Abstract:Cellobiase is a hydrolytic enzyme which acts on various compounds with #beta#-D-glucosidic linkages. It plays a key role in hydrolyzing cellobiose to glucose. Cellobiase bglII gene was cloned by RT-PCR using first strand cDNA as template after reverse transcripting T. reesei total RNA. After sequenced, the bglII cDNA was then inserted directionally into a temperature-inducible prokaryotic expression vector pJW2. The bglII gene was expressed in E. coli DH5#alpha#.

Abstract:AIM: To clone human sCD40L(184-831) cDNA and construct a prokaryotic vector for expression in E. coli. METHODS: The human sCD40L(184-831 ) cDNA was amplified with the total RNA from the human tonsil by reverse transcriptase (RT)-PCR, and was analyzed by enzymatic digestion and sequencing. Then, the insert of human sCD40L(184-831) cDNA was cloned into the vector pET-28a+ for expression in BL21, and then the interest protein was identified by Western blot. RESULTS: The result of sequencing was accordant with the one reported. The prokaryotic expression vector was constructed successfully and the interest protein was expressed in BL21. CONCLUSION: The human sCD40L(184-831) cDNA has been successfully cloned and a prokaryotic system been constructed for expression. The interest protein was expressed successfully in BL21.
High expression of human Era and human Era C-terminal domain in E.coli

Abstract:AIM　To highly express human Era (h-Era)　and h-Era C-terminal domain protein in E.coli. METHODS　Human era(h-era) cDNA and h-era cDNA C-terminal domain gene were amplified by PCR by using a plasmid containing h-era cDNA as a template, and ligated with prokaryotic expression vector pRSET-C and pDH respectively. E.coli BL21(DE3) transformed with the recombinant plasmid pRSET-C-h-era was induced with IPTG to express (His)6-h-Era fusion protein. E.coli TAP106 transformed with the recombinant plasmid pDH-h-era-C was induced at 42℃ to express h-Era C-terminal domain protein. The expression products were identified by SDS-PAGE and gel thin-layer chromatography-scanning. RESULTS　the expressed (His)6-h-Era fusion protein was about 80% of total bacterial protein. The expressed h-Era C-terminal domain protein was about 40% of total bacterial protein. CONCLUSION　(His)6-h-Era fusion protein and h-Era C-terminal domain protein had been successfully highly expressed in E.coli.
Cloning of Human CDH22 Gene and Construction of Its Eukaryotic Expression Vectors

Abstract:Objective To obtain entirely coding human CDH22 gene and construct its eukaryotic expression vectors.Methods With total RNA extracted from the human colorectal carcinoma cell sw480 as template,CDH22 gene was amplified by RT-PCR with the designed primers based on the public sequence of GeneBank,and then was inserted into pGEM-T Easy vector.The recombinant plasmid pGEM-T-CDH22 was identified by Restriction endonuclease analysis and DNA sequencing.Digested by restriction endonuclease,the CDH22 was cloned into multiclone sites of the eukaryotic expression vector pCDNA3/HA.The expression of CDH22 gene was detected by Western Blot and immunofluorescence cytochemistry using mouse anti-HA antibody after the recombinant plasmid pCDNA3/HA-CDH22 was transfected into HEK293A cells.Results The entirely coding human CDH22 gene was cloned.Recombinant pCDNA3/HA-CDH22 were successfully constructed and expressed.Conclusion The successfully constructed pCDNA3/HA-CDH22 would provide a basis for a further study of the relationship between the tumorigenesis and CDH22 gene.

Abstract:According to the mRNA sequence of human Lysozyme(hLYZ) derived from Genbank,a pair of primers was designed.Human lysozyme(hLYZ) cDNA was amplified by RT-PCR from total RNA of human placenta which was 0.85 kb.The cloned fragment was sequencd and compared with hLYZ cDNA which was published,the result indicated that the sequence homology was 99%.Sequence analysis with blastp indicated that the deduced amino acid sequence has the homology of 97% and 99% respectively with the precursor protein of hLYZ and mature peptide.Which is the vital basis for the further construction of mammary gland-specific expression vector.

Abstract:The total RNA of HepG2 cell was extracted as the template by ultrocentrifuge method. The full length cDNA (0.8 kb) encoding the human stem cell factor (hSCF) was amplified by RT-PCR method. The cDNA encoding mature hSCF (0.5 kb) was sequenced and was recombined into the expression vector (PBV-220). The expression level of rhSCF in E. coli DH5 alpha was about 15% of the total protein.

Abstract:Copper is one of the most important trace elements to life. Human HCUTA is a novel cDNA encoding a 156aa protein, which may participate in human copper tolerance system. The HCUTA protein is highly similar to protein CUT A1 of E. coli . The whole opening reading frame of HCUTA cDNA was amplified by PCR and cloned into pET28a + express vector, and the HCUTA protein was effectively expressed in E. coli BL21 (DE3).

Abstract:A novel cDNA, HGLP, which encodes a G-protein coupled receptor (GPCR) like protein, has been isolated and cloned. The coding region of the human HGLP predicts a 7-transmembrane region protein with motifs of rhodopsin-like GPCR superfamily. Northern blot analysis reveals a 3-kb transcript in various human tissues examined. The N- and C-terminal coding regions of HGLP, which are deduced as non-transmembrane regions, have been amplified by PCR and cloned into pET30a(+) vector. Then the recombinant proteins are highly expressed in E. coli.

Abstract:The ubiquitin-proteasome system is an essential mechanism for protein degradation in eukaryotes. Protein ubiquitination is composed of a series of enzymatic reactions. The ubiquitin-conjugating enzyme (E2) is one of the important enzymes involved in the process. A cDNA encoding an E2 enzyme was cloned from a Clonorchis sinensis cDNA library by large-scale sequencing. This new cDNA contains 862 bp with a putative open reading frame of 156 amino acids. The deduced amino acid sequence is 77% identical to the human E2, HHR6A and HHR6B. The coding region of this cDNA was expressed in E. coli as a GST-tagged protein, and was purified to electrophoretic homogeneity. Enzymatic assays showed that this E2 had the capacity to form a thiolester linkage, and could conjugate ubiquitin to histone H2A in an E3-independent manner in vitro, which indicated that the expressed protein was functionally active. The nucleotide sequence reported in this paper has been submitted to the Genbank Database with accession number AY632078.

Abstract:Objective To clone and partially sequence of human cartilage－ derived protein－ 1(CDMP－ 1 ) encoding mature protein. Methods In the study, total RNA was extracted respectivelyby the improved acid guanidinium thiocyanata－ phenol－ chloroform method from the human cartilagetissue, and chondrocytes from the human cartilage through collagenase digestion. The desired DNAproduct was obtained from the total RNA by RT－ PCR with the primers including Oligo(dt) and twogene specific primers. The segment (about 1.0 kbp) was inserted into the pBluescript vector and the in-teresting plasmid was transformed into the E. coli host strain DHSa. The double － stranded DNA of thepositive clone was analyzed by restriction endonuclease mapping and DNA sequencing. Results Therestriction endonuclease map and sequence of human CDMP－ 1 encoding mature protein were consistentwith those of the references published. Conclusion The CDMP－ 1 cDNA encoding mature proteinmay be obtained for further research.

Abstract:Objective To clone and express the 582 bp cDNA fragment of the extracellular domain of human CD40.Methods The cDNA of human CD40 antigen was amplified from total RNA of XG-2 cells by RT-PCR,and it was inserted into pMD18-T.The extracelluar domain of human CD40 was amplified from the recombinant vector pMD18-T/CD40,then it was subcloned into expression vector pGEX-5x-3,E.coli BL21(DE3) was transfected with the recombinant vector and the expression was induced with IPTG.Results DNA sequencing showed that the obtained 582 bp cDNA sequence was identical with the reported CD40 gene.The target fragment was expressed in bacterial cells.Conclusion The expressed product of 582 bp cDNA fragment of human CD40 obtained lays the foundation for studing the functions of human CD40 antigen.

Abstract:Objective:In order to obtain the DNA sequence encoding human interferon-γand fitting for exˉpression in E.coli.,the codons were all changed into favorite for bacteria.Methods:The human interferon-γgene coding sequence of DNA modified according to the known E.coli favorite codon was divided into six fragˉments and synthesized them chemically.The full length DNA was connected by serious PCR.experiments.The final product of PCR was cloned into plasmid pGEM-T and the recombinant plasmid was sequenced.Results:It was proved by sequence analysis the recombinant plasmid pGEM-hIFN-γdid contain the expected DNA sequence encoding the amino-acid sequence of human interferon-γ.Conclusion:A new coding sequence of hIFN-γgene was cloned successfully,and it was predicted for enhancing expression as done by other gene in bacteria.

Abstract:The porcine CD3e cDNA was amplified and cloned from total RNA of peripheral blood lymphocytes cultured under stimulation of concanavalin A (Con A) by RT-PCR and 3′RACE (rapid amplification of cDNA ends). Sequence analysis revealed that the amplified porcine CD3e cDNA was 1 241 nt in length, including 80nt of 5′un-translated region,591 nt of coding region and 570 nt of 3′un-translated region. The coding region encoded a predicted molecular mass of 22 kD protein precursor with 196 amino acids residues lacking N-glycosylation site. The porcine CD3e contained several characteristic motifs highly conserved across the species within the cytoplasmic domain, namely Src homology 3(SH3),endoplasmic reticulum (ER) retention motif and the immunoreceptor tyrosine-based activation motif(ITAM) which consisted of two Src homology 2(SH2) and two potential tyrosine phosphorylation sites at Tyr 177 and Tyr 188. These motifs were associated with signal transduction on T-cell activation as well as assembly and expression of TCR-CD3 complex. Comparison of the deduced amino acids sequence of porcine CD3e with those of human,mouse,dog,bovine and sheep showed that the amino acids homology were 61.2%, 58.7%,58.7%,65.1% and 65.6%, respectively.

Abstract:AIM:To clone human PD-L1 cDNA and construct a prokaryotic expression vector for the extracellular domain of PD-L1 gene.METHODS:The human PD-L1 cDNA was cloned by RT-PCR from the total RNA of activated human peripheral lymphocytes. The prokaryotic expression vector for the extracellular domain of PD-L1 gene was constructed and protein expression in E.coli strain BL21(DE3) was determined.RESULTS:The full-length gene coding sequence of PD-L1 was cloned and confirmed by DNA sequencing. The prokaryotic expression vector for the extracellular domain of PD-L1 gene fused with His(6) tag at the C-terminus was then constructed. The recombinant protein was expressed in E.coli after IPTG induction and identified by Western blot. The expressed product had a relative molecular mass (M(r)) being 22 000, which was consistent with theoretical value.CONCLUSION:The PD-L1 gene was successfully cloned and the extracellular domain of the protein was expressed in E.coli, which lays the foundation for further study of the function of PD-L1.

Abstract:Objective: To clone THANK gene and express its extracelluar fragment. Methods: Using RNA isolated from HL 60 cell lines, THANK cDNA was amplified by RT PCR. The fragment was linked to pMD18 T vector and sequenced, and then the extracellular fragment of THANK was subcloned into pET vector and THANK protein expression was induced.Results: A 858 bp DNA fragment was amplified and the cDNA sequence was identical with the published sequence encoding THANK gene. Western blot showed that THANK protein with a relative molecular weight of 2.6×10 4 was expressed. Conclusion: Human THANK gene was cloned and expressed successfully, which provides a base of further research of THANK gene. [

Abstract:Aim: To clone the cDNA of human EGFR extracellular domain and to construct the mammalian cell expression vector for soluble EGFR (sEGFR). Methods: The cDNA encoding EGFR extracellular domain was cloned by RT-PCR from human placenta and its sequence was determined. The DNA fragment was inserted into pEGFP-N1 to construct an expression vector for sEGFR. Results: The cDNA encoding the extracellular domain of EGFR was cloned from human placenta villi and the expression vector for sEGFR was constructed by PCR using a primer pair to introduce a Kozak sequence before ATG and to insert a stop codon. The protein encoded by this gene contained domains L1, S1, L2, S2 as well as signal peptide. Sequence analysis revealed that there were 3 nucleotides different from the previously published gene, with G1562→A, G1620→C and T1887→A, respectively. The former two nucleotide substitutions led to amino acid changes at position 497 and 516 from Arg and Lys to Lys and Asn respectively, whereas the latter one was a synonymous mutation (Thr605). Conclusion: The cDNA encoding the extracellular domain of EGFR was successfully cloned and the expression vector for sEGFR was constructed.
Cloning and Expression of Human LXR-LBD Cdna in E.coli

Abstract:To obtain PPARγ-LBD cDNA, the total RNA was isolated from facil adipose tissue of Chinese and cDNA was synthesized by RT-PCR. The specific amplification product was cloned and sequenced. Then PPARγ-LBD cDNA was inserted into prokaryotic expression vector pET28a and made it expressed in E. coli. The expression product fused with 6 x His at N-terminal was analyzed by Western-blot, and purified by the Ni2+-NTA ion exchange resin and the purity of PPARγ-LBD was analyzed by SDS-PAGE. The results showed that human PPARγ-LBD cDNA has been obtained and was identical with Gene Bank reports. The molecular weight of expressed product analyzed by Western-blot was about 34 kDa. The purity of the recom-bined PPARγ-LBD was more than 90% after purified by the Ni2+ -NTA ion exchange resin. Thus, The sequence of Chinese PPARγ-LBD was achived, and PPARγ-LBD was expressed and purified successfully in E. coli.

Abstract:According to the cDNA sequence of HSA(human serum albumin HSA) derived from the Genebank(V00495), a pair of primers was designed. RT-PCR was applied to amplify the whole cDNA of HSA gene. The DNA fragment was cloned into vector pMD-18T and transformed into E.coli DH5α host bacteria. XbaⅠendonuclease digestion and sequenced from selected positive cloned bacteria. The results showed that the whole 1995 bp cDNA of HSA were cloned successfully and compared with the sequence deposited in Gene Banks, and the homology was 99%.

Abstract:A 3300‐bp cDNA (6A8) has been isolated from a human tonsil cell λgt11 cDNA library (GenBank accession number: AF044414). The 6A8 gene is localized on human chromosome 13q31–32. Its cDNA has an open reading frame from position 57 bp to 3243 bp, encoding a 1062 amino‐acid polypeptide. The sequence of the polypeptide has 89% identity to rat liver ER α‐mannosidase. Homogenates of COS‐7 cells transfected with 6A8 cDNA showed an enhanced enzymatic activity with p‐nitro‐phenyl‐α‐D‐mannopyranoside, which was not inhibited by swainsonine. These data suggest that 6A8 α‐mannosidase belongs to the class II α‐mannosidase. Western blot analysis showed a band for 6A8 cDNA encoded protein of ≈120 kDa. Northern blot analysis revealed two 6A8 mRNA transcripts with different tissue distribution. Enhanced concanavalin A (ConA) binding to CNE‐2L2 cells transfected with a reverse 6A8 DNA was observed, indicating that the 6A8 protein is an important cellular α‐mannosidase.

Abstract:Objective:To clone and express human renalase gene.Methods:Total RNA was extracted in human kidney and the cDNA of renalase was obtained by reverse-transcription PCR.The renalase cDNA was inserted into the expression vector pET42b(+)and was transformed into E coli BL21.The fusion protein was induced by IPTG and purified by affinity chromatography.The products were identified by SDS-PAGE and Western blot analysis.Results:The renalase cDNA was obtained and was 1 029 bp.The renalase fusion protein was obtained,whose molecular weight was 38 kD.Conclusion:The human renalase gene was cloned and expressed in E coli successfully,which paved the way for further research of the renalase function.
