Expression of speckle-type POZ protein in bladder cancer

Abstract:Objective To explore the expression and significance of speckle-type POZ protein (SPOP) in bladder cell cancer.Methods Totally 32 samples of bladder carcinoma and 20 samples of normal bladder either from mucosa tissue ＞ 5 cm outside of the cancer tissue or from the normal bladder tissue of prostate operation were selected.The expression of SPOP in bladder cancer and normal bladder tissues were detected by immunohistochemistry and Western blot;the expressions of SPOP in bladder cancer lines 5 637,T24,J82 and normal bladder cell line SV-HUC-1 were also detected by Western blot.Results The positive rate of SPOP expression in bladder cancer was 65.6％ (21/32),lower than that in normal bladder tissue 100％ (20/20);the expression level of SPOP was significantly lower in bladder cancer tissues than that in normal bladder tissue (0.21 ±0.08 vs 0.73 ± 0.06),and was significantly lower in bladder cancer cell lines 5 637,T24,J82 than that in normal bladder cell line SV-HUC-1 (0.43±0.05,0.14±0.06,0.03±0.01 vs0.72±0.14)(P＜0.01).Conclusion SPOP may be a molecular bio-marker for bladder cancer,and associated with the development and progression of bladder cancer.
