were evaluated every 24 h by trypan blue dye exclusion; cell attachment assay was carried out in a microculture well pre-coated with fi-bronectin (FN) ; the invasive activity of tumor cells was assayed in a Transwell cell culture chamber; FAK expression and cell cycle were evaluated by functional assay ( FCM ) ; cell apoptosis analysis was performed with TUNEL method. Results Genistein significantly inhibited the growth of Bel 7402 cells, and the inhibitory rate of tumor cells in the G5 and G10 group was about 26. 71% and 42. 64% , respectively. The attachment of Bel 7402 cells to FN pre-coated plastic wells was significantly inhibited by genistein, and the inhibitory rate of the attachment within 40 min in the G5 and G10 group was about 43. 52% and 84. 18% , respectively. A lower rate of invasion in vitro detected in the Transwell cell culture chamber was also observed in the group treated with genistein, and the inhibitory rate of invasion was about 28% in the G10 group. Besides, genistein caused cell arrest in G2/M transition and S phase was decreased significantly. The occurrence of apoptosis detected by the TUNEL method in the group treated with genistein was increased significantly. The expression of FAK in the group treated with genistein 10 μg/mL (12.89 × 0.36)% was significantly lower than that in the control group (19. 75 ×1. 12)% (P 0. 05). Conclusion Genistein significantly inhibits the in vitro invasive growth of Bel 7402 hepatocellular carcinoma cells. These inhibitory effects of genistein on Bel 7402 cells are closely related to the alteration of FAK expression.
