Isolation of a corynebacterium glutamate capable of degrading of ZEN and AFB1, and investigation of its degrading characteristics

Abstract:The aim of this study was to screen bacterial isolates capable of simultaneously degrading zearalenone (ZEN) and aflatoxin B1 (AFB1) and to preliminarily study the degradation characteristics. Bacterial strains were isolated from soil and chyme of chicken and donkey, and nine strains were found to be able to effectively degrade both ZEN and AFB1. The strain named 406 was identified as a Corynebacterium glutamate by morphological observation and 16SrDNA sequence analysis. The ZEN and AFB1 degrading activity was predominantly attributed to fermentation supernatant of Corynebacteri?um glutamate 406 and the degradation ratio of ZEN and AFB1 was 50% and 58% respectively. The fer?mentation supernatant of 406 was treated with heating, trypsin and sodium dodecyl sulfate (SDS) to de?termine active components, indicating that extracellular enzyme(s) were responsible for the degradation of ZEN and AFB1. The optimal temperature for the maximal degradation of ZEN and AFB1 was 67 ℃and the optimal initial pH was 8. Corynebacterium glutamate 406 capable of simultaneously degrading ZEN and AFB1 might be a promising candidate for ZEN and AFB1 biodegradation in food and feed.
