Association of TNF-αG-308A gene polymorphism with pathogenesis of type 2 diabetic nephropathy

Abstract:Objective To investigate the association of TNF-αG-308A gene polymorphism with type 2 diabetic nephropathy in Han population. Methods From March 2014 to Jan,2015,113 cases of type 2 diabetes were randomly selected in our hospital,and diagnosed with a DN as case group(DN group);108 cases of healthy individuals were selected as control group. Use polymerase chain reaction restriction fragment length polymorphism(PCR-RFLP)to detect the three genotypes of GG,GA,AA of TNF-αG-308A,and then correlation analysis of samples TNF-αG-308A gene polymorphism and type 2 diabetic nephropathy was carried out. At the same time,the individual indicators were detected,including body mass index (BMI),systolic blood pressure(SBP),diastolic blood pressure(DBP),total cholesterol(TC),triglycerides(TG), HbAlc, blood urea nitrogen(BUN)and 24 hour urine protein content and other clinical and biochemical indexes. Results The sample distribution between DN group and normal control group conformed to the law of Hardy-Weinberg equilibrium (P>0.05), which represent the selected sample possess representative characteristics. The clinical parameters of type 2 diabetic nephropathy that indicated by clinical trials index analysis,which include BMI,SBP,BUN,TG,24 h urine protein,were significantly higher than those of the control group (P<0.05). The GG genotype in DN group was significantly higher than that of the control group (χ2=6.51,P<0.05). The frequency distribution of G allele in DN group and normal control was 56.8% and 38.1%,respectively. The data of DN group was significantly higher than the control group(χ2=3.32,P<0.05). Logistic regression analysis data showed that TNF-αG-308A polymorphism and 24 h urine protein levels were risk factors for the development and progression of DN. Conclusion The TNF-α G-308A gene polymorphisms was closely related with type 2 diabetic nephropathy and development,and the G allele might be a genetic susceptibility factor for type 2 diabetic nephropathy.
