Knockout of mic3 gene in Toxoplasma gondii using CRISPR/Cas9 technique

Abstract:Toxoplasma gondii is an obligate intracellular protozoan,which can infect almost all warm-blooded animals.Successful invasion and infection of host cells by this parasite require proteins secreted from specialized secretory organelles,which include micronemes.Recently,the CRISPR/Cas9 genome-editing technique has become an important tool for molecular biology studies.To better understand the function of micronemal proteins during host infection,the mic3knockout strains were constructed in the type I strain (RH)and type Ⅱ strain(PRU)backgrounds using the CRISPR/Cas9 system.After drug selection and single clone screening,mic3 knockout strains were successfully constructed in both strain types.Then picked RHΔMIC3 for phenotype studies,it showed slightly faster growth in vitro and higher virulence in vivo than the parental strain RH.Thus,this study not only demonstrated that the CRISPR/Cas9 system could be used for genome engineering in any strain of Toxoplasma gondii,but also provided a basis to investigate the biological functions of mic3 in Toxoplasma gondii.
