Abstract:An efficient micropropagation system has been developed for Cymbidium nanutum Y. S. Wu et S. C. Chen using immature seeds. Seed germination frequency was 45% after 60 days culture and protocorms subsequently formed rhizomes after 90 days culture on half strength Murashige and Skoog (1/2 MS) medium with 0.5 mg l(-1) NAA, 30 g l(-1) sucrose, 1 g l(-1) activated charcoal, and 100 ml l(-1) coconut milk. The highest average proliferation rate was 4.4 and was obtained after 60 days of culture on half strength MS medium containing 2.0 mg l(-1) NAA, 0.5 mg l(-1) BAP, 10 g l(-1) sucrose, and 1 g l(-1) activated charcoal. Approximately I cm long rhizomes produced an average of 3.7 shoot after 50 days of culture on half strength MS differentiation medium supplemented with 0.1 mg l(-1) TDZ, 0.25 mg l(-1) NAA, 20 g l(-1) sucrose, and 1.0 g l(-1) citric acid. Hyponex medium (1.5 g l(-1) Hyponex1 and 1.5 g l(-1) Hyponex2) supplemented with 0.5 mg l(-1) NAA, 30 g l(-1) sucrose, 2.0 g l(-1) activated charcoal, 100 ml l(-1) coconut milk and 100 g l(-1) banana homogenate was best for shoot bud growth and rooting induction (69.7% rooting). The plantlets survival rate was 60% after they were transfered to pots filled with a 1 : 1 : 1 (v : v: v) sand : sieved peat : perlite in greenhouse 30 days after the transferring.
