Abstract:During 2007 to 2009,an extensive survey and morphological identification on root-knot nematodes,Meloidogyne spp.,occurring in vegetable crops of Zhejiang,China,were carried out.Based on diagnostic characters and morphometrics of females,males,and second-stage juveniles,especially the perineal patterns of female,two species were identified,i.e.M.incognita and M.javanica,in which M.incognita was the preponderant species,widely distributed in vegetable crops of Zhejiang,whereas M.javanica was only found in Wenzhou of Zhejiang,and this was the first record of the species in Zhejiang.

Abstract:In recent years, with the rapid development of installation cultivation of vegetable, many questions which constraint its development appeared. Among them, nematode disease is one of the most important constraint factors, one of which the harm caused by the root-knot nematodes (Meloidogyne spp.) is the most serious. Root-knot nematode is internal parasites settlement in plant roots, which has become one of most governing pathogen threaten to agricultural for its widespread and more parasites. In the past, chemicals were used to control this disease, combined with agricultural control and the use of resistant varieties, but has its limitations. With the advancement of technology and enhanced awareness of environmental protection, biological control has aroused a great attention. This paper reviews the advances of various biological control factors, and important factors influenced the efficiencies of biological control to nematode were analyzed thoroughly, in the last, the application prospect of the biological control were reviewed.

Abstract:The rice root-knot nematode Meloidogyne graminicola is a severe pest of rice. In China, it was first reported from Hainan Province, and later from several other provinces. In the present study, a rice root-knot nematode population found from the rice cultivation areas of Zhejiang Province, China is characterized via molecular analysis using internal transcribed spacer (ITS) and cytochrome c oxidase subunit II (coxII)-16S rRNA genes and scanning electron microscopy (SEM) observations of males and the second-stage juveniles. Morphometric data and molecular sequence comparisons for all M. graminicola populations occurring in China are also provided. The overall morphology of M. graminicola found in Zhejiang match well with the original description, though males have a slightly longer body and stylet, and a shorter tail, while the second-stage juvenile is also slightly longer than in the original description. This is the first report of M. graminicola from Zhejiang. Phylogenetic studies based on coxII suggest that all the Chinese populations belong to Type B. This study expands knowledge of the increasing distribution and phylogenetic relationships of M. graminicola that occur in China.

Abstract:Stunt nematodes belong to an important group that feeds on the roots of many types of crops worldwide. They include Tylenchorhynchus spp. and Merlinius spp., which although, at present are classified as minor pests, but are a potential biosecurity threats to global agriculture production and food security. Molecular and morphological studies of Tylenchorhynchus thermophilus, T. annulatus and Merlinius brevidens were conducted in this study with the main aim of accurate identification and classification of the pest. This is necessary in planning of protective and preventive measures to reduce the risk of transmission of these nematodes. The phylogenetic relationships were analyzed using ITS 1 and 18S genes of ribosomal DNA. These analyses indicated that sequence identity ranged from 97.1% to 99.8% among six Merlinius populations and 91.4% to 98.9% between two Tylenchorhynchus populations. The results of this study are significant in biosecurity measures at plant quarantine stations at sea and airports and also useful to scientists, extension agencies and farmers involved in nematode research and control. Abstract:According to the morphological characters and morphometrics,three species in genus Hirschmanniella were identified.They were H.oryzae,H.mucronata and H.gracilis.Some morphometrics of H.oryzae and H.mucronata varied slightly from the original descriptions of type specimens,most of other morphometric data were within the reference value range.Compared to the original descriptions of type specimen,the data of H.gracilis were smaller.This was the first report of H.gracilis in Zhejiang Province,eastern China.

Abstract:The species of the genus Meloidogyne are globally distributed and generally induce the formation of galls or nodules in the roots of a wide variety of plants, due to this; this nematode is known as "Root knot nematode". The affected plants show loss of turgidity of the leaves, chlorosis and the withering, which leads to a reduction in crop yields. In Ecuador, the presence of several species of the root knot nematode has been registered, in wild and cultivated plants. For the diagnosis, the identification of this plant parasitic nematode, initially was by the use of morphological characters and later was incorporated the use of the analysis of the isoenzyme phenotypes in polyacrylamide gel. However, molecular techniques, which include the analysis of partial or total sequences of one or more genes of taxonomic importance, have generated a revolution in Taxonomy in general, so it is currently one of the strategies with enormous potential for the identification and genetic study of Meloidogyne spp. due to its high sensitivity and reliability of the result. The modern identification techniques will allow a correct identification of Meloidogyne spp., which contributes to the design of adequate management strategies for this phytopathogenic nematode.
Species Diversity of Ring Nematodes of the Genus Criconemoides (nematoda: Criconematidae) Based on Three New Species from China, Using Integrative Taxonomy

Abstract:Ring nematodes of the genus Criconemoides are a group of polyphagous, migratory root-ectoparasites of plants. In a nematological survey of Zhejiang Province, three new Criconemoides species were isolated from the rhizosphere of Citrus sp., Euonymus alatus and Pterocarya stenoptera trees. In the present study, the new species characterized using an integrative taxonomic approach and described as Criconemoides parainformis n. sp., C. geraerti n. sp. and C. rotundicaudatus n. sp. Three of the species shares truncated anteriorly flattened lip region comprised of two annuli, body annuli margin smooth with few anastomoses, (except C. rotundicaudatus n. sp. it has crenated annuli margins and numerous anastomoses), excretory pore 1–2 annuli posterior to pharyngeal bulb base, (except C. geraerti n. sp. it has excretory pore at the same level, or 1–2 annuli anterior to pharyngeal bulb base), vulva closed, anus hardly visible 1–3 annuli posterior to vulva. The new species can be distinguished based on their en face view pattern under scanning electron microscope (SEM) and the tail shapes. i.e. C. parainformis n. sp. has an ellipsoid oral disc, slightly fused pseudolips and tail ending in a bilobed terminus. C. geraerti n. sp. has a hexagonal oral disc, separated pseudolips, and a bluntly conoid tail. C. rotundicaudatus n. sp. has a rounded oral disc, well developed pseudolips, and a short broadly rounded tail. Besides detailed morphological studies using SEM, genomic and mitochondrial molecular data were obtained for the three described new species. In inferred phylogenies, these three species clustered with known Criconemoides species in 18S, 28S, ITS and coxI trees. Currently, only four known Criconemoides species occur in China and the discovery of the three new Criconemoides species in present study has increased our knowledge on the diversity of Criconemoides in China.

During May 2016, rice plants in irrigated fields in Jinhua, Zhejiang Province, eastern China, showed symptoms of root-knot nematode infestations; above ground symptoms were stunted growth and leaf chlorosis while infected root tips became swollen and hooked. Several pear shaped females of Meloidogyne sp. were observed with gelatinous egg masses often completely embedded inside the root galls. The perineal pattern of the females(n = 9) was oval-shaped with low and round dorsal arches, and striae were smooth and sometimes few lines converge at either end of vulva. Lateral fields obscure or absent. Measurements (mean ±sd, range) of J2 (n = 20): body length (456.7 ± 31. 8 µm, 402.7 to 509.0 µm), stylet (12.1 ± 0.7 µm, 10.6 to 13.1 µm), tail length (70.2 ± 6.1 µm, 61.2 to 79.8 µm) and hyaline tail terminus (19.5 ± 1.8 µm, 16.5 to 22.6 µm); male (n = 17) abundant, body cylindrical and fairly long (1,270 ± 158.8 µm, 1,043.4 to 1,553.4 µm), stylet (17.2 ± 1.0 µm, 15.2 to 18.9 µm), and spicule (21.1 ± 0.8 µm, 20.1 to 21.9 µm). Seven individual females were transferred to seven different tubes for DNA extraction. The D2/D3 region of 28S RNA was amplified with D2A (5′-ACAAGTACCGTGAGGGAAAGTTG-3′) and D3B (5′-TCGGAAGGAACCAGCTACTA-3′) (De Ley et al. 1999) yielded a PCR fragment of 766 bp. The ITS region was amplified with AB28 (5′-ATATGCTTAAGTTCAGCGGGT-3′) and TW81 (5′-GTTTCCGTAGGTGAACCTGC-3′) (Curran et al. 1994) yielded a PCR fragment of 579 bp. BLAST results of D2/D3 region of 28S RNA and ITS sequences exhibited 99 to 100% similarity with those of M. graminicola isolates available in GenBank. The sequences of this study were deposited in GenBank (28S D2/D3 accession no. KY660544–KY660545, ITS accession no. KY660542–KY660543). Both morphological and molecular characterization confirmed the status of this nematode as M. graminicola. To assess reproductive rate of M. graminicola, a 2-week-old rice seedling (Oryza sativa cv. Xiushui 11) was individually transplanted in a standard tube (25 mm × 95 mm) containing 10 ml Pluronic F-127 medium, inoculated with 100 second stage juveniles, and incubated in a growth chamber under 16/8 h light/dark photoperiod at 28/26°C for 15 days (Cappello et al. 2016). There were 21 inoculated replicates and 11 noninoculated as control. The egg masses were examined 15 days after inoculation and the multiplication factor [(number of egg masses × number of eggs per egg mass)/nematode inoculum level] values were 8.94 to 11.58. All inoculated plants were stunted and leaves became chlorotic, galls were observed on roots, and root tips become hook-shaped. No galls were found on the control roots. All the parameters confirmed the pathogenicity of the isolated M. graminicola population on rice. Investigation of Root-knot Nematodes in East China

Abstract:The fauna of the root knot nematodes was investigated from 104 samples in six provinces of East China. The root knot nematodes on 65 species of host plants were identified as Meloidogyne incognita, M.arenaria and M.javanica by the perineal patterns and the esterase isozymes. The occurrence frequencies of them were 88%, 4% and 8%, respectively. M.incognita was the most common species. 26 of the 65 host plants were recorded for the first time in China. The results of investigation can be used as reference to work out a control scheme of the root knot nematodes in this region.

Abstract:AbstractRoot-knot nematodes (RKN) are sedentary parasites of the roots of plants and are considered some of the most damaging pests in agriculture. Since RKN target the root vascular system, they provoke host nutrient deprivation and defective water transport, causing above-ground symptoms of growth stunting, wilting, chlorosis, and reduced crop yields. In Mexico RKN infestations are primarily dealt with by treating with synthetic chemically based nematicides that are preferred by farmers over available bioproducts. However, due to environmental and human health concerns chemical control is increasingly restricted. Biological control of RKNs can help reduce the use of chemical nematicides as it is achieved with antagonistic organisms, mainly bacteria, fungi, other nematodes, or consortia of diverse microorganisms, which control nematodes directly by predation and parasitism at different stages: eggs, juveniles, or adults; or indirectly by the action of toxic diffusible inhibitory metabolites. The need to increase agricultural production and reduce negative environmental impact creates an opportunity for optimizing biological control agents to suppress nematode populations, but this endeavour remains challenging as researchers around the world try to understand diverse control mechanisms, nematode and microbe life cycles, ecology, metabolite production, predatory behaviours, molecular and biochemical interactions, in order to generate attractive products with the approval of local regulatory bodies. Here, we provide a brief review of the biology of the genus Meloidogyne, biological control strategies, and a comparison between chemical and bioproducts in the Mexican market, and guidelines emitted by national agencies to ensure safety and effectiveness of new developments.

Advances in phylogenetic studies of Nematoda

Abstract:Nematoda is a metazoan group with extremely high diversity only next to Insecta. Caenorhabditis elegans is now a favorable experimental model animal in modern developmental biology, genetics and genomics studies. However, the phylogeny of Nematoda and the phylogenetic position of the phylum within animal kingdom have long been in debate. Recent molecular phylogenetic studies gave great challenges to the traditional nematode classification. The new phylogenies not only placed the Nematoda in the Ecdysozoan and divided the phylum into five clades, but also provided new insights into animal molecular identification and phylogenetic biodiversity studies. The present paper reviews major progress and remaining problems in the current molecular phylogenetic studies of Nematoda, and prospects the developmental tendencies of this field.
Identification and potential utility of Metarhizium anisopliae (ITCC9014) for the management of root-knot nematode, Meloidogyne incognita

Abstract:Root-knot nematodes (RKNs) of the genus Meloidogyne are agriculturally important pests, causing serious damage to agricultural crops. Biocontrol strategies have gained unprecedented importance to combat RKNs due to their eco-friendly nature. Here, we have developed a repository of fungi selectively isolated using Caenorhabditis elegans as bait. The isolated fungus was identified morphologically and further confirmed using molecular markers. The fungus was identified as Metarhizium anisopliae. Furthermore, in vitro bioassay of M. incognita showed about 97.67% juvenile parasitization. Additionally, the studied isolate was evaluated for its effect on nematode infection and growth parameters under in vivo conditions. The results revealed that M. anisopliae (ITCC9014) isolate significantly reduced nematode disease burden reflecting parasitic success measured as number of galls, endoparasites, eggmasses, eggs per eggmass which ultimately resulted in 82.3% reduction in the nematode multiplication factor. These results establish that the studied entomopathogenic fungus could be highly promising as nematophagous fungus for RKNs management.
An Improved Polymerase Chain Reaction Method for Identification of Four Major Root Knot Nematodes

Abstract:A rapid and sensitive polymerase chain reaction(PCR)method for the detection and identification of four most common and economically important Meloidogyne spp.was developed.By using previously published primer #C2F3/#1108,fragments of three sizes were detected.The M.incognita and M.javanica reactions produced a 1.7 kb fragment;the M.arenaria reaction,a 1.1 kb fragment;and the M.hapla reaction resulted in a 0.5 kb fragment.Based on the sequence of oesophageal gland protein gene SEC-1,specific primer MI-F/MI-R was designed and tested for its amplification specificity and efficiency against populations of M.incognita.This resulted in two pairs of primers that were used in combination to reliably identify Meloidogyne incognita,M.arenaria,M.javanica and M.hapla.

Abstract:Abstract In Europe, root-knot nematodes are increasingly important. Out of more than 90 Meloidogyne species currently described, 23 have been found on the continent. In the cooler climates, Meloidogyne hapla, M. naasi, M. chitwoodi and M. fallax are prevalent. Meloidogyne arenaria, M. javanica and M. incognita are the most common species in warmer conditions of southern Europe, but also in glasshouses in northern Europe. Morphological identification of root-knot nematodes is difficult and time consuming; therefore, many research groups have been developing molecular techniques for identification of Meloidogyne species. Meloidogyne chitwoodi and M. fallax are quarantine organisms and subject to regulations, and the highly aggressive M. enterolobii has been added to the EPPO alert list. Differences between temperate and tropical Meloidogyne species and their prevalence in Europe imply the need for different management strategies in south and north Europe. Possible crop rotations for the control of root-knot nematodes are limited due to the wide host range of several important species. The banning of methyl bromide and restrictions on other fumigant pesticides in the EU have increased the application of biofumigation significantly in south Europe. The egg-parasitising fungus Paecilomyces lilacinus is commercialised in Germany and applied as dispersible granules for application in water. Intensive research is conducted on the egg-parasitising fungus Pochonia chlamydosporia, and the obligate parasitic bacterium Pasteuria penetrans. European research has paid much attention to resistance breeding and selection. The Mi gene of tomato is widely used but resistance-breaking populations of M. incognita and M. javanica have been reported in different countries.

Abstract:Plant parasitic nematodes (PPNs) cause an important class of diseases that occur in almost all types of crops, seriously affecting yield and quality and causing great economic losses. Accurate and rapid diagnosis of nematodes is the basis for their control. PPNs often have interspecific overlays and large intraspecific variations in morphology, therefore identification is difficult based on morphological characters alone. Instead, molecular approaches have been developed to complement morphology-based approaches and/or avoid these issues with various degrees of achievement. A large number of PPNs species have been successfully detected by biochemical and molecular techniques. Newly developed isothermal amplification technologies and remote sensing methods have been recently introduced to diagnose PPNs directly in the field. These methods have been useful because they are fast, accurate, and cost-effective, but the use of integrative diagnosis, which combines remote sensing and molecular methods, is more appropriate in the field. In this paper, we review the latest research advances and the status of diagnostic approaches and techniques for PPNs, with the goal of improving PPNs identification and detection.

Abstract:Root-knot nematodes are a growing concern for vegetable producers, because chemical nematicides are gradually disappearing. Alternative techniques based on agronomic practices are needed to solve the problem. This review analyzes the most recent studies related to these techniques and their combinations and identifies the most effective ones. Based on an agronomic point of view, the analysis focuses on a description of agricultural factors and practices, rather than on biological processes. Several alternative techniques are considered, including sanitation, soil management, organic amendments, fertilization, biological control and heat-based methods. We analyzed the effects of each practice and interactions among techniques and found large variations among studies. Many practices are only partially effective for nematode control; thus, combining control methods in a systemic analysis presents a challenge. We outline such an ongoing systemic approach and identify key future research studies.

Abstract:Columbia root-knot nematode, Meloidogyne chitwoodi, parasitizes potato plants and causes visible small brown dots in the tuber flesh that dramatically reduce the market value of the crop. In the Pacific Northwest (PNW), two races of M. chitwoodi, Race 1 and Race 2 and a pathotype Race 1Roza exist. The races and pathotype of M. chitwoodi are primarily differentiated based on host tests. Currently, M. chitwoodi can be differentiated from M. hapla or other Meloidogyne Sp. based on morphology and by molecular markers however, the different pathotypes of M. chitwoodi cannot be differentiated using the aforementioned techniques. Based on the in silico genome comparison of M. chitwoodi Race 1, Race 2 and Race 1Roza, we developed diagnostic molecular markers that could successfully differentiate between M. chitwoodi races and pathotype. Further, we also developed three race-specific markers that can specifically amplify Race 1, Race 2, or Race 1Roza. Overall, we have potential markers that can successfully differentiate all the two races and one pathotype of M. chitwoodi through simplex PCR, which has potential application in plant disease diagnostics.
Identification of Meloidogyne Species on Traditional Chinese Medicine Plants in the Qinling Mountain Area of China and Their Aggressiveness to Different Medicinal Herbs

Abstract:Root-knot nematodes (Meloidogyne spp.) are plant-parasitic nematodes that cause serious damage worldwide. There are many species of traditional Chinese medicine (TCM) plants, but only a few have been reported to be infected by Meloidogyne species. From 2020 to 2022, a survey was conducted in the Qinling mountain area, which is the main production region of TCM plants in China. Obvious galling symptoms were observed on the root systems of 15 species of TCM plants. Females were collected from diverse diseased TCM plants and subsequently identified at morphological and molecular levels. Among the 20 diseased root samples collected, Meloidogyne hapla populations were identified in 12 samples (60%), and M. incognita populations were identified in eight samples (40%). Among the 15 species of diseased TCM plants, eight species, namely, Scutellaria baicalensis, Leonurus japonicus, Dioscorea zingiberensis, Cornus officinalis, Viola philippica, Achyranthes bidentata, Senecio scandens, and Plantago depressa, were reported to be infected by Meloidogyne species for the first time. The host status of five species of TCM plants for two M. hapla isolates and one M. incognita isolate from TCM plants in this study was then evaluated. Differences in TCM plants' response to nematode infection were apparent when susceptibility was evaluated by the egg counts per gram of fresh root and the reproduction factor of the nematodes. Among the five species of TCM plants tested, Salvia miltiorrhiza and Gynostemma pentaphyllum were the most susceptible, while S. baicalensis and V. philippica were not considered suitable hosts for M. hapla or M. incognita.

Abstract:Introduction: The root-knot nematodes (RKN), especially Meloidogyne spp., are globally emerging harmful animals for many agricultural crops. Methods: To explore microbial agents for biological control of these nematodes, the microbial communities of the rhizosphere soils and roots of sponge gourd ( Luffa cylindrica ) infected and non-infected by M. incognita nematodes, were investigated using culture-dependent and -independent methods. Results: Thirty-two culturable bacterial and eight fungal species, along with 10,561 bacterial and 2,427 fungal operational taxonomic units (OTUs), were identified. Nine culturable bacterial species, 955 bacterial and 701 fungal OTUs were shared in both four groups. More culturable bacterial and fungal isolates were detected from the uninfected soils and roots than from the infected soils and roots (except no fungi detected from the uninfected roots), and among all samples, nine bacterial species ( Arthrobacter sp., Bacillus sp., Burkholderia ambifaria , Enterobacteriaceae sp., Fictibacillus barbaricus , Microbacterium sp., Micrococcaceae sp., Rhizobiaceae sp., and Serratia sp.) were shared, with Arthrobacter sp. and Bacillus sp. being dominant. Pseudomonas nitroreducens was exclusively present in the infested soils, while Mammaliicoccus sciuri , Microbacterium azadirachtae , and Priestia sp., together with Mucor irregularis , Penicillium sp., P. commune , and Sordariomycetes sp. were found only in the uninfected soils. Cupriavidus metallidurans , Gordonia sp., Streptomyces viridobrunneus , and Terribacillus sp. were only in the uninfected roots while Aspergillus sp. only in infected roots. After M. incognita infestation, 319 bacterial OTUs (such as Chryseobacterium ) and 171 fungal OTUs (such as Spizellomyces ) were increased in rhizosphere soils, while 181 bacterial OTUs (such as Pasteuria ) and 166 fungal OTUs (such as Exophiala ) rose their abundance in plant roots. Meanwhile, much more decreased bacterial or fungal OTUs were identified from rhizosphere soils rather than from plant roots, exhibiting the protective effects of host plant on endophytes. Among the detected bacterial isolates, Streptomyces sp. TR27 was discovered to exhibit nematocidal activity, and B. amyloliquefaciens , Bacillus sp. P35, and M. azadirachtae to show repellent potentials for the second stage M. incognita juveniles, which can be used to develop RKN bio-control agents. Discussion: These findings provided insights into the interactions among root-knot nematodes, host plants, and microorganisms, which will inspire explorations of novel nematicides.
