Dissection of the Differentiation of Human Monocyte Subtypes to Osteoclasts at Single-Cell Resolution

Abstract:Peripheral blood monocytes (PBMs) were derived from the bone marrow hematopoietic stem cells (HSCs), which were involved in inflammatory responses and tissue homeostasis. Monocytes could differentiate into dendritic cells, macrophages, and osteoclasts. Although previous studies have identified several monocyte subtypes and analyzed their functions, few studies comprehensively explored the differentiation of monocyte subtypes to osteoclasts via state-of-the-art bioinformatics methods at single-cell resolution. Here, we innovatively used the DrImpute method to impute the single-cell RNA sequencing (scRNA-seq) dataset of PBMs in a healthy normal subject. Based on the imputed gene expression patterns and cell lineage reconstruction, we identified five distinct clusters including classical subtype, nonclassical subtype, and three intermediate subtypes. Intermediate1 subpopulation expressed PLD4 and FCER1A genes was a novel subpopulation and had the potential function in the establishment of protein localization to membrane and neutrophil activation. To further explore the functions of different monocyte subtypes in osteoclast differentiation, we performed functional enrichment analyses, protein-protein interaction (PPI) network analyses, and cell-to-cell communication analyses. We found that some signaling pathways and cytokines promoting osteoclast differentiation were enriched in the classical and intermediate2 monocyte, while some signaling pathways that inhibit osteoclast differentiation were enriched in the nonclassical monocytes. Therefore, we inferred that the classical subtype and intermediate2 subtype were the main subtypes that could differentiate into osteoclasts, and maintain bone mass balance. Based on PPI analysis, we identified eight interesting genes [FCGR3A and FCGR3B, TYROBP, FOS, TAB1, IL1R1, PIK3R2, and MAP2K ] invovled in the osteoclast differentiation of PBMs. Our studies provide a novel perspective to explore the relationship between monocytes and bone homeostasis at the single-cell level.
