Effect of salinities on haemocyte of the white shrimp Litopenaeus vannamei

Abstract:To study the effects of salinities on physiological and immune status of Litopenaeus vannamei,this research analyzed the haemocyte indicators of Litopenaeus vannamei cultured in two salinities (5‰ and 20‰) using flow cytometry (FCM).Results showed that THC and non-specific esterase activity of shrimp cultured in 20‰ were 1.35 and 1.90 times higher than those of shrimp cultured in 5‰ respectively.Compared to the shrimp cultured in 5‰,small granular cells and large granular cells were increased,whereas hyaline cells were decreased in the shrimp cultured in 20‰.Plasma phenoloxidase (PO) activity of shrimp cultured in 20‰ were significantly higher than that of shrimp cultured in 5‰.These results indicate that shrimp cultured in 5‰ possess weaker cellular immunity than shrimp cultured in 20‰.Intracellular Ca2+ concentration of shrimp cultured in 20‰ were 1.26 times higher than that of shrimp cultured in 5‰,suggesting that shrimp haemocyte might increase the intracellular Ca2+ concentration to keep normal osmotic pressure between intracellular and extracellular environments under high environmental salinity.Reactive oxygen species(ROS) and NO of shrimp cultured in 5‰ were 1.49 and 1.55 times higher than those of shrimp cultured in 20‰,suggesting that shrimp cultured in low salinity might suffer chronic stress for a long time.There was no significant difference in the rate of apoptosis between the two salinity groups.Shrimp cultured in 5‰ might mobilize more antioxidant activity to eliminate superfluous ROS and NO.This study indicates that the cultured salinity significantly affect the physiological and immune status of shrimp,and the cellular immunity of shrimp cultured in 5‰ was weaker due to the chronic stress of low salinity.
