Abstract:OBJECTIVE:To observe the changes of auditory electrophysiology and inner ear pathology in rat cochlea after the noise exposure, and to offer the experimental data for exploring the mechanism of noise-damaged cochlea. METHOD:The rats in the study group were exposed to a intense narrow band noise centered at 4 kHz at the leave of 120 dB (SPL) for 4 h. The exposed cochleae were collected at various intervals (1 or 21 days) after the noise exposure. Auditory function was monitored by measuring thresholds of auditory brain stem responses (ABR). The morphological changes in rat cochlear hair cell (HC) were examined by HC nuclei stained with Propidium iodide (PI), a fluorescent dye specifically labelling the nuclear DNA and scanning electron microscopy (SEM). The number of spiral ganglion cells was calculated using pathologic technique. RESULT:The thresholds of ABR in the study group were significantly greater than that in the normal control group (P < 0.01). Examined at 1 day after the noise exposure, normal, apoptosis, necrotic and missing out hair cell (OHC) could be distinguished with PI staining, whereas the apoptosis OHC were not found at 21 days. Significant OHC loss was found in as compared to the normal control group (P < 0.01). There was not significant difference in the calculation of spiral ganglion cells (P > 0.05). SEM revealed the injured stereocilia of OHC (disarrangement, collapse) and OHC loss in the study group, which was more severe in OHC3 than the other two rows of OHC. CONCLUSION:The intense noise used in our study could injure the rat cochlea and bring permanent threshold shift (PTS). Under this condition, the death modes of HC in the cochlea include apoptosis and necrosis in the fore part, whereas necrotic is the major mode in the evening of exposure. The injured stereocilia of OHC and OHC loss could remain the most consistent correlate of PTS.
Effect of Hypoxia on Expression of AQP4 and HIF-1αin Protein in Prefrontal Cortex of Rats

Abstract:Objective To investigate the expression of AQP4 and HIF-1 αprotein in the prefrontal cortex of SD rats under hypoxic conditions for the basis of elucidating the mechanism of hypoxic brain edema. Methods Twelve SD rats were randomly divided into the normal oxygen group and the hypoxia group.Rats in hypoxia group were exposed to hypoxia (7.8% O2 ,equal to 7 000 m altitude)in a hypobaric chamber for 8 h.The expression of AQP4 and HIF-1 αprotein in rat prefrontal cortex were determined by Western blot. Results AQP4 and HIF-1 α protein were enhanced in the hypoxic rat prefrontal cortex after exposure to 7 000 m hypoxia for 8 h (P <0.05)by Western blot. Conclusion The expression of AQP4 and HIF-1 αprotein in the prefrontal cortex of SD rats were increased after 8 h hypoxia.It was showed that HIF-1 might contribute to hypoxic brain edema by increasing the expression of AQP4 protein.
High Salt Challenge Augments Expression of Proinflammatory Cytokines and Contributes to Elevated Neuronal Activity in the Hypothalamic Paraventricular Nucleus

Abstract:Previous studies have demonstrated that brain proinflammatory cytokines (PIC) play an important role in neurogenic hypertension. However, little is known about the role of brain inflammation in salt sensitive hypertension (SSHTN). In this study, we tested the hypothesis that the PIC expression is increased in the hypothalamic paraventricular nucleus (PVN) and contributes to increased neuronal activity in SSHTN. To test this hypothesis, 8‐week‐old Dahl salt sensitive (DS) rats were divided into two groups (n=3~ 5/group) and fed with either a high salt (HS, 4% NaCI) or normal salt (NS, 0.4% NaCl) diet. Four weeks following diet treatment, rats were euthanized, the PVN was punched out, and real time PCR were performed to assay genes of interest. HS treatment resulted in respective 8 fold, 4 fold, and 7 fold increases in TNFα, IL1β and IL6 mRNA levels in the PVN. In addition, Fra1, a chronic neuronal activation marker, was also increased by 4 fold. Additionally, in order to examine the action of acute hypertonic NaCI on the expression of PIC and neuronal activation marker, adult male SD rats were divided into two groups (n=3~4 per group) and received intracerbroventricular infusion of either hypertonic (2M, 2μl), or isotonic (0.15 M, 2μl) NaCI under anesthetized condition. Three hours following the infusion, rats were euthanized and PVN mRNA levels of PIC and Fra1 assayed. Results indicate that hypertonic saline infusion induced significant increases in mRNA levels of PIC including TNFα (6‐fold), IL1β (13‐fold) and IL6 (21‐fold), as well as neuronal activation marker Fra1 (8‐fold). Finally, we tested whether or not the increase in PIC expression occurred in neurons. Incubation of hypertonic saline (10 mM NaCI plus full medium) for 3 hours resulted in a 12‐fold increase in IL6 and 2.8 fold increase in Fra1 mRNA levels in neuronal cultures from the hypothalamus containing the PVN of neonatal SD rats. Abstract:This study was designed to examine the effect of infrasound exposure (5 Hz at 130 dB) on whole-cell L-type Ca 2+ currents (WLCC) in rat ventricular myocytes and the underlying mechanism(s) involved. Thirty-two adult Sprague-Dawley rats were randomly assigned to infrasound exposure and control groups. [Ca 2+ ] i , WLCC, mRNA expression of the a 1c subunit of L-type Ca 2+ channels (LCC), and SERCA2 protein were examined on day 1, 7, and 14 after initiation of infrasound exposure. Fluo-3/AM fluorescence and the laser scanning confocal microscope techniques were used to measure [Ca 2+ ] i in freshly isolated ventricular myocytes. The Ca 2+ fluorescence intensity (FI), denoting [Ca 2+ ] i in cardiomyocytes, was significantly elevated in a time-dependent manner in the exposure groups. There was a significant increase in WLCC in the 1-day group and a further significant increase in the 7- and 14-day groups. LCC mRNA expression measured by RT-PCR revealed a significant rise in the 1-day group and a significant additional rise in the 7- and 14-day groups compared with control group. SERCA2 expression was significantly upregulated in the 1-day group followed by an overt decrease in the 7- and 14-day groups. Prolonged exposure of infrasound altered WLCC in rat cardiomyocytes by shifting the steady-state inactivation curves to the right (more depolarized direction) without altering the slope and biophysical properties of I Ca,L . Taken together, our data suggest that changes in [Ca 2+ ] I levels as well as expression of LCC and SERCA2 may contribute to the infrasound exposure-elicited cardiac response.

Abstract:OBJECTIVE: This study aims to investigate the biological significance of heat shock protein (HSP70) in adaptation to high altitude hypoxia.METHODS: Ninety male SD rats were randomly divided into 10 groups: Acute plateau hypoxia group (group 5) and chronic high altitude hypoxia group (group 3), and control group (group 2).Changes in HSP gene and HSP protein expression in brain tissues of SD rats at different altitudes were determined by Western blot and conventional RT-PCR, while an optical and transmission electron microscope was used to observe the cell structure changes of animal brain tissues.RESULTS: HSP70 expression rapidly increased at high altitudes in SD rats under high-altitude hypoxia environments, and HSP70 increased with altitude. Morphological and structural damage in SD rats in each group increased with altitude.CONCLUSION: The rapid synthesis of HSP70 in heat shock response is beneficial for maintaining the normal physiological function of cells during hypoxia stress, and the amount of HSP70 production is positively correlated with hypoxia tolerance ability.

Induction of Heat Shock Proteins 27, 60, 70, and 90 in the Cerebellum of Rats After Hyperthermia During Postnatal Development

Abstract:Heat shock proteins (HSPs) are induced in response to stressful stimuli and play an important role in cell repair and protection. This study, using immunohistochemistry, aimed to determine whether HSPs are induced in the cerebellum of rats subjected to hyperthermia during postnatal development (PND). The results showed that unlike HSP27 and HSP70, HSP60 and HSP90 were constitutively expressed in the cerebellum of rats. However, hyperthermia induced HSP27 in the white matter (WM) and HSP70 in the Bergmann glial cells, the internal granule layer (IGL), and the WM. In the WM, HSP27 induction was only observed on days PND20, PND25, and PND30, and HSP27 expression was higher on day PND30 compared with days PND20 and PND25 (p < 0.001). In the Bergmann glial cells, HSP70 induction was only observed on days PND5, PND10, and PND20, and HSP70 expression was greater on days PND5 and PND10 compared with day PND20 (p < 0.001). In the IGL and the WM, HSP70 expression was higher on days PND20, PND25, and PND30 compared with days PND5 and PND10 (p < 0.001). These findings indicate that unlike HSP60 and HSP90, HSP27 and HSP70 have different expression patterns in the cerebellum of rats after hyperthermia during PND.
[Effects of Infrasound on Activities of 3beta Hydroxysteroid Dehydrogenase and Acid Phosphatase of Polygonal Cells in Adrenal Cortex Zona Fasciculate in Mice].

Abstract:OBJECTIVE:To explore the biological effects of infrasound on the polygonal cells in adrenal cortex zona fasciculation in mice.METHODS:The biological effects of infrasound on the activities of 3beta hydroxysteroid dehydrogenase (3-betaHSDH) and acid phosphatase(ACP) of the polygonal cells in adrenal cortex zona fasciculate were observed when exposure to 8 and 16 Hz infrasound at 80, 90, 100, 110, 120 and 130 dB for 1 day, 7 days and 14 days or 14 days after the exposure.RESULTS:When exposure to 8 Hz infrasound, the enzyme activities of 3-betaHSDH increase as the sound pressure levels increase. Only when the sound pressure levels reach 130 dB, the enzyme activities began to decrease exceptionally. When exposure to 16 Hz, 80 dB infrasound, no significant difference between the treatment and control group in the activities of 3-betaHSDH could be observed, but the injury of the polygonal cells had appeared. When exposure to 16 Hz, 100 dB infrasound, the activities of 3-betaHSDH started to increase. The cell injury still existed. When exposed to 16 Hz, 120 dB infrasound, the local tissue damage represented. Fourteen days after the mice exposure to 8 Hz, 90 dB and 130 dB infrasound for 14 days continuously, the local tissue injury of the adrenal cortex zona fasciculation began to recover at certain extent, but the higher the exposure sound pressure level, the poorer the tissue recovery.CONCLUSION:The biological effects of infrasound on the polygonal cells in adrenal cortex zona fasciculation response to the frequency of the infrasound are found at certain action strength range, but this characteristic usually is covered by the severe tissue injury. When exposure to infrasound is stopped for a period of time, the local tissue injury of the adrenal cortex zona fasciculation could recovers at certain extent, but the higher the exposure sound pressure level, the more poorer the tissue recovery.

Abstract:Objective To observe the expression of heat shock protein 70(HSP 70)in rat spinal cord with acute contusion.Methods Sixty-five SD rats were divided into 3 groups:5 in control group,30 in operative group,30 in spinal cord injury group.The injuries of spinal cord were induced by Allen's model.Spinal cords observed in operative group were collected from 5 rats each time at 6 time points(2 h,6 h,12 h,24 h,48 h and 72 h after management),as same as the spinal cord injury group.HSP 70 expression in spinal cords from each group at different time points was studied by method of immunohistochemistry.Results There was not constitutive expression of HSP 70 in the normal spinal cord.But expression of HSP 70 had been already found at 2 h after the spinal cord contusion,reached the peak at 24～48 h,and could been observed at 72 h after contusion.Conclusion Expression of HSP 70 is a part of stress reaction for organism.HSP 70 obviously increased in gray nucleus of spinal cord from 2 h to 72 h after acute injury.HSP 70 maybe make for preventing secondary lesion of spinal cord.
Study of expression of heat shock protein 70 in Guinea Pigs after the irradiation of carbon dioxide laser

Abstract:[Objective] To study the relationship among the expression of the heat shock protein 70(Hsp70), the change of ultrastructure and hearing function of the cochlea after the irradiation of the carbon dioxide laser in guinea pigs. [Methods] 36 guinea pigs were equally divided into 3 groups, irradiated with carbon dioxide laser of 1w, 2w and 3w. Auditory brainstem response(ABR) were recorded respectively. Scanning electron microscopies were performed to observe the morphological alteration. The expression of the Hsp70 was detected by SABC. [Results] It was elucidated that the thresholds of ABR immediately after irradiation were higher than those preoperative in three groups, the ABR thresholds three weeks later were higher than those before irradiation in group 2w and 3w, no significance was noted in group 1w. Three weeks later, through scanning electron microscopies. It was found that cochlea hair cells were slightly affected by low power(1w) CO2 laser, with 2w CO2 laser, the outer hair cells and stereocilia in the Corti′s organ were disarranged, with 3w CO2 laser, regional disorder or serious loss of stereocilia. The level of immunoreactivity in cochlea immediately was higher than that in control group, Hsp70 presented in 2w and 3w group three weeks later was stronger than that in control group, whereas no difference was observed in 1w group. The rise of the ABR thresholds was correlated with the strong expression of the Hsp70 both immediately and three weeks later in A and B group, and immediately in C group, but no significance was noted three weeks later in group C. [Conclusion] Our findings indicate that the expression of Hsp70 was induced in guinea pigs cochlea after laser irradiation, which relates to the damage degree of the cochlea and the protection of the auditory function.

Abstract:We demonstrated previously that hypoxia activated CRF and CRF mRNA in PVN, and CRF receptor 1 (CRFR1) mRNA in rat pituitary. The aim of the study is to test whether the hypoxia-activated CRF and CRF mRNA is associated with triggering CRFR1. Rats were exposed to hypobaric hypoxia at altitude of 2 and 5 km. CRF and CRF mRNA were assayed by immunostaining and in situ hybridization. CRFR1 mRNA was assayed by RT-PCR. Results showed that 5 km continual hypoxia increased CRF and CRF mRNA in PVN, CRFR1 mRNA in pituitary, and plasma corticosterone. The hypoxia-increased CRF, CRF mRNA, CRFR1 mRNA, and corticosterone were blocked by CRFR1 antagonist (CP-154,526), suggesting that CRFR1 in PVN and pituitary are responsible for the hypoxia-increased CRF and CRF mRNA in PVN. Abstract:Exposure to intense sounds often leads to loss of hearing of environmental sounds and hearing of a monotonous tonal sound not actually present, a condition known as tinnitus. Chronic physiological effects of exposure to intense tones have been reported for animals and should be accompanied by chemical changes present at long times after the intense sound exposure. By using a microdissection mapping procedure combined with high-performance liquid chromatography (HPLC), we have measured concentrations of nine amino acids, including those used as neurotransmitters, in the cochlear nucleus, inferior colliculus, medial geniculate, and auditory cortex of hamsters 5 months after exposure to an intense tone, compared with control hamsters of the same age. No very large differences in amino acid concentrations were found between exposed and control hamsters. However, increases of glutamate and ?-aminobutyrate (GABA) in some parts of the inferior colliculus of exposed hamsters were statistically significant. The most consistent differences between exposed and control hamsters were higher aspartate and lower taurine concentrations in virtually all regions of exposed hamsters, which reached statistical significance in many cases. Although these amino acids are not considered likely neurotransmitters, they indirectly have roles in excitatory and inhibitory neurotransmission, respectively. Thus, there is evidence for small, widespread, long-term increases in excitatory transmission and decreases in inhibitory transmission after a level of acoustic trauma previously shown to produce hearing loss and tinnitus. Abstract:As a kind of environmental noise, infrasonic noise has negative effects on various human organs. To date, research has shown that infrasound impairs cognitive function, especially the ability for learning and memory. Previously, we demonstrated that impaired learning and memory induced by infrasound was closely related with glia activation; however, the underlying mechanisms remain unclear. Connexin 43 hemichannels (Cx43 HCs), which are mainly expressed in hippocampal astrocytes, are activated under pathological conditions, lending support to the hypothesis that Cx43 HCs might function in the impaired learning and memory induced by infrasound. This study revealed that that blocking hippocampal Cx43 HCs or downregulating hippocampal Cx43 expression significantly alleviated impaired learning and memory induced by infrasound. We also observed that infrasound exposure led to the abundant release of glutamate and ATP through Cx43 HCs. In addition, the abundant release of glutamate and ATP depended on proinflammatory cytokines. Our finds suggested that the enhanced release of ATP and glutamate by astroglial Cx43 HCs may be involved in the learning and memory deficits caused by infrasound exposure.

Expression of heat shock protein 70 and inducible nitric oxide synthetase in guinea pig cochlea after irradiation of carbon dioxide laser

Abstract:Objective To investigate the relationships among the expression of heat shock protein(Hsp70),inducible nitric oxide synthase(iNOS) and the hearing function after the irradiation of carbon dioxide laser in guinea pig cochlea.Methods Thirty-six guinea pigs after the fenestration on the basal turn of the left cochlea were divided into 3 groups(group A,B and C),who were irradiated with carbon dioxide laser with 1,2 and 3 W respectively.Auditory brainstem response(ABR) was recorded one day before the operation and antemortem respectively.Animals were decapitated in batch immediately and three weeks after irradiation.The expression of Hsp70 and iNOS was detected by streptavidin-biotin-peroxidase complex immunohistochemistry method and the average gray densities were quantified with a light microscope image analysis system.Results It was found that the thresholds of ABR immediately after irradiation were all higher than those preoperative in all three groups(P0.05).The ABR thresholds three weeks later were higher than those before irradiation in group B and C(P0.05),no significance being noted in group A(P0.05).The level of immunoreactivity in cochlea immediately after irradiation was higher than that in the control group(P0.05).The expression of Hsp70 and iNOS in group B and C three weeks after irradiation was stronger than that in the control group(P0.05),whereas no difference was observed in group A(P0.05).The rise of the ABR thresholds in guinea pigs was correlated with the strong expression of Hsp70 and iNOS both immediately and three weeks after laser irradiation in group A and B(P0.05),and immediately after irradiation in group C(P0.05),but no significance was noted three weeks later after irradiation in group C(P0.05).Conclusions Our findings indicate that the expression of Hsp70 and iNOS was induced in guinea pigs cochlea after laser irradiation,which relates to the damage degree of the cochlea.
Hsp70 ameliorates sleep deprivation-induced anxiety-like behavior and cognitive impairment in mice

Abstract:Background: Many neurobehavioral processes, including psychomotor, cognitive, and affection are negatively impacted by sleep deprivation (SD), which may be harmful to a person's physical and mental health. Heat shock proteins (Hsps) have been demonstrated to play a protective role in a number of neurodegenerative diseases and are essential for maintaining intracellular protein homeostasis, but their roles in SD remain elusive. Methods: A mouse SD model was constructed using a modified multi-platform water environment method. The cognitive function was tested by novel object recognition test and Y-maze test, and anxiety-like behaviors were assessed by open field test (OFT). Protein expression was determined by Western blotting assay and ELISA assay. Results: We found that SD could profoundly enhance anxiety levels and impair cognitive function in mice. SD also reduced the expression levels of p-cAMP-response element binding protein (CREB) and brain-derived neurotrophic factor (BDNF) and increased microglial activation and neuroinflammatory response in the hippocampus of mice. The intranasal injection of human recombinant Hsp70 protein could alleviate SD-induced anxiety and cognitive impairment, as well as restore pCREB and BDNF levels and reduce microglia-induced neuroinflammation in the hippocampus of SD mice. Conclusions: Hsp70 treatment might serve as a potential treatment for mitigating SD-related unfavorable symptoms.

Abstract:Objective To study the characteristics of infrasound and its effects on the worker8 at power plants.Methods The audible noise and infrasound in three thermoelectricity plants were measured and by using Symptom Checklist 90(SCL-90),the subjective sensation,and the physiological indices were compared between exposed workers and controls.Results The infrasound levels were different at different measure sites of the three thermoelectricity works ranging from 40 to 98 dB.There was still 40～80 dB infrasound even when the soot blower stopped running.Different apparatus produced different infrasound levels with the highest ranging from 62 to 115 dB.A single frequency(16 Hz)infrasound was produced in certain department during working hour with sound pmssme levels of 110 to 120 dB.but the audible noise sound pressure level was less than 70 dB.There was no significant difference in the indices representing vision fatigue and neurobehaviour function between exposed workers and controls.Workers at certain department experienced evident subjeetive sensation of neurobehavioral dysfunction,and the scores of somatization,depression,hostility,phobic anxiety,and psychotism in the SCL-90 were significantly higher in the exposed group than in the control and the norm in China(P＜0.05).Conclusion Infrasound is ubiquitous in the working environment,but usaully,the noise levels are less than 120 dB.In some special production department.there is persistent infrasound above 110 to 120 dB.No obvious health effects are found among those who are exposed to infrasound below 100 dB.However,the workers who are chronically exposed to infrasound above 110 to 120 dB present notable subjective sensation of autonomic neurobehavioral dysfunction.and their psychological health status is not as good as those in the control and those in the domestic normal pattern.
A terrified-sound stress induced proteomic changes in adult male rat hippocampus

Abstract:In this study, we investigated the biochemical mechanisms in the adult rat hippocampus underlying the relationship between a terrified-sound induced psychological stress and spatial learning. Adult male rats were exposed to a terrified-sound stress, and the Morris water maze (MWM) has been used to evaluate changes in spatial learning and memory. The protein expression profile of the hippocampus was examined using two-dimensional gel electrophoresis (2DE), matrix-assisted laser desorption/ionization time-of-flight mass spectrometry, and bioinformatics analysis. The data from the MWM tests suggested that a terrified-sound stress improved spatial learning. The proteomic analysis revealed that the expression of 52 proteins was down-regulated, while that of 35 proteins were up-regulated, in the hippocampus of the stressed rats. We identified and validated six of the most significant differentially expressed proteins that demonstrated the greatest stress-induced changes. Our study provides the first evidence that a terrified-sound stress improves spatial learning in rats, and that the enhanced spatial learning coincides with changes in protein expression in rat hippocampus.

Abstract:Objective: To investigate the effects of acute noise stress on two amine stress hormones, norepinephrine (NE) and 5-hydroxyindoleacetic acid (5-HIAA) in the brain and plasma of mice after noise exposure. Methods: Mice were grouped into the control and noise groups. Mice in the noise group were exposed to white noise of 110dB sound pressure level for 60min. Auditory brainstem response thresholds, distortion product otoacoustic emissions, the organ of Corti grading scores, western blots of NE/5-HIAA in the whole brain and hippocampus, and the plasma levels of NE/5-HIAA were compared between the two groups. Results: Significant hearing loss and cochlear damage were demonstrated in the noise group. NE and 5-HIAA in the hippocampus were elevated in the noise group (p=0.019/0.022 for NE/5-HIAA vs. the control). Plasma levels of NE and 5-HIAA were not statistically different between the groups (p=0.052/0.671 for NE/5-HIAA). Conclusion: Hearing loss with outer hair cell dysfunction and morphological changes of the organ of Corti after noise exposure in C57BL/6 mice proved the reliability of our animal model as an acute noise stress model. NE and 5-HIAA are suggested to be the potential biomarkers for acute noise stress in the hippocampus.
Psychophysical and neural correlates of noised-induced tinnitus in animals: Intra- and inter-auditory and non-auditory brain structure studies

Abstract:Tinnitus, a ringing in the ear or head without an external sound source, is a prevalent health problem. It is often associated with a number of limbic-associated disorders such as anxiety, sleep disturbance, and emotional distress. Thus, to investigate tinnitus, it is important to consider both auditory and non-auditory brain structures. This paper summarizes the psychophysical, immunocytochemical and electrophysiological evidence found in rats or hamsters with behavioral evidence of tinnitus. Behaviorally, we tested for tinnitus using a conditioned suppression/avoidance paradigm, gap detection acoustic reflex behavioral paradigm, and our newly developed conditioned licking suppression paradigm. Our new tinnitus behavioral paradigm requires relatively short baseline training, examines frequency specification of tinnitus perception, and achieves sensitive tinnitus testing at an individual level. To test for tinnitus-related anxiety and cognitive impairment, we used the elevated plus maze and Morris water maze. Our results showed that not all animals with tinnitus demonstrate anxiety and cognitive impairment. Immunocytochemically, we found that animals with tinnitus manifested increased Fos-like immunoreactivity (FLI) in both auditory and non-auditory structures. The manner in which FLI appeared suggests that lower brainstem structures may be involved in acute tinnitus whereas the midbrain and cortex are involved in more chronic tinnitus. Meanwhile, animals with tinnitus also manifested increased FLI in non-auditory brain structures that are involved in autonomic reactions, stress, arousal and attention. Electrophysiologically, we found that rats with tinnitus developed increased spontaneous firing in the auditory cortex (AC) and amygdala (AMG), as well as intra- and inter-AC and AMG neurosynchrony, which demonstrate that tinnitus may be actively produced and maintained by the interactions between the AC and AMG.
Expression of HSP 70 in the basilar arteries of rabbits after experimental subarachnoid hemorrhage and its meanings

Abstract:Objective To study the role of heat shock protein 70 (HSP70) in the cerebral vasospasm after subarachnoid hemorrhage (SAH) in rabbits.Methods Fifteen Japanese rabbits were randomly divided into control group (n=3) and experimental groups (n=12),who were redivided into 4 subgroups of 3 animal each according to the time from SAH to sacrifice.The SAH was produced in the rabbits of all the experimental groups by injecting autologous fresh arterial blood into the cisternae magna.The basilar arteries were taken from the animals,who in the experimental group were sacrificed respectively 3,7,14 and 21 days after SAH.The gross and histological changes in the basilar arteries of all the rabbits were observed.The expressions of HSP70 in the basilar arteries of all the rabbits was determined by immunohistochemical technique.Results The gross and histological changes in the basilar arteries after SAH included thickening of the arterial walls,narrowing of the arterial lumina,swelling and deformation of the endothelial cells,wrinkling of the elastic interna and so on.The expression levels of HSP70 in the endothelial cells of the basilar arteries were significantly lower 3,7 and 14 days after SAH than that in the control group (P <0.01).Conclusion It is suggested that the cerebral vasospasm after SAH may be related to the decrease in HSP70 expression level in the cerebrovascular endothelial cells.

Abstract:Background: Noise induced injury of the cochlea causes shifts in activation thresholds and changes of frequency response in the inferior colliculus (IC). Noise overexposure also induces pathological changes in the cochlea, and is highly correlated to hearing loss. However, the underlying mechanism has not been fully elucidated. In this study, we hypothesized that overexposure to noise induces substantial electrophysiological changes in the IC of guinea pigs.Results: During the noise exposure experiment, the animals were undergoing a bilateral exposure to noise. Additionally, various techniques were employed including confocal microscopy for the detection of cochlea hair cells and single neuron recording for spontaneous firing activity measurement. There were alterations among three types of frequency response area (FRA) from sound pressure levels, including V-, M-, and N-types. Our results indicate that overexposure to noise generates different patterns in the FRAs. Following a short recovery (one day after the noise treatment), the percentage of V-type FRAs considerably decreased, whereas the percentage of M-types increased. This was often caused by a notch in the frequency response that occurred at 4 kHz (noise frequency). Following a long recovery from noise exposure (11-21 days), the percentage of V-types resumed to a normal level, but the portion of M-types remained high. Interestingly, the spontaneous firing in the IC was enhanced in both short and long recovery groups.Conclusion: Our data suggest that noise overexposure changes the pattern of the FRAs and stimulates spontaneous firing in the IC in a unique way, which may likely relate to the mechanism of tinnitus.
