P53 Overexpression and Mutation in Endometrial Carcinoma

Abstract:：Objective To investigate p53 gene mutations in endometrial carcinoma（EC）,and their correlation with clinical features and prognosis. Methods Using immunohistochemical staining（IMH）and polymerase chain reaction-single strand conformation polymorphism（PCR-SSCP）methods, examine the protein expression and gene mutation of p53 in EC. Results The general positive rate of P53 protein was 34.15%. The expression of p53 was not correlated with age，clinical stage，myometrial invasion ，lymph node or ovarian metastasis（Ｐ0.05），but the pathologic grade（Ｐ0.05）.Survival curves determined by Kaplan-Meier method showed that p53 overexpression was associated with a poor prognosis in EC（Ｐ0.05）.The positive rates of p53 gene mutation was 23.08% in EC, and the mutation were located on exon 5，6，and 7.The concordance rate of IMH and PCR-SSCP was 76.92%. Conclusion The mutation of p53 gene were associated with the pathologic grade and poor prognosis in EC.
