Cloning and Genetic Analysis of Interleukin-2 Gene in Gushi-chicken

Abstract:A pair of primers of RT-PCR were designed and synthesized according to the published gene sequence of chicken interleukin-2.The target DNA sequence was cloned by RT-PCR depending on the template of total RNA isolated from ConA-stimulated spleen cell.Recombined pGEM-T-GSChIL-2 was transformed into JM109,identified by enzyme digecting and DNA sequencing.Sequence anylasis by comparing with other chicken breeds showed that the cloned seqence had very high similarity.The homologous ratio of DNA sequence is 99.3%,99.1%,98.4%,99.3%,98.6%,99.5%,and that of amino acid sequence is 97.9%,97.9%,95.8%,97.9%,96.5%,98.6%,respectively,compared with Chengren chicken,Indian broiler,Silky,Leghorn,Xianju chicken,and Xiaoshan chicken.
