Abstract:DNA vaccine is a new vaccine after live attenuated,whole killed,subunit and recombinant one,etc.It’s better than them at efficiency,safety and ease to produce.The article is mainly discussed construction,effect mechanism,immunization methods,efficiency and safety of DNA vaccine used to fish.It is expected to boost the research and application of DNA vaccines in fish,which is due to expedite the development of aquaculture in our country,and to provide some evidence for studying deeply and grasping the mechanism of fish immunoprotection.

Abstract:The research on DNA vaccines has opened up a new model in the prevention and treatment of clinical diseases.TANK-binding kinase-1(TBK-1),a key signaling molecule in innate immunity,can not only act importantly in the innate immune response which is mediated by TLR or RIG-Ⅰ,but also promote the adaptive immune response.Recent findings show that TBK-1 plays an important role in the immune response elicited by DNA vaccines.In this article,some progress on the research are reviewed.

Abstract:As a novel vaccine set up in early 1990s,DNA vaccine has been developed rapidly in recent years and played an important role in the prevention and treatment of viral diseases and tumor. Compared with the traditional vaccines,it had some advantages,such as good immune effect,low production cost,and convenient usage for the clinical application,but it could also be found safety concerns. To provide the references for DNA vaccine researchers,the development,mechanism,and advantage are reviewed in this paper,and my personal viewpoint about its safety is presented as well.

Abstract:Adenoviruses represent the most widely used viral-vectored platform for vaccine design, showing a great potential in the fight against intracellular infectious diseases to which either there is a lack of effective vaccines or the traditional vaccination strategy is suboptimal. The extensive understanding of the molecular biology of adenoviruses has made the new technologies and reagents available to efficient generation of adenoviral-vectored vaccines for both preclinical and clinical evaluation. The novel adenoviral vectors including nonhuman adenoviral vectors have emerged to be the further improved vectors for vaccine design. In this review, we discuss the latest adenoviral technologies and their utilization in vaccine development. We particularly focus on the application of adenoviral-vectored vaccines in mucosal immunization strategies against mucosal pathogens including Mycobacterium tuberculosis, flu virus, and human immunodeficiency virus.

Abstract:The concept of DNA vaccination was introduced in the early 1990s. Since then, advancements in the augmentation of the immunogenicity of DNA vaccines have brought this technology to the market, especially in veterinary medicine, to prevent many diseases. Along with the successful COVID mRNA vaccines, the first DNA vaccine for human use, the Indian ZyCovD vaccine against SARS-CoV-2, was approved in 2021. In the current review, we first give an overview of the DNA vaccine focusing on the science, including adjuvants and delivery methods. We then cover some of the emerging science in the field of DNA vaccines, notably efforts to optimize delivery systems, better engineer delivery apparatuses, identify optimal delivery sites, personalize cancer immunotherapy through DNA vaccination, enhance adjuvant science through gene adjuvants, enhance off-target and heritable immunity through epigenetic modification, and predict epitopes with bioinformatic approaches. We also discuss the major limitations of DNA vaccines and we aim to address many theoretical concerns.

Abstract:Reverse-genetic engineering of foot and mouth disease virus (FMDV) can improve the productivity, antigen matching, antigen stability, immune response ability, and biological safety of vaccines, so vaccine candidates with anticipated biological characteristics can be promptly achieved. Negative influence in taming of virulent strains can also be decreased or avoided. Reverse genetics not only make up for deficiencies like limitation of viral nature, low success rate, and time and energy consuming, but also realize more active designing of vaccines. Therefore, reverse genetics is significant in improving integral quality and efficiency of vaccines. In this review, we use FMDV vaccines as an example to summarize improvement in biological characteristics of virulent strains and provide a reference for related researches.

Abstract:Vaccines have proven effective in the treatment and prevention of numerous diseases. However, traditional attenuated and inactivated vaccines suffer from certain drawbacks such as complex preparation, limited efficacy, potential risks and others. These limitations restrict their widespread use, especially in the face of an increasingly diverse range of diseases. With the ongoing advancements in genetic engineering vaccines, DNA vaccines have emerged as a highly promising approach in the treatment of both genetic diseases and acquired diseases. While several DNA vaccines have demonstrated substantial success in animal models of diseases, certain challenges need to be addressed before application in human subjects. The primary obstacle lies in the absence of an optimal delivery system, which significantly hampers the immunogenicity of DNA vaccines. We conduct a comprehensive analysis of the current status and limitations of DNA vaccines by focusing on both viral and non-viral DNA delivery systems, as they play crucial roles in the exploration of novel DNA vaccines. We provide an evaluation of their strengths and weaknesses based on our critical assessment. Additionally, the review summarizes the most recent advancements and breakthroughs in pre-clinical and clinical studies, highlighting the need for further clinical trials in this rapidly evolving field.

Abstract:Vaccines play a critical role in the prevention of life-threatening infectious disease. However, the development of effective vaccines against many immune-evading pathogens such as HIV has proven challenging, and existing vaccines against some diseases such as tuberculosis and malaria have limited efficacy. The historically slow rate of vaccine development and limited pan-variant immune responses also limit existing vaccine utility against rapidly emerging and mutating pathogens such as influenza and SARS-CoV-2. Additionally, reactogenic effects can contribute to vaccine hesitancy, further undermining the ability of vaccination campaigns to generate herd immunity. These limitations are fuelling the development of novel vaccine technologies to more effectively combat infectious diseases. Towards this end, advances in vaccine delivery systems, adjuvants, antigens and other technologies are paving the way for the next generation of vaccines. This Review focuses on recent advances in synthetic vaccine systems and their associated challenges, highlighting innovation in the field of nano- and nucleic acid-based vaccines.

Abstract:The escalating global changes have fostered conditions for the expansion and transmission of diverse biological factors, leading to the rise of emerging and reemerging infectious diseases. Complex viral infections, such as COVID-19, influenza, HIV, and Ebola, continue to surface, necessitating the development of effective vaccine technologies. This review article highlights recent advancements in molecular biology, virology, and genomics that have propelled the design and development of innovative molecular tools. These tools have promoted new vaccine research platforms and directly improved vaccine efficacy. The review summarizes the cutting-edge molecular engineering tools used in creating novel vaccines and explores the rapidly expanding molecular tools landscape and potential directions for future vaccine development. The strategic application of advanced molecular engineering tools can address conventional vaccine limitations, enhance the overall efficacy of vaccine products, promote diversification in vaccine platforms, and form the foundation for future vaccine development. Prioritizing safety considerations of these novel molecular tools during vaccine development is crucial.

Abstract:Human diseases, particularly infectious diseases and cancers, pose unprecedented challenges to public health security and the global economy. The development and distribution of novel prophylactic and therapeutic vaccines are the prioritized countermeasures of human disease. Among all vaccine platforms, viral vector vaccines offer distinguished advantages and represent prominent choices for pathogens that have hampered control efforts based on conventional vaccine approaches. Currently, viral vector vaccines remain one of the best strategies for induction of robust humoral and cellular immunity against human diseases. Numerous viruses of different families and origins, including vesicular stomatitis virus, rabies virus, parainfluenza virus, measles virus, Newcastle disease virus, influenza virus, adenovirus and poxvirus, are deemed to be prominent viral vectors that differ in structural characteristics, design strategy, antigen presentation capability, immunogenicity and protective efficacy. This review summarized the overall profile of the design strategies, progress in advance and steps taken to address barriers to the deployment of these viral vector vaccines, simultaneously highlighting their potential for mucosal delivery, therapeutic application in cancer as well as other key aspects concerning the rational application of these viral vector vaccines. Appropriate and accurate technological advances in viral vector vaccines would consolidate their position as a leading approach to accelerate breakthroughs in novel vaccines and facilitate a rapid response to public health emergencies.

Abstract:In recent years, the CRISPR/Cas9-based gene-editing techniques have been well developed and applied widely in several aspects of research in the biological sciences, in many species, including humans, animals, plants, and even in viruses. Modification of the viral genome is crucial for revealing gene function, virus pathogenesis, gene therapy, genetic engineering, and vaccine development. Herein, we have provided a brief review of the different technologies for the modification of the viral genomes. Particularly, we have focused on the recently developed CRISPR/Cas9-based gene-editing system, detailing its origin, functional principles, and touching on its latest achievements in virology research and applications in vaccine development, especially in large DNA viruses of humans and animals. Future prospects of CRISPR/Cas9-based gene-editing technology in virology research, including the potential shortcomings, are also discussed.

Abstract:The design of DNA vaccines and their mode of application need to be thoroughly improved to mount strong and sustained immune responses in humans, for example, by deleting prokaryotic sequences known to exert detrimental effects from the vector backbone, or by nanoformulated delivery that allows cell type‐specific transfection and codelivery of adjuvants. Despite the great potential of DNA vaccines for a broad range of applications, ranging from prevention of infections, over treatment of autoimmune and allergic diseases to cancer immunotherapies, the implementation of such therapies for clinical treatment is far behind the expectations up to now. The main reason is the poor immunogenicity of DNA vaccines in humans. Consequently, the improvement of the performance of DNA vaccines in vivo is required. This mini‐review provides an overview of the current state of DNA vaccines and the various strategies to enhance the immunogenic potential of DNA vaccines, including (i) the optimization of the DNA construct itself regarding size, nuclear transfer and transcriptional regulation; (ii) the use of appropriate adjuvants; and (iii) improved delivery, for example, by careful choice of the administration route, physical methods such as electroporation and nanomaterials that may allow cell type‐specific targeting. Moreover, combining nanoformulated DNA vaccines with other immunotherapies and prime‐boost strategies may help to enhance success of treatment.

Abstract:This article mainly discusses the prospects of DNA vaccines as a future form of immunization. DNA vaccines, also known as nucleic acid vaccines or gene vaccines, inject genes encoding specific protein antigens directly into animals. By utilizing recombinant eukaryotic expression vectors, they activate the immune system to produce specific humoral and cellular immune responses, providing comprehensive protection against specific pathogens. Compared with traditional vaccines, DNA vaccines have shown new promise in addressing many viral infections. Although no DNA vaccines have been approved for use in humans at present, research on DNA vaccines for specific human diseases, such as prostate cancer, is ongoing. Additionally, the delivery methods of DNA vaccines, including oral vaccines and particle-mediated epidermal vaccination, as well as the challenges and directions for improvement in their clinical application, are explored. Finally, despite these challenges, the research and application prospects of DNA vaccines are broad.

Abstract:COVID 19 Pandemic is caused by the viral pathogen severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Scientific fraternity worldwide swiftly developed various types of vaccines for the prevention and as mitigation measures for curbing the pandemic. Traditional inactivated vaccines, mRNA vaccines (protein subunits such as spike proteins), and viral vector vaccines (non-replicating vectors with protein subunits) have been approved by World Health Organisation (WHO) for emergency use. The emergence of many mutated variants has been a worrying factor in the fight against the pandemic. There has been continuous research in the quest for more therapeutics, especially vaccines to curb and stop the pandemic. According to WHO, there are 194 vaccines in pre-clinical trials belonging to various types out of which sixteen is DNA vaccines. In this review, we have discussed the advantages and disadvantages of the DNA vaccines for Covid - 19. This article tried to explore the available information on DNA vaccines and their current status against Covid – 19 which are in pre-clinical trials.
