Study on the Effects of Derivatives of OGP(10-14) on Bone Mineral Density and Bone Biomechanics in Ovariectomized Rats

Abstract:Objective: To study the effects of derivatives of OGP(10~14) G38I and G38K on bone metabolic characteristics, bone mineral density (BMD) and bone biomechanics in ovariectomized rats, and investigate the relationships between these indexes. Methods: Forty female Sprague-Dawley rats with three month old were randomly divided into 5 groups including sham-operated (S) group and ovariectomy model (M) group as negative control groups and other three groups with G38I, G38K and alendronate respectively after bilaterally ovariectomized (OVX). All rats were sacrificed 60 days after the operation. The serum Ca, P and ALP, the BMD of femora and lumbar vertebra, the ratio of ash weight to dry weight and the biomechanical parameters were measured. Results: Serum Ca and P increased and the ratio of ash weight to dry weight decreased in group M compared with group S. All these indexes returned back to the level of group S after treated with G38I and G38K, the lumbar vertebral BMD significantly increased, and the femoral maximal stress enhanced in group M. The ratio of ash weight to dry weight showed a positive correlation with BMD. There was no relationship between BMD and biomechanical parameters. Conclusion: The bone loss decreases and the biomechanics improves in ovariectomized rats with the treatment of G38I and G38K. It's important to pay attention to the bone quantity and quality in the evaluation of drug effects.
