Expression of HSP7O and MHC-I antigen in hepatocellular carcinoma and its significance

Abstract:AIM: To detect the expression of HSP70 and MHC-I antigen in human hepatocellular carcinoma and to investigate the relationship between their expression and biologic behavior of the tumor. METHODS: Using the immunohistochemical S-P method, HSP70 and MHC-I antigen expression was detected in liver tissue from 10 normal individuals, 40 patients with HCC and adjacent noncancerous liver tissues. RESULTS: The expression rate of HSP70 was l0% (1/10), 45 % (18/ 40), 80 % (32/ 40) in normal liver, adjacent noncancerous liver tissues, hepatocellular carcinoma, respectively. Significantly higher expression of HSP70 in HCC tissues was observed, compared with adjacent noncancerous liver and normal liver tissues, hepatocelular carcinoma, respectively. Significantly higher expression of HSP70 in HCC tissues was observed, compared with adjacent noncancerous liver and liver tissues (X12 = 10.45, X22 = 14.49, P < 0.05). The expression rate of MHC-I antigen was 100 % (10/ 10), 95 % (38/ 40), 55 % (22/ 40) in normal liver, adjacent noncancerous liver tissues and hepatocellular carcinoma, respectively. A significant lower distribution of MHC-I antigen in HCC tissues was detected than those in adjacent noncancerous liver tissues and normal liver (X2= 5.21, X22= 17.06, P < 0.05). Expression of HSP70 was not associated with tumor lymphocytic infiltration (X2 = 1.4, P > 0.05) and tumor metastasis (X2= 1.9, P > 0.05), but they were associated with histological grading. Positive expression rate of HSP70 was significantly different among well differentiated type (50 %, 5/ 10), moderately differentiated type (78.6 %, 11/]4) and poorly differentiated type (100 %,16/16)of HCC (X2=9.40, P < 0.05). Compared with other cases, differentiation of the tumor was much better in HSP70 + and HLA - A/B/C + cases(X2= 8.13, P < 0.05). CONCLUSION: HSPT70 is overexpressed in human hepatocellular carcinoma. HSP70 can be used as an indicator to judge the differentiation of HCC. The differentiation of tumor is much better in HSP70 + and HLA - A/ B/ C + cases than that in other cases. HSP70 and MHC-I antigen are likely involved in the oncogenesis of HCC.
