Abstract:Mongolian pine, as one of the major tree species of the Three-North Shelterbelt Project in the northern semiarid region of China, is very important for ensuring ecological and environmental security. Ca, as an imperative mineral element for plant development and a second messenger, partakes in photosynthesis and affects the growth of plants. However, the optimal calcium concentration for its growth in different soil types is still unclear. In this study, fifteen treatments with combinations of three different soils (e.g., sandy soil, cinnamon soil, dark brown soil) and five calcium concentration gradients (e.g., 0, 100, 200, 400, and 800 mg∙kg -1 ) were conducted to investigate this effect by measuring the indices of growth, biomass, photosynthetic pigment, gas exchange rate, photosynthates, chlorophyll fluorescence parameters and water use efficiency (iWUE) based on a pot experiment. The results showed that the optimal calcium concentration of Mongolian pine seedlings in sandy soil and cinnamon soil was 0–100 mg∙kg -1 , and the optimal calcium concentration of Mongolian pine seedlings in dark brown soil was 100–200 mg∙kg -1 . In other words, the results showed that there was an optimal calcium concentration for the growth of Mongolian pine seedlings, and the optimal calcium concentration was different under different soil types. And if the calcium concentration in the soil was too high, it would have an inhibitory effect on Mongolian pine seedlings, low calcium concentration maybe don't work. The addition of an appropriate amount of exogenous calcium could promote the growth of Mongolian pine seedlings in different soil types. The plant height, basal diameter and biomass of Mongolian pine seedlings all increased significantly after applying an appropriate amount of calcium ( p < 0.05); the addition of an appropriate amount of exogenous calcium could promote the photosynthetic characteristics of Mongolian pine seedlings in different soil types. Under sandy soil and cinnamon soil, the peaks for Pn, Gs, Tr, accumulation of soluble sugar and starch of Mongolian pine seedlings occurred at 0–100 mg∙kg -1 , and the decreasing trend of the Fv/Fm value was significant in the case of exceeding 200 mg∙kg -1 ( p < 0.05), indicating that the growth of Mongolian pine seedlings was affected when calcium concentrations higher than 200 mg∙kg -1 were applied. Moreover, under dark brown soil, the peaks for Pn, Gs, Tr, and accumulation of soluble sugar and starch of Mongolian pine seedlings occurred at 100–200 mg∙kg -1 ; similarly, the growth of Mongolian pine seedlings was affected when calcium concentrations higher than 200 mg∙kg -1 were applied. Compared with the treatment without calcium, after applying an appropriate amount of exogenous calcium, the water use efficiency of Mongolian pine seedlings in different soil types was significantly improved, though it was significantly reduced at 800 mg∙kg -1 ( p < 0.05).

Abstract:In this paper,the effects of 6-BA and NAA combinations of different organs and different concentrations on the rapid culture of Caryopteris clandonensis are lucubrated.The results show that the germination and rootability of the middle section of newly-growing twigs are the highest;the inducing ratio is higher than 85%;MS + 6-BA 0.5 mg/L + NAA 0.5 mg/L is the optimal successive transfer culture medium,and the germination and rootability of the twigs of Caryopteris clandonensis are high.The rootability of stem apex seedlings is high during rooting,and the rooting ratio of stem apex seedlings can be up to 85% in short time.Short but strong seedlings can be cultured in PP333,thus,the survival ratio of the seedlings of Caryopteris clandonensis can be increased.

Abstract:Herbaceous peony (Paeonia lactiflora Pall.) is a renowned traditional Chinese flower, which has been cherished by people since ancient times. It holds significant value as both an agricultural cash crop and an ornamental and medicinal plant. However, the lack of in vitro propagation system has been long hindered the commercial production of this species and the development of new cultivars. In this study, the low chilling requirement cultivar Paeonia lactiflora 'Hang Baishao' that originated from low latitudes of China, was used to study the in vitro culture of embryos. The results of our experiment showed that the optimal medium for in vitro culture of embryos was a 1/2 Murashige and Skoog (MS) medium supplemented with 1.0 mg/L 6-benzyladenine (6-BA), 1.0 mg/L gibberellic acid (GA3) and 2.0 g/L polyvinyl pyrrolidone (PVP). Additionally, histological observations revealed the crucial role of vascular bundle development in the survival of transplants. Plants with well-developed vascular tissue exhibited better transplanting success. This study presents, for the first time, a protocol for embryos propagation and transplanting of 'Hang Baishao', a cultivar with low chilling requirement (CR) during winter. This study provides an efficient method for in vitro culture of 'Hang Baishao', and makes it possible for large-scale production, utilization, and germplasm preservation of this species.

Abstract:A regeneration protocol for cultured endosperm of Actinidia chinensis were set up via organogenesis from callus tissues derived from endosperm explants of Actinidia chinensis ＇ Jintao＇ in vitro. The effects of different media, plant growth regulators, sucrose concentrations and dark treatment on callus induction and shoot formation were studied. The addition of 2,4-D significantly increased percentage of calli formation form the endosperm explants. Addition of 20g/L of sucrose resulted in better organogenesis form, endosperm callus. Dark culture could promote the growth of endosperm callus and organ formation, and the best efficiency was obtained after 7d of culture in the dark.

Abstract:The aim was to study the proper ratio of hormones for seed germination.Lilium speciosum var.gloriosoides widely distributed in Zhejiang Province,was used as matericals.The seeds of induction and germination medium adding different concentration of hormones were studied,and the MS basal medium without any hormones was used as control.The germination rate,shoot induction rate,number of shoots inducted were measured,and the growth of plantlets were observed during the experiment.Meanwhile,the seeds induced form plantlets were analyzed using the conventional chromosome preparation in this paper.The results showed that medium adding hormones could enhance the germination rate significantly,the optimum medium was MS media supplemented with 6-BA 1.0 mg/L and NAA 0.1 mg/L,the germination percentage,induction rate and average indefinite buds were 88.3%,88.9% and 2.5,respectively.Seeds without peeling treatment did not germinate after inoculating 30 days at all.The induced seedlings were transferred to MS medium,the growth condition sustained as before,which illustrated testa was the key factor limiting seed germination and hormone treatment or other physical measures can effectively improve the germination rate.The ploidy levels of seeds induction plantlets were diploid(2n=2x=24),and no genetic variation was found.
Effects of gibberellin A4/7, 6-benzylaminopurine and chlormequat chloride on the number of male and female strobili and immature cones in Chinese pine (Pinus tabuliformis) with foliar sprays

Abstract:Three kinds of plant growth regulators, gibberellinA4/7 (GA4/7), 6-benzylaminopurine (BA), and chlormequat chloride (CCC), were evaluated for their ability to promote strobilus and cone production in a Chinese pine (Pinus tabuliformis Carr.) clonal seed orchard. Treatments (0, 250, 500, or 1000 mg·L−1) were applied during three periods (June–July, July–August and August–September) in 2005. Of the three plant growth regulators, GA4/7 was the best for promoting flower and cone production. Trees sprayed with GA4/7 (500 mg·L−1) from June to September had significantly more female strobili and immature cones than controls and other treatments (p≤0.0001). The best time to apply GA4/7 was in June. BA at 500 mg·L−1 significantly increased female flower and immature cone production, compared to other treatments and controls (p≤0.0001), while promotion of male strobili was unchanged. Chlormequat chloride at 1000 mg·L−1 significantly increased the number of male strobili, compared to the other treatments (p≤0.0001). Spraying CCC at 500 mg·L−1 significantly increased the number of female strobili, compared to controls (p<0.05), but it had no effect on immature cone production (p>0.05). These results are important for improving seed production and seed orchard management of Chinese pine.

Abstract:Juglans mandshurica is an important component of Korean pine mixed and secondary forests in Northeast China and is also a precious timber and economic species. In this study, plant regeneration of J. mandshurica has been established using stems with bud and mature zygotic embryos as explants. The results showed that placing the stems with buds in low-temperature conditions (0 to 4°C) and in darkness for 5 d inhibited the browning of the explants and promoted bud sprouting; Na 2 S 2 O 3 also inhibited the browning of mature zygotic embryos significantly. The suitable basal media and hormones for bud sprouting and zygotic embryo germination were (1) WPM medium (woody plant medium) containing 2.0 mg·L −1 6-benzylaminopurine (6-BA) and 0.01 mg·L −1 3-indolebutyric acid (IBA) and (2) DKW medium (Driver and Kuniyuki medium) containing 2.5 mg·L −1 6-BA and 0.1 mg·L −1 IBA. The suitable basal medium and hormone for rooting of shoots from buds was 1/2 Murashige and Skoog (MS) medium (the macro-elements were half-strength of MS medium) supplemented with 1.0 mg·L −1 IBA, and the suitable basal medium and hormone for rooting of seedlings derived from zygotic embryo was 1/2 MS medium containing 1.0 mg·L −1 IBA, and DKW medium with 2.5 mg·L −1 6-BA, 0.1 mg·L −1 IBA, 0.2 mg·L −1 gibberellin A3 (GA 3 ), and 0.2 g·L −1 activated carbon (AC). About 95% of the rooted plantlets survived after acclimation and transplantation. These results provided a foundation for the scale propagation to meet the demands of forestry production of the species.

Abstract:To solve the problems of weak growth and low rooting potential of long period subcultured Dendrobium nobile protocorm-like bodies(PLBs) in vitro,the effects of organic additives,plant growth regulators,sucrose and heights of the shoots on the growth,proliferation and rooting of the PLBs were systematically studied.The results indicate that 0.5 mg/L BA was suitable for the growth and proliferation of the PLBs,and 20 g/L sucrose suitable for rooting.Although IBA stimulated rooting of the shoots,the height of shoot was crucial.On the basal medium supplemented with 0.05 mg/L IBA,the rooting percentage of the 2.5 cm high shoots reached 51.11%.F/P rate(increasing rate of fresh weight of PLBs/increasing rate of proliferation of PLBs) is a key parameter to distinguish whether the growth of the PLBs is normal or not.When F/P≥0.6,the PLBs grew well.

Abstract:Object To optimize the fitting culture media for multiplication of adventitious bud and ra-dication of Dendrobium candidum Wall. ex Lindl. and the length of adventitious bud for succesive transfer multiplication. Methods In the same culture condition, the adventitious bud with different length developed in the various culture media for multiplication and radication. Facing the different growth status, the diversity test and general analysis were carried on for them. Results There were significant difference in different length of adventitious bud, difference between the multiplication and radication of culture media. Conclusion The adventitious buds in (1.2±0.1) cm length are superior for succesive transfer multiplication, MS adding BA 1.0 mg/L, NAA 0.2 mg/L is the best media for the adventitious buds multiplication, 1/2 MS adding NAA 0.2 mg/L is the test media for radication.
Effect of Plant Growth Regulators, Temperature and Sucrose on Shoot Proliferation from the Stem Disc of Chinese Jiaotou ( Allium Chinense ) and in Vitro Bulblet Formation

Abstract:In vitro shoot proliferation from stem disc of Allium chinense, a vegetatively propagated plant, was investigated in this experiment. In the present study, shoots were formed directly on stem discs on a medium containing 1 mg/l N6-benzyladenine (BA) and 0.5 mg/lα-naphthaleneacetic acid (NAA). These shoots were further cultured on MS media supplemented with various levels of BA in combination with NAA, and new shoot clusters developed easily from the explants cultured despite considerable differences in the induction of shoot clusters with different levels of BA and NAA. The most productive combination of growth regulators proved to be 1.0 mg/l BA and 1.0 mg/l NAA, in which about 17 shoots were produced per cluster in 8 weeks culture. Most of the formed shoots were rooted 15 days after being cultured on MS media supplemented with 0.1–1.0 mg/l NAA. The survival rate of the plantlets under ex vitro conditions was 95% in pots filled with a peat: sand (2:1 v/v) mixture after two weeks. In vitro bulblet formation were strongly promoted by the high temperature of 30°C compared to that at 25, 20 and 15°C, and 12% (w/v) sucrose appeared to be optimal for bulblet development. Results from this study demonstrated that A. chinense could be in vitro propagated by using stem discs and in vitro bulblet formation could be achieved.

Abstract:The aim of this study was to establish an efficient protocol for androgenetic embryoid induction in asparagus anther culture.Cultivar THIELIM F_1 was used in the experiment with MS as the basal medium.Orthogonal arrays of L_(16)(4~5) were adopted to investigate the effect of NAA,6BA,2,4-D,sucrose,glutamine on response of embryoid induction.The results showed that NAA was the most effective element for embryoid induction in asparagus anther culture,followed by 6-BA and sucrose with the least effective for 2,4D and glutamine.The optimized combination was derived with 0.01 mg/LNAA,2.0 mg/L 6-BA,1.0 mg/L 2,4-D,5 g/L sucrose and 0 mg/L glutamine.
Enhanced Root and Stem Growth and Physiological Changes in Pinus bungeana Zucc. Seedlings by Microbial Inoculant Application

Abstract:Background and Objectives: As an extensively used tree species in landscaping and afforestation in China, lacebark pine (Pinus bungeana Zucc.) seedlings are in high demand. However, the small number of fine roots and the low growth rate of lacebark pine seedlings increase the risks encountered during transplant and extend the nursery time for outplanting. We aimed to find out whether a microbial inoculant would promote root growth and accordingly, shorten the nursery cultivation time. Materials and Methods: One-year-old lacebark pine seedlings were treated with the inoculant Bacillus subtilis 8–32 six times from June to September. At each application time, five treatments of undiluted microbial inoculants (UM), 30 times diluted microbial inoculants (30 DM), 40 times diluted microbial inoculants (40 DM), 50 times diluted microbial inoculants (50 DM), and distilled water as a control (CTRL) were administered to the seedlings. In the end, all the seedlings were harvested to measure the root growth, aboveground growth, and the physiological indices. Results: Root and stem growth was enhanced by the inoculants in terms of the increased number of root tips, the length and surface area of the roots, the biomass of the roots and stems, as well as the increase in height and basal stem diameter. The chlorophyll a/b of the needles was increased, in spite of the fact that the total chlorophyll content was decreased by the microbial inoculant treatments at the end of the growth phase. Meanwhile, the maximum photochemical efficiency (Fv/Fm) of the needles was increased by the inoculant treatments. The soluble sugar content was additionally translocated into the stems in the UM treatment, suggesting the change in carbon allocation. The content of available potassium, phosphorus, and ammonium nitrogen in the potting soil was increased in the 30 DM group, and the content of soil organic matter was increased in all the inoculant treatments. Conclusions: The microbial inoculant Bacillus subtilis 8–32, in appropriate concentrations, could be applied to promote root and shoot growth and improve the seedling quality of the lacebark pine during cultivation.

Inducing and Proliferating Culture of Adventitious Buds from Catalpa Bungei

Abstract:In this experiment,sterile tender stems of Catalpa bungei were selected as explants to explore induction,proliferation and rooting of adventitious buds by applying different kinds of auxin(NAA,IAA,IBA,2,4-D)and concentrations(0.05,0.1,0.2 mg/L).The results showed that(1)The optimum induction and growth were obtained by applying IAA;(2)by applying IAA(0.1 mg/L)combined with 6-BA(0.8 mg/L),C.bungei not only showed a 100% induction rate but also robust growth;(3)After transferring to the proliferation medium,the adventitious buds cultured in IAA(0.1 mg/L)combined with 6-BA(0.8 mg/L)medium showed the highest proliferation ratio which was 3.23 and robust growth;(4)adding of IAA(0.1 mg/L)was good for rooting of adventitious buds.
[Study on PLB induction and proliferation of Dendrobium officinale]

Abstract:Objective: To study on the optimal medium ingredients for PLB induction and proliferation of Dendrobium officinale. Methods: Seed embryos of Dendrobium officinale were cultivated in MS medium as the basic medium, along with different plant hormones like 6-BA, NAA,2,4-D and KT or their combinations added with organic additives like PE, BE, AE and CM. Results: BA and NAA combination was not conductive to germination and the germination ratio was even lower than that of MS medium; 10% PE and CM was beneficial to PLB induction; 2,4-D was not conductive to growth and proliferation; A certain concentration of BA, KT and NAA was beneficial to PLB proliferation; KT at 1.0 mg/L recorded the highest 40 d PLB proliferation times at 9.0; PE, CM and AE could promote the PLB proliferation at different levels, among which 10% CM was the most effective. Conclusion: The optimized medium ingredients suitable for PLB induction are MS +10% CM +1.0g/L AC; The optimized medium ingredients suitable for PLB proliferation are MS + 1.0 mg/L KT + 0.2 mg/L NAA + 10% CM.
Pretreated Eucalyptus globulus and Pinus radiata Barks: Potential Substrates to Improve Seed Germination for a Sustainable Horticulture

Abstract:Commercial forest plantations in Chile are dominated by pine (Pinus radiata) and eucalyptus (Eucalyptus globulus). Tree bark is the main by-product of the forestry industry and has low value, but great potential for use as an agricultural substrate. However, the direct use of bark fibers may cause plant phytotoxicity due to the presence of polyphenolic and other compounds. This study aims to evaluate the physicochemical properties of E. globulus and P. radiata bark after water extraction treatments. The phytotoxicity of the resulting extracted bark alone and that mixed with commercial substrates (coconut fiber, moss, peat, and composted pine) at different ratios (25 to 75 wt%) were assessed using the Munoo-Liisa vitality index (MLVI) test. For all treatments, the seed germination and growth of radish (Raphanus sativus) and Chinese cabbage (Brassica rapa) species were evaluated and compared to a commercial growing medium (peat) as a control. The optimal mixture for seed growth was determined to be 75% extracted E. globulus bark fiber and 25% commercial substrates such as peat (P), coconut fiber (C), moss (M), and composted pine (CP), as indicated by the MLVI and germination results. Two phytostimulant products, chitosan and alginate-encapsulated fulvic acid, were added to the best substrate mixture, with the purpose of improving their performance. Encapsulated fulvic acid at 0.1% w/v was effective in promoting plant growth, while chitosan at all of the concentrations studied was effective only for mixture 75E-25CP. The mixture of E. globulus fiber and commercial substrates, containing a high proportion of water-extracted fiber (75%), shows the potential to be used in the growth of horticultural crops and in the plant nursery industry.

Substrate selection for container seedling breeding of Pinus flexilis.

Abstract:A container seedling breeding experiment was carried out with seeds of four Pinus flexilis provenances introduced from North America by four types of mixed substrate with different proportions(surface soil substrate was used as control).The most suitable substrate selected was B2(30% sand,50% peat,20% vermiculite) through measuring eight growth indexes and four physiological indexes of the one-year-old container bred seedlings of the four provenances.The correlations between the physical and chemical characters of the different substrates and the morphological and physiological indexes showed that the physiological indexes of the substrates were the most important factors influencing the growth and development of the one-year-old Pinus flexilis container bred seedlings.

Moench [Hibiscus esculentus L.] plants were studied,the stem segments with one leaf and one axillary bud of seedlings derived from sterile germination were used as explants.The results showed that when the explants were inoculated on 1/2MS basic medium,in a certain concentration range,the increase of IAA concentrations significantly promoted the ratio of differentiation and proliferation,and the presence of 6-BA could increase the role of IAA treatments,but not obviously,so the optimal combination was IAA 1.0 mg/L+6-BA 2.0 mg/L.After the buds grew to more than 1 cm,the single buds were cut out to inoculate on 1/4MS basic medium adding different NAA concentrations,and the optimal rooting rate and growth performance was found in the treatment of NAA 0.1 mg/L.
An Advanced Protocol for the Establishment of Plantlets Originating from Somatic Embryos in Pinus Massoniana.

Abstract:This study describes critical factors affecting germination of somatic embryos and plantlet regeneration in Pinus massoniana. Somatic embryos from the same embryogenic line 27 of P. massoniana were used as test materials. The supplementation of activated charcoal (AC) in the medium was essential for the germination of mature somatic embryos, while the addition of excessive AC to the medium was prohibitive for somatic embryo germination. The highest germination rate was found on the medium containing 10 g/l AC, and the addition of 5 g/l AC to the medium was optimal to the growth of germinating somatic embryos. Thidiazuron (TDZ) was linearly related to the number of sprouting axillary buds. However, the growth of sprouting buds was retarded when > 4 µmol/l TDZ was added into culture medium. Exogenous plant growth regulators added to the medium significantly improved the root regeneration capacity of shoots. The highest root regeneration rate was observed under the treatment of 1.2 µmol/l ɑ-naphthaleneacetic acid (NAA) plus 2 µmol/l paclobutrazol (PBZ), reaching 96.3%. One year after the field transfer, the growth performance of plant height, caliper, and survival rate for rooted shoots was significantly better than that of plantlets directly developed via somatic embryogenesis. The presented results provide useful instruction for the establishment of plantlets originating from somatic embryos, and would be able to make a great contribution to the clonal forestry of P. massoniana.

Abstract:Liriope spicata is an important traditional Chinese medicine and is also used as an ornamental groundcover plant. A tissue culture method, such as somatic embryo regeneration, that can generate large numbers of genetically identical plants would help maintain the characteristics of specific varieties. This study aimed to screen and optimize plant growth regulators for all aspects of somatic embryo regeneration in L. spicata . The plant growth regulators 2,4-dichlorophenoxyacetic acid (2,4-D), 6-benzylaminopurine (6-BA), naphthalene acetic acid (NAA), gibberellic acid (GA 3 ), indole-3-acetic acid (IAA), and kinetin (KT) were screened in tissue culture media. We found that 2 mg/L 2,4-D + 2 mg/L 6-BA + 0.1 mg/L NAA had the best induction efficiency of 30.95%. During the screening of the somatic embryo proliferation media, we found that MS + 1 mg/L 6-BA + 0.1 mg/L NAA + 20 g/L glucose + 7 g/L agar, with a pH of 5.8, had the best proliferation efficiency of 86.67%. For the germination of somatic embryos, we found that MS + 0.1 mg/L 6-BA + 0.2 mg/L NAA + 1 mg/L GA 3 had the best effect, with an efficiency of 29.03%. Indole-3-butyric acid (IBA) had a significant effect on the induction of adventitious shoots, with the formula 1/2 MS + 0.1 mg/L IBA resulting in an efficiency of 100%. The establishment of the regeneration pathway for L. spicata somatic embryos not only ensures the industrial production of species but also provides reference for the application of plant growth regulators in plant tissue culture.

Abstract:The propagation of adult pine trees by tissue culture has been studied for a few decades but problems related to the juvenile to adult phase change of trees have limited the practical applications of these tissue culture procedures. This study describes a micropropagation protocol for the in vitro propagation of mature Pinus massoniana trees. In this study, nodal segments were used as the explants for in vitro regeneration of adult P. massoniana trees (25-30 years old). The sampled explants were cultured on four media (DCR, SH, GD and modified MS (MMS)) supplemented with three cytokinins (BA, KT and ZT) at three different concentrations (5, 10 and 20 mu M). The evaluated culture media and cytokinin treatments showed significant differences in organogenic responses. The highest organogenic response was obtained with nodal segments cultured on MMS media and by explants cultured on medium supplemented with 10 mu M zeatin (ZT). A successful and efficient protocol is first developed for the micropropagation of adult P. massoniana trees by biotechnological approach.
