Role of the interaction between Caveolin-1 and protease-activated receptor 1 in regulating brain microvascular endothelial cell barrier after oxygen glucose deprivation

Abstract:Objective: To explore the effect of Caveolin-1(Cav-1) and its interaction with protease-activated receptor 1 (PAR-1) on the endothelial cell tight junction protein by establishing an oxygen-glucose deprivation(OGD) model in vitro. Method:Mouse brain microvascular endothelial cells(bEND3) cultured in vitro, after 2 hours of OGD. The expression of Cav-1 and occludin were determined by Western Blot. Interactions of Cav-1 and PAR-1 were detected by co-immunoprecipitation. The co-localization of Cav-1 and endothelial protein C receptor(EPCR) was detected by immunofluorescence assay. Results:2 hours of OGD lead to occludin degradation in bEND3 but not Cav-1 expression variation. Interactions between Cav-1 and PAR-1 in bEND3 decreased. The co-localization of Cav-1 and EPCR increased. Conclusion: After OGD, the dissociation of Cav-1 and EPCR in endothelial cells, and the enhanced interaction of Cav-1 and PAR-1, aggravating endothelial cell barrier injury.
