Abstract:Objective To determine whether overload cytosolic calcium ion was involved in down regulation of β-adrenoceptor densities in patients with chronic congestive heart failure(CHF).Methods 26 Patients with CHF (average age 56±7.5ys)(NYHA Ⅱ,Ⅲ)were randomly admitted to the study.15 Additional subjects (average age 55±5.6ys),with no history of cardiac disease, hypertension or other cardiovascular disorders, had early morning resting blood specimens obtained as control.[3H]-Dihydroalprenolol (DHA)radioligand binding assay was used to measure lymphocyte β-AR density, fluorescent indicator Fura2-AM to estimate platelet cytosolic calcium concentration (PCC) and 125I-radioimmunoassay to measure plasma norepinephrine (NE)levels. Results β-AR densities in CHF patients were lower than that in control group (P<0.001).PCC and plasma NE levels in CHF patients were significantly higher than that in control group (P<0.001,respectively).Results of correlation analysis showed that there was significant negative correlation between β-AR and PCC in CHF patients (r2=0.58,P<0.001).But there was no significant correlation between β-AR and plasma NE levels in CHF patients. In control group,no significant correlation either between β-AR and PCC or between β-AR and plasma NE levels was observed in this study. Conclusion Overload cytosolic calcium ion was involved in the down regulation of β-adrenoceptor densities in CHF patients.
