Effect of Curcumin on Rat Hepatic Fibrosis Induced by Carbon Tetrachoride

Abstract:Aim:To investigate the effect of curcumin(Cur) on rat liver fibrosis induced by carbon tetrachoride (CCl4) and its possible molecular mechanism.Methods:A total of 64 male rats were randomly divided into six groups: normal control group(n=10),model group(n=14),silibinin group(n=10) and low,middle,and high-dose curcumin groups(n=10).Apart from normal control group,those in other five groups were injected subcutaneously 40% CCl4 for seven weeks to induce the model of liver fibrosis.At the same time,the rats in silibinin groups were intragastrically administered with 0.5 g/L silibinin,the rats in curcumin groups were intragastrically administered with 1,2,4 g/L curcumin once per day,respectively. The serum levels of ALT,AST,ALP,TP,albumin were measured by enzymatic assays.The levels of hyaluronic acid(HA),laminin(LN),procollagenⅢ N-terminal peptide(PⅢNP),and collagen Ⅳ in serum were measured using radioimmunity technique. Liver samples were stained with HE,and was used to observe the pathological changes of liver tissues under light microscope. Immunohistochemistry were performed to detect the activity of collagenⅠ and peroxisome proliferator-activated receptor γ (PPAR-γ).Results: Administration of curcumin significantly decreased the serum transaminase activities,and the levels of HA,LN,PⅢ NP and C-Ⅳ,while increased the ratio of albumin to globulin(F=11.535,19.927,16.415,17.596,19.692,21.535,4.831,P0.05).Histopathological scores showed curcumin had inhibitory effect on the progression of hepatic fibrosis. CCl4-administered rats depicted siginificant increases in collagenⅠ production while curcumin remarkably suppressed expression levels of collagenⅠand promoted the expression levels of PPAR-γ(F=7.541,6.519,P0.05).Conclusion:Curcumin effectively reverses CCl4-induced hepatic fibrosis by means of activating specific nuclear receptor PPAR-γ and inhibiting the production of collagenⅠ.
