Effects of Antioxidant N-Acetylcysteine on Caveolin-3/eNOS/NO Signaling Pathway in Early Diabetic Rats

Abstract:Objective:To investigate the effects of N-acetylcysteine (NAC) on the myocardial function and caveolin-3/eNOS/NO signaling in early diabetic rat hearts.Methods:Pathogen-free male Sprague-Dawley rats,weighing (260±10) g,were used in this study.Twenty-four rats were randomly allocated into three groups (n =8/group) by using a random number table:control group (group NC),diabetic group (group DM) and diabetes with NAC treatment group (group DM+NAC).Diabetes was induced by tail vein injection of streptozotocin at the dose of 60 mg/kg.One week after induction of diabetes,the rats of DM+ NAC group were treated by oral gavage with NAC for 4 weeks at the dose of 1.5 g/(kg · d),and NC group and DM group were given equal volume of saline for four weeks.Upon completion of treatment,cardiac function were assessed by echocardiography.The levels of free 15-F2t-isoprostane,total antioxidants capacity and SOD activity in serum,as well as myocardial 15-F2t-isoprostane,O2-·,Nitrotyrosine,and NO were detected by commercial kits.Myocardial Caveolin-3,p-eNOS and eNOS expression were assessed by Western Blotting.Results:Compared with NC group,myocardial diastolic function in DM group was significantly decreased,but myocardial systolic function was not affected;the levels of 15-F2t-isoprostane and the total antioxidants capacity in serum were significantly increased.While,the levels of SOD and NO were decreased.15-F2t-isoprostane,O2-· and Nitrotyrosine levels in myocardial tissue were significantly increased,and cardiac Caveolin-3 and p-eNOS protein expression were significantly attenuated (P<0.05).Compared with DM group,NAC treatment could improve myocardial diastolic function,decrease the levels of 15-F2t-isoprostane,total antioxidant capacity,O2-· and Nitrotyrosine levels,increase SOD,NO activity and up-regulate cardiac expression of Caveolin-3 and p-eNOS (P<0.05).Conclusion:The expression of Caveolin-3 and p-eNOS may play an important role in the regulation of oxidative stress and cardiac function in early diabetic rats.N-acetylcysteine inhibits oxidative stress and improves diabetic heart function by improving the expression of Caveolin-3 and p-eNOS.
