Separation and Bioinformatics Analysis of One Candidate STK-class Resistance Gene from Cassava

Abstract:Abstact One pair of degenerated primers were designed based the domain I and VIII of the Serine/Threonine (STK) kinase-like proteins. Cassava germplasm E1340 and GR911 were resistant and susceptible to cassava bacterial blight separately, and their genome DNAs were extracted and used as template. The genome DNAs were amplified to produce fragments of about 0.5 kb by PCR reaction. These two products were purified, cloned and sequenced, from which the same sequence of 505nt was obtained. Sequence alignment showed that the cassava germplasm AM560 anchored nucleotide fragment with high homology to this sequence. Gene prediction was made of the sequence of the long fragment about 2.0 kb upstream and downstream from the homogenous fragment, and one gene with four exons and the ORF length of 1866 nt was predicted and named SSK1. The sequence analysis showed that the SSK1 encoded 621 aa. This protein has cell wall receptor kinase binding and coupling sites and eight transmembrane domains, which were typical conserved structures characteristic of STK class resistance gene. SSK1 was a candidate resistance gene and may play an important role in the resistance reaction of cassava to diseases.
