Effects of ALA on Mice TM3 Leydig Cell Viability and Relative Gene Expression

Abstract:As one of the essential fatty acids, α-linolenic acid (ALA) plays an important role in the growth and development of the body. In the male reproductive system, ALA can convert into docosahexaenoic acid and eicosapentaenoic acid, and contribute to male fertility. The present study was undertaken to elucidate direct ALA effects on mice TM3 Leydig cell viability and gene expression using WST-1, JC-1 and real time PCR techniques. The results showed that the addition of different concentrations of ALA (10, 50, 100, 200 and 400 μmol/L) in the culture system for 24 h significantly up-regulated TM3 cell viability in a dose-dependent manner (P<0.05). Meanwhile, the mitochondrial membrane potential was remarkably boosted after being treated with 50 μmol/L ALA for 24 h. ALA also inhibited apoptosis related gene Caspase-3 expression (P<0.05) and stimulated steroid metabolism related gene StAR expression (P<0.05). To conclude, the present study demonstrated that ALA increased mice TM3 Leydig cell viability and stimulated proliferation and StAR expression, however, inhibited Caspase-3 expression.
