Influence of Negative Pressure Wound Therapy Combined with Topical Oxygen Therapy on Burn Wound Healing in Rats with Diabetes Mellitus

Abstract:Objective To observe influence of negative pressure wound therapy (NPWT) combined with topical oxygen therapy on burn wound healing in rats with diabetes mellitus.Methods Diabetic SD rat models were established using Streptozotocin to induce by intraperitoneal injection,and the skin scald models were made using steam sprayer on the back of the model.A total of 90 diabetic SD rats with back burn were randomly divided into diabetic scald control group (group A),NPWT treatment group (group B) and NPWT combined with topical oxygen therapy group (group C,n =30 for each group),and all rats were given corresponding treatments.Wound healing rates were measured at 7th d,14th d and 21th d after injury,and the specimens were collected for routine pathological examination.Expressions of vascular endothelial growth factor (VEGF) in wound tissues were detected by Western blotting.Results Values of wound healing rates and VEGF protein expressions in group B and C were significantly higher than those in group A at 7th d,14th d and 21th d after injury (P <0.01).Values of wound healing rates and VEGF protein expressions in group C were significantly higher than those in group B at 7th d and 14th d after injury (P < 0.01).Pathological examination showed that the wound had little inflammatory reaction and faster growth of granulation tissues and epithelial cells.Conclusion NPWT combined with topical oxygen therapy can promote the growth of new blood vessels in the wound tissues,improve the efficiency of wound healing and significantly shorten the wound healing time compared with those by NPWT treatment.
