Abstract:Ophiostomatoid fungi mainly refer to the order Ophiostomatales (Ascomycota), which are known to be symbiotic with bark beetles infesting pine trees. These fungi are primarily associated with Dendroctonus armandi and other bark beetles, significantly impacting the local pines Pinus armandii in western China. However, this fungal diversity must be further explored. This study aims to identify Ophiostomatoid fungi associated with P. armandii in Ningshaan, Shaanxi Province. A total of 225 Ophiostomatalean isolates were obtained from 46 gallery samples, accounting for 77.6 % of the total number of isolated strains. Twenty species were identified based on the phylogenetic analyses of ITS, LSU, β-tubulin, and EF-1α, as well as morphological characteristics. These include seven new taxa, , , , , , , , and thirteen known species. This study laid a theoretical foundation for understanding the relationship between the beetles and the ophiostomatalean fungi in the Ningshan region, China.

Abstract:Climate change has exacerbated outbreaks of forest pests worldwide. In recent years, bark beetles have caused significant damage to coniferous forests of the Northern Hemisphere. Polygraphus bark beetles are widely distributed secondary pests. Recently, tree mortality caused by these beetles on the Qinghai-Tibet Plateau has been increasing; however, few studies have focused on their fungal associations. In the present study, we explored the diversity of ophiostomatalean fungi associated with these beetles on the north-eastern and southern Qinghai-Tibet Plateau. We isolated 442 ophiostomatalean strains from adult beetles and their fresh galleries, specifically targeting Polygraphus poligraphus and Polygraphus rudis infesting Picea crassifolia and/or Pinus griffithii . Based on phylogenetic and morphological features, we assigned the 442 strains to 16 species belonging to Grosmannia spp., Leptographium spp. and Ophiostoma spp. Amongst these, Ophiostoma maixiuense and Ophiostoma bicolor were the most frequently isolated species, accounting for 20.8% and 18.1% of the total number of ophiostomatalean assemblages, respectively. By comparing their fungal communities, we found that the different patterns of fungal assemblages of bark beetles from the north-eastern and southern Qinghai-Tibet Plateau may be influenced by biogeographic barriers and host tree species. The results of this study enhance our understanding of bark beetle fungal assemblages, especially Polygraphus , on the Qinghai–Tibet Plateau, with implications for forest management under changing climate.

Abstract:Leptoporus is a rare and remarkable genus, mainly occurring in coniferous forests in the Northern Hemisphere. Recent phylogenetic studies showed that Leptoporus belongs to Irpicaceae in the phlebioid clade. It is worth noting that most species in the phlebioid clade can cause white-rot decay, except for the Leptoporus species, which can cause a brown-rot decay. In this study, we performed phylogenetic and taxonomic studies of Leptoporus and related genera. Molecular phylogenetic analyses were conducted based on sequences from multiple loci including the internal transcribed spacer (ITS) regions, the large subunit of nuclear ribosomal RNA gene (nLSU), the largest subunit of RNA polymerase II gene ( RPB1 ), the second largest subunit of RNA polymerase II gene ( RPB2 ), and the translation elongation factor 1-α gene ( TEF1 ). Combined with morphological characteristics, a new species, Leptoporus submollis sp. nov., is discovered and illustrated from Southwest China.

Abstract:Fungi of Ophiostomatales (Sordariomycetes, Ascomycota) always have a symbiotic relationship with bark beetles (Coleoptera: Scolytinae). In this study, we investigated the fungi associated with and in Fushun. Based on the four gene fragments of TUB, ITS, LSU, and TEF and morphological characteristics, 206 strains of Ophiostomatales were identified to be distributed in 21 taxa; these included an undescribed taxon. Here, we described it as sp. nov. The results of this study provided valuable information on the relationship between fungi and bark beetles.

Abstract:Ophiostomatalean fungi refers to the species in the order Ophiostomatales of Ascomycota. This group of fungi comprises wood blue-stain agents and some species that are important pathogens of trees, nematodes, or humans. Most species in the Ophiostomatales have beneficial relationships with wood-boring insects such as bark beetles. Recent surveys were conducted to investigate the diversity of Ophiostomatoid fungi associated with Cyrtogenius luteus infesting Pinus massoniana in some areas of Guangdong, China. This study, 382 fungal strains were obtained, and 224 were identified as belonging to the Ophiostomatales order. Identification of these strains revealed that they spanned seven distinct genera. A new species, sp. nov., was discovered and identified through a combination of morphological and phylogenetic analyses across four gene regions: beta-tubulin (TUBB), internal transcribed spacer (ITS), large subunit ribosomal RNA (LSU), and translation elongation factor 1 alpha (EF1A).

Abstract:is a genus of fungi that falls under the order Microascales of Ascomycota. Some species in this genus can establish a unique symbiotic relationship with the pine-infesting bark beetles, while others are typically found in wood or soil habitats. To comprehensively investigate the diversity of species of these fungi, recent field trips were conducted in seven provinces (Fujian, Guangdong, Guizhou, Guangxi, Liaoning, Shaanxi, and Shandong) in China. 96 pure isolates of were obtained by sequences from 361 samples. Nineteen representative strains were carefully selected to generate sequencing data from four gene regions (ITS, LSU, EF1A and TUBB), then used to construct phylogenetic trees for the genus. The results revealed the discovery of two new species, namely sp. nov. and sp. nov., and was the most common species in various pine hosts.

Abstract:As a result of an ongoing survey of microfungi associated with garden and ornamental plants in Qijing, Yunnan, China, several saprobic fungal taxa were isolated from Magnolia grandiflora . Both morphological and combined SSU, LSU, ITS, tef1 , and rpb2 locus phylogenetic analyses (maximum-likelihood and Bayesian analyses) were carried out to identify the fungal taxa. Three new species are introduced in Pleosporales, viz ., Lonicericola qujingensis ( Parabambusicolaceae ), Phragmocamarosporium magnoliae , and Periacma qujingensis ( Lentitheciaceae ). Botryosphaeria dothidea, Diplodia mutila , and Diplodia seriata (in Botryosphaeriaceae ) are reported from Magnolia grandiflora for the first time in China. Angustimassarina populi ( Amorosiaceae ) is reported for the first time on M. grandiflora from China, and this is the first report of a member of this genus outside Europe. Shearia formosa is also reported for the first time on M. grandiflora from China.

Diaporthe Sinensis, a New Fungus from Amaranthus Sp. in China

Abstract:During an exploration of plant pathogens in vegetables occuring in Zhejiang province, China, a novel fungal species, was found. Three strains ZJUP0033-4, ZJUP0038-3 and ZJUP0132 were isolated from black round lesions in the stems and leaves of Amaranthus sp. Phylogenetic analyses based on sequences from four genes including rDNA internal transcribed spacer (ITS), translation elongation factor 1-α (EF1-α), histone (HIS) and β-tubulin (TUB) indicated that D. sinensis clustered in a distinct clade closely related to D. neoarctii, D. angelicae, D. subordinaria, D. arctii, D. cuppatea, D. lusitanicae, D. novem, D. infecunda, D. ganjae and D. manihotia. Morphologically, D. sinensis is distinguished by brown, scattered, globose pycnidia and ellipsoid alpha conidia with bi- to multiguttulate.

Abstract:Six species of the Leucophenga argentata species group from East Asia are examined (including one known and five new ones): L. argentata (de Meijere, 1914); L. bicuspidata sp. nov.; L. fuscivena sp. nov.; L. longipenis sp. nov.; L. quadricuspidata sp. nov.; L. tricuspidata sp. nov.; the diagnoses for Asian species of the argentata group and a key to these species are provided. A total of 44 DNA sequences of the mitochondrial COI (cytochrome c oxidase subunit I) gene with BOLD process ID and GenBank accession numbers are provided for these species. The intra- and interspecific pairwise p-distances are summarized. The NJ (Neighbor-joining) and the Bayesian analyses are used to conduct a molecular phylogenetic analysis for the above-mentioned species. The molecular data are used as complementary evidence for the identification of the argentata group species with sexually dimorphic abdominal tergites.
New and Interesting Pine-Associated Hyphomycetes from China

Abstract:Pine trees play a crucial role in the forests of Sichuan Province, boasting rich species diversity and a lengthy evolutionary history. However, research and investigation on fungi associated with pine trees are insufficient. This study investigated the diversity of hyphomycetes fungi associated with pine trees in Sichuan Province, China. During the survey, we collected five specimens of hyphomycetes from branches and bark of species of Pinus. Five barcodes were selected for study and sequenced, including ITS, SSU, LSU, TEF1, and RPB2. Morphological examination and multi-locus phylogenetic analyses revealed three new species, viz. Catenulostroma pini sp. nov. within Teratosphaeriaceae, Kirschsteiniothelia longisporum sp. nov. within Kirschsteiniotheliaceae, Sporidesmiella sichuanensis sp. nov. within Junewangiaceae, and two known species, Paradictyoarthrinium diffractum and P. hydei within Paradictyoarthriniaceae, which are the new host records from Pinus species. Catenulostroma pini, distinguished from other species in the genus by its unique morphology, has three conidial morphologies: small terminal helicoconidia, scolecoconidia with many septa, and phragmoconidia conidia. Kirschsteiniothelia longisporum has longer spores when compared to the other species in the genus. According to phylogenetic analysis, Sporidesmiella sichuanensis formed an independent clade sister to S. aquatica and S. juncicola, distinguished by differences in conidial size.

Abstract:The taxonomy of Taxus Linn. remains controversial due to its continuous phenotypic variation and unstable topology, thus adversely affecting the formulation of scientific conservation strategies for this genus. Recently, a new ecotype, known as Qinling type , is mainly distributed in the Qinling Mountains and belongs to a monophyletic group. Here, we employed multiple methods including leaf phenotype comparison (leaf shapes and microstructure), DNA barcoding identification (ITS + trn L -trn F + rbc L), and niche analysis to ascertain the taxonomic status of the Qinling type .

Abstract:Two fungal strains were isolated from submerged decaying woods in the freshwater habitats, which are characterized by cylindrical, septate, verrucose, brown conidia, composed of four vertical columns of cells, 3-septate in each column, with four appendages at the base which are slightly tapering toward the apex and each with 5–10-septate. Multigene phylogenetic analyses of combined ITS, LSU, and SSU sequences data showed that the two isolates clustered into a separated clade in Tetraploa. Herein, we report it as a new hyphomycetous species, Tetraploa lignicola. The descriptions, illustrations, and a phylogenetic tree show the placement of T. lignicola, the comparisons between the new species and other morphologically similar taxa in Tetraploa are also provided.

Abstract:During a survey of the diversity of lignicolous fungi in Yunnan Province, China, we collected and identified five microfungi species from dead woody litters of Castanopsis trees in terrestrial habitats. Through both morphological comparisons and phylogenetic analyses of multi-gene sequences, we identified two taxa as new species and three collections as new host records within Pleosporales . Pseudolophiostoma lincangense sp. nov. is introduced as a sexual morph in Lophiostomataceae , Pleopunctum baoshanense sp. nov. is introduced as a hyphomycetous fungi in Phaeoseptaceae , and Paraphoma aquatica as a first report of sexual morph in Paraphoma . In addition, Occultibambusa kunmingensis and Pleopunctum megalosporum were isolated for the first time from the dead twigs of Castanopsis delavayi and C. calathiformis , respectively. Comprehensive morphological descriptions, illustrations, and phylogenetic analysis results are provided for the above-mentioned species.

Abstract:China has a complex and diverse forest ecological environment, which breeds abundant forest macrofungi, including some edible, medicinal, and poisonous species. During the investigations of macrofungi in the Saihanba National Nature Reserve, North China, we collected abundant specimens of Agaricales and Polyporales within the Agaricomycetes. Based on the morphological characters and molecular evidence of DNA sequences including the internal transcribed spacer (ITS) regions, the large subunit of nuclear ribosomal RNA gene (nLSU), the small subunit of mitochondrial rRNA gene (mtSSU), the small subunit of nuclear ribosomal RNA gene (nuSSU), the largest subunit of RNA polymerase II (RPB1), the second largest subunit of RNA polymerase II gene (RPB2), the beta-tubulin gene (TUB), and the translation elongation factor 1-alpha gene (TEF1), this study identifies ten species of Agaricales and Polyporales new to science, viz. Cyanosporus subpopuli, Gelatinofungus betulina, Lycoperdon pseudoperlatum, Macrocystidia hebeiensis, Mycena subbrunnea, M. subpura, M. variispora, M. violocea-ardesiaca, Picipes griseus, and Pleuroflammula hebeiensis. Detailed morphological descriptions, fruiting bodies, and microscopic structure diagrams of these ten novel species are provided.
Multigene Phylogeny and Morphology of Ophiocordyceps alboperitheciata sp. nov., A New Entomopathogenic Fungus Attacking Lepidopteran Larva from Yunnan, China

Abstract:A new fungus, Ophiocordyceps alboperitheciata, parasitic on the larva of Noctuidae (Lepidoptera) was identified from a survey of entomopathogenic fungi in Kunming Wild Duck Forest Park, Yunnan Province, China. It can be primarily distinguished from relatives by its longer fertile parts, sterile tips, superficial perithecia, narrower asci, and smaller septa of ascospores. As revealed from phylogenetic analyses inferred from nrSSU, nrLSU, tef-1α, rpb1, and rpb2 sequence data, O. alboperitheciata belongs to the Hirsutella citriformis clade in the genus Ophiocordyceps of Ophiocordycipitaceae, and forms a separated clade from other related species. The uniqueness of the taxon is significantly evidenced by both molecular phylogeny and morphology. Furthermore, the interspecific relationships in the H. citriformis clade are discussed.

Abstract:We describe a new species of forest bark beetle, Tomicus armandii Li & Zhang, collected from Pinus armandii in Yunnan, China. We used the D2 fragment of 28S rDNA to improve the taxonomy of Tomicus. The new species can easily be distinguished from the other Tomicus species using the following two morphological characters: punctures of interstria 2 on declivity appearing evenly biseriate or triseriate; erect interstrial setae on the declivity short, about 0.5× as long as distance between striae. The genetic distances of 28S rDNA measured between T. armandii and other species of Tomicus are similar to the distances between other Tomicus species, and these are much higher than intraspecific distances. The phylogenetic analysis of 28S rDNA agrees with the groupings obtained from morphological identification. DNA analysis has commonly been used in Tomicus taxonomy and is helpful for resolving taxon identification problems.

Zoosystematics and Evolution

Abstract:Four morphologically similar species of the Pholcus yichengicus species group, occurring in geographic proximity of China's Qinling Mountains, were recognised, based on morphology and four methods of molecular species delimitation. They comprise two new species, namely Pholcus ankang sp. nov. and P. baoji sp. nov. and two previously described species: P. ovatus Yao & Li, 2012 and P. taibaiensis Wang & Zhu, 1992. Their DNA barcodes were obtained to estimate p-distances and K2P distances. In addition, an identification key for the four closely-related species is presented.
