Effect of cholesterol on infectious bronchitis virus infection of CEKs

Abstract:Cholesterol is an important component to maintain the stable structure of lipid rafts on cellular membrane.For some enveloped viruses,the infection of host cell depends on the participation of cholesterol.In this paper,pretreatment of chicken embryo kidney cells (CEKs) with methyl-β-cyclodextrin (MβCD),a drug used to deplete cholesterol from membranous structure,to analyze whether the cellular or viral membrane cholesterol played a role in chicken infectious bronchitis virus (IBV) infection of CEKs.The results showed that significantly decrease of IBV infection in a dose-dependent manner through real time RT-PCR and virus titer test,and IBV infection could be partially restored by supplementing exogenous cholesterol,indicated that IBV infection depended on cellular membrane cholesterol.Further studies discovered that the removal of cellular membrane cholesterol affected IBV attachment and release stage.However,the removal of viral envelope cholesterol had no effect on IBV infection.These results demonstrated that IBV infection of CEKs was dependent on cellular membrane and no viral envelope cholesterol.
