Role of Atorvastatin in Stabling Artery Plaque and Reducing Thrombotic Events in Rabbits

Abstract:Objective To establish rabbit models of atherosclerotic vulnerable plaques and evaluate the effect of atorvastatin on plaques stability and thrombosis.Methods Atherosclerotic vulnerable plaques were induced with high-fat feed and balloon injury in 18 male rabbits.All rabbits were randomly divided into high-fat group(n=9) and atorvastatin group(n=9),and were given high-fat diet,meanwhile atorvastatin was given to atorvastatin group.After pharmacological triggering,all survival rabbits were sacrificed to obtain the aorta and processed HE staining and immunohistochemical staining.Results The percentages of each model of atherosclerosis plaque were 5.29% of type Ⅱ,13.94% of type Ⅲ,26.44% of type Ⅳ,25.00% of type Ⅴ,and 29.33% of type Ⅵ in high-fat group.Vulnerable plaques had large plaque area,thin fibrous cap,small lipid core ratio,and more infiltrating inflammatory cells.Compared with high-fat group,atorvastatin group had fewer vulnerable plaques(29.33% vs 13.08%),and more stable plaques(70.67% vs 86.92%)(P0.05).The percentage of plaque area in total lumen area and plaque thickness were much smaller in atorvastatin group((23.48±9.94)%,(311.27±190.98) μm) than those in high-fat group((50.23±10.08)%,(737.47±248.73) μm)(P0.05).The thrombosis incidence was lower in atorvastatin group(33%) than that in high-fat group(75%)(P0.05).Conclusion Atorvastatin can improve plaque stability and reduce plaque rupture and thrombosis.
