Activated macrophages for regeneration of spinal function after spinal injuries in rats: the opportunity and the effect

Abstract:Objective: To investigate the correlation between the opportunity of activated macrophages transplantation and the restoration after the spinal cord functions of the spinal cord injury (SCI).Methods: Sixty-two semi-resected BALB C rats whith SCI were grouped in A(n=21),B(n=21) and C(n=20) ,randomly . When the spinal cord had been semi-resected, the activated macrophages were injected into the gap of the spinal cord in group A after one hour and in group B after eight hours. 8 rats were taken respectively in A,B and C group randomly, they were observed the restoration of the motor functions by the Basso, Beattie, and Bresnahan (BBB) loco motor rating scale and the improvement of the somatosensory evoked potential(SEP)following every week after injury. The other 8 rats in A, B, and C group randomly were killed 1 day, 4 days, 1 week, 2,4, 8,12 and 16 weeks, then spinal cords were fixed and cut into transverse frozen sections, and were immunostained and stained with hematoxylin and eosin (HE) .The numbers of cells or neurofilaments were counted under microscope. Results: There was a significant difference of BBB scales and SEP between group A and B, C (ANOVA,P0.05). It was proved that the motor functional restoration was obviously. The axons of group A increased quickly. Conclusion: Suitable opportunity is essential for the activated macrophages transplantation to promote the recovery of the spinal cord functions.
