The Effect of CsA and rIL-2 on T Lymphocyte Subsets in Rat Peripheral Blood Following Orthotopic Liver Transplantation

Abstract:Objective To explore whether CD4+/CD8+ T cell ratios could be used as a mark in early diagnosing liver transplant rejection or not and whether rIL-2 could reverse CsA-induced tolerance or not.Methods Animals were divided into groups:1.WistarWistar+rIL-2,group 2.SDWistar,and group 3.SDWistar+CsA+rIL-2.T lymphocyte subsets in peripheral blood were assayed by immunofluorescence staining and flow cytometry on the pre-set time points in the period of 35 days after orthotopic liver transplantation in rats.Results Median survival was 14 days in untreated allogeneic rats.In the syngeneic WistarWistar+rIL-2,the CD4+/CD8+ T cell ration remained unchanged.There was a significant fall in CD4+/CD8+ T cell ration in allogeneic untreated SDWistar from the 3~14 day,particularly on the 9th day reaching the lowest level 0.84±0.14.This abnormality in CD4+/CD8+ T cell ratio occurred 7~9d earlier than the classic signs of acute liver allogeneic rejection in histology examination.In the allogeneic SDWistar+CsA+rIL-2,there was a fall in CD4+/CD8+ T cell ratio (ranged from 0.72±0.01 to 1.11±0.04)during the CsA-treated period,which was caused by a decrease in CD4+/CD8+ T cell.On the 30th day,however,this ratio increased marginally.In the following rIL-2 treatment period,the CD4+/CD8+ T cell ratio declined again and all animals died within 8.5 days. Conclusions Serial changes in peripheral blood T lymphocyte subsets may be useful in prognosticating acute rejection and the CsA-induced to lerance could be reversed by injecting rIL-2 into rats subsets.
