The Role of Apoptosis in Lung Injury Induced by Brain Death

Abstract:Objective:To study on changes of cell apoptosis and related genes in pulmonary tissues at different time points after brain death of rabbits,and to research the mechanism of lung injury induced by brain death.Methods:Forty rabbits were separated into two groups averagely,shamoperation group and brain death group.Other treatments in the two groups are the same except that the sham-operation don't use high intracranial pressure to establish a state of brain death.We recorded mean arterial pressure and heart rate of the two groups in 2,6,8 hours after operation,observed the cell apoptosis with TUNEL,and detected the expression of apoptosis genes including Bax and caspase 3 with RT-PCR and immunohistochemistry.Results:There were no significant differences in mean arterial pressure and heart rate at 2,6 and 8 h after brain death.The level of caspase 3 in brain death group was significantly higher than that in sham-operated group.Apoptotic index increased according to time,and the differences are statistically significant,which was consistent with those of the mRNA expression of Bax in brain death group.Conclusion:Brain death can lead to the lung damage,which gets worse over time,and the damage is related to over-expression of apoptosis genes.The lung apoptosis peaks at 8 h after brain death.
